      • C<sub>2</sub>H <i>N</i> = 1 − 0 and N<sub>2</sub>H<sup>+</sup><i>J</i> = 1 − 0 observations of <i>Planck</i> Galactic cold clumps

        <P>A survey of C2H <I>N</I> = 1 − 0 and N2H<SUP>+</SUP><I>J</I> = 1 − 0 toward <I>Planck</I> Galactic cold clumps (PGCCs) was performed using the Purple Mountain Observatory’s 13.7 m telescope. C2H and N2H<SUP>+</SUP> were chosen to study the chemical evolutionary states of PGCCs. Among 121 observed molecular cores associated with PGCCs, 71 and 58 are detected with C2H <I>N</I> = 1 − 0 and N2H<SUP>+</SUP><I>J</I> = 1 − 0, respectively. The detected lines of most sources can be fitted with a single component with compatible <I>V</I>LSR and line widths, which confirms that these PGCC cores are very cold (with gas temperatures 9-21 K) and quiescent while still dominanted by turbulence. The ratio between the column densities of C2H and N2H<SUP>+</SUP> (<I>N</I>(C2H)/<I>N</I>(N2H<SUP>+</SUP>)) is found to be a good tracer for the evolutionary states of PGCC cores. Gas-grain chemical model can reproduce the decreasing trend of <I>N</I>(C2H)/<I>N</I>(N2H<SUP>+</SUP>) as a function of time. The cores with the lowest abundances of N2H<SUP>+</SUP> (<I>X</I>[N2H<SUP>+</SUP>] < 10<SUP>−10</SUP>) are the youngest, and have nearly constant abundances of C2H. In evolved cores with <I>X</I>[N2H<SUP>+</SUP>] ~10<SUP>−9</SUP>, abundances of C2H drop quickly as the exhaustion of carbon atoms. Although these PGCC cores are in different evolutionary states, they are all quite young (< 5 × 10<SUP>5</SUP> yr) with <I>N</I>(C2H) > <I>N</I>(N2H<SUP>+</SUP>). Mapping observations are carried out toward 20 PGCC cores. The PGCC cores in Cepheus have lower <I>N</I>(C2H)/<I>N</I>(N2H<SUP>+</SUP>) and larger line widths compared with those in Taurus. This implies that PGCC cores in Taurus are less chemically evolved than those in Cepheus.</P>

        Effects of Nitrogen Application Rate on the Yields, Nutritive Value and Silage Fermentation Quality of Whole-crop Wheat

        Whole-crop wheat (Triticum aestivum L.) as forage has been extensively used in the world. In this study, the effects of N application rates on the yields, nutritive value and silage quality were investigated. The N application rates were 0, 75, 150, 225, and 300 kg/ha. The research results indicated that the dry matter yield of whole-crop wheat increased significantly with increasing N rate up to 150 kg/ha, and then leveled off. The crude protein content and in vitro dry matter digestibility of whole-crop wheat increased significantly with increasing N up to 225 kg/ha, while they no longer increased at N 300 kg/ha. On the contrary, the content of various fibers tended to decrease with the increase of N application. The content of lactic acid, acetic acid and propionic acid in silages increased with the increase of N rate (p<0.05). The ammonia-N content of silages with higher N application rates (${\geq}225kg/ha$) was significantly higher than that with lower N application rates (${\leq}150kg/ha$). Whole-crop wheat applied with high levels of N accumulated more nitrate-N. In conclusion, taking account of yields, nutritive value, silage quality and safety, the optimum N application to whole-crop wheat should be about 150 kg/ha at the present experiment conditions.

      • c-Cbl-Mediated Neddylation Antagonizes Ubiquitination and Degradation of the TGF-β Type II Receptor

        Transforming growth factor β (TGF-β) is a potent antiproliferative factor in multiple types of cells. Deregulation of TGF-β signaling is associated with the development of many cancers, including leukemia, though the molecular mechanisms are largely unclear. Here, we show that Casitas B-lineage lymphoma (c-Cbl), a known proto-oncogene encoding an ubiquitin E3 ligase, promotes TGF-β signaling by neddylating and stabilizing the type II receptor (TβRII). Knockout of c-Cbl decreases the TβRII protein level and desensitizes hematopoietic stem or progenitor cells to TGF-β stimulation, while c-Cbl overexpression stabilizes TβRII and sensitizes leukemia cells to TGF-β. c-Cbl conjugates neural precursor cell-expressed, developmentally downregulated 8 (NEDD8), a ubiquitin-like protein, to TβRII at Lys556 and Lys567. Neddylation of TβRII promotes its endocytosis to EEA1-positive early endosomes while preventing its endocytosis to caveolin-positive compartments, therefore inhibiting TβRII ubiquitination and degradation. We have also identified a neddylation-activity-defective c-Cbl mutation from leukemia patients, implying a link between aberrant TβRII neddylation and leukemia development.

        Expression Analysis of the Mx Gene and Its Genome Structure in Chickens

        Among the known interferon-induced antiviral mechanisms, the Mx pathway is one of the most powerful pathways. The Mx protein has direct antiviral activity and inhibits a wide range of viruses by blocking an early stage of the viral replication cycle. Cloning, characterization, and expression of Mx in vivo and in vitro have been conducted. The chicken Mx gene spans 21 kb and is made up of 14 exons and 13 introns, of which the promoter region was analyzed. The real-time PCR results showed that Mx expression was increased in chicken embryo fibroblasts (CEF) after 12- and 24-h induction with polyI: C. Induction of Mx expression by poly I: C in vivo revealed tissue-specific patterns among the chicken tissues tested. A trace expression of Mx was detected in healthy chicken liver tissues from adult chickens without inducement; the expression levels in the liver, heart, and gizzard were higher than in the muscle and kidney. This is the first report to demonstrate the expression of a glutathione-S-transferase-tagged-Mx fusion protein of 75 KDa, as well as the biological activity tested by SDS-PAGE and western blotting.

        Effect of night light regimen on growth performance, antioxidant status and health of broiler chickens from 1 to 21 days of age

        Objective: The study was conducted to evaluate the effects of night light regimen on growth performance, antioxidant status and health of Lingnan Yellow broiler chickens from 1 to 21 days of age. Methods: A completely randomized factorial design involved 2 photoperiods (constant lighting [CL], 24 L:0 D and intermittent lighting [INL], 17 L:3 D:1 L:3 D)×2 light intensities (10 lx and 30 lx). A total of one thousand six hundred and eighty 1-d-old Lingnan Yellow broiler chicks were randomly divided into 4 treatments with 6 replicates (70 birds per replicate). The experiment lasted for 21 d. Results: Photoperiods and light intensities had no effect on average daily gain, feed conversion ratio, and mortality of the broiler chickens (p>0.05). The INL had a significant effect on average daily feed intake (p<0.05) of broiler chickens compared with CL. Photoperiod and light intensity had an interactive effect on melatonin (MT) concentration (p<0.05). At CL, reducing light intensity increased MT concentration; INL birds had higher MT but MT concentration was not affected by light intensity. There was an interactive effect on glutathione peroxidase (GPx) and catalase (CAT) in serum and total antioxidant capability (T-AOC) in liver between photoperiod and light intensity. With the decrease of light intensity, the activities of GPx and CAT in serum and T-AOC in liver increased in CL group (p<0.05). Broiler chickens reared under INL had better antioxidant status and 10 lx treatments had higher activities of CAT in serum than 30 lx (p<0.05). Different photoperiods and light intensities had no effect on malondialdehyde. There was an interaction between photoperiod and light intensity on serum creatine kinase (CK) concentration (p<0.05). At CL, the elevated light intensity resulted in an increase in CK content; INL birds had lower CK concentration especially in low light intensity group. Besides, INL and low light intensity significantly reduced the concentration of serum corticosterone and heat shock protein 70 (p<0.05). Serum immunoglobulin M contents were increased in broiler chickens reared under the INL compared with CL group (p<0.05). Conclusion: Results above suggest that the night light regimen of INL and 10 lx could be beneficial to the broiler chickens from 1 to 21 days of age due to the better health status and electricity savings.

        Deletion of the Lmna Gene Induces Growth Delay and Serum Biochemical Changes in C57BL/6 Mice

        The A-type lamin deficient mouse line ($Lmna^{-/-}$) has become one of the most frequently used models for providing insights into many different aspects of A-type lamin function. To elucidate the function of Lmna in the growth and metabolism of mice, tissue growth and blood biochemistry were monitored in Lmna-deficient mice, heterozygous ($Lmna^{+/-}$) and wide-type ($Lmna^{+/+}$) backcrossed to C57BL/6 background. At 4 weeks after birth, the weight of various organs of the $Lmna^{-/-}$, $Lmna^{+/-}$ and $Lmna^{+/+}$ mice was measured. A panel of biochemical analyses consisting of 15 serological tests was examined. The results showed that Lmna deficient mice had significantly decreased body weight and increased the ratio of organ to body weight in most of tissues. Compared with $Lmna^{+/+}$ and $Lmna^{+/-}$ mice, $Lmna^{-/-}$ mice exhibited lower levels of ALP (alkaline phosphatase), Chol (cholesterol), CR (creatinine), GLU (glucose), HDL (high-density lipoprotein cholesterol) and higher levels of ALT (alanine aminotransferase) (p<0.05). $Lmna^{-/-}$ mice displayed higher AST (aspartate aminotransferase) values and lower LDL (lowdensity lipoprotein cholesterol), CK-MB (creatine kinase-MB) levels than $Lmna^{+/+}$ mice (p<0.05). There were no significant differences among the three groups of mice with respect to BUN (blood urea nitrogen), CK (creatine kinase), Cyc C (cystatin C), TP (total protein), TG (triacylglycerols) and UA (uric acid) levels (p>0.05). These changes of serological parameters may provide an experimental basis for the elucidation of Lmna gene functions.

        Elimination of the cryptic plasmid in Leuconostoc citreum by CRISPR/Cas9 system

        <P>Leuconostoc spp. are important lactic acid bacteria for the fermentation of foods. In particular, L. citreum strains isolated from various foods have been used as host strains for genetic and metabolic engineering studies. In order to develop a food-grade genetic engineering system, L. citreum CB2567 was isolated from Kimchi. However, the isolated bacterium contained a cryptic plasmid which was difficult to eliminate. As the existence of the plasmid might hinder strain engineering, we eliminated the plasmid using an RNA-guided DNA endonuclease CRISPR/Cas9 system. We demonstrated that a plasmid-free L. citreum CB2567 host strain could be efficiently constructed through a two-step procedure: 1) transformation of the 'killer' plasmid expressing Cas9 endonuclease and a guide RNA (gRNA) targeting for a specific sequence in the cryptic plasmid, and 2) serial subculture without antibiotics for curing the killer plasmid. When the crude extract of L. citreum expressing Cas9 and the guide RNA was incubated with a PCR fragment containing the specific sequence recognized by the guide RNA, the PCR fragment was cleaved. Also, the cryptic plasmid pCB42 was successfully eliminated from the host strain after transforming the plasmid harboring Cas9 and the guide RNA. The Cas9 and gRNA expression plasmid used in this study can be applied for genome engineering purposes by additionally introducing an editing DNA template to repair the double strand DNA breakage caused by Cas9 in the genome of L. citreum. This study demonstrates the feasibility of developing CRISPR/Cas9-based genetic engineering tools to develop a safe host strain and construct food-grade lactic acid bacteria without residual antibiotic markers.</P>

        Conjugated Linoleic Acid in Rumen Fluid and Milk Fat, and Methane Emission of Lactating Goats Fed a Soybean Oil-based Diet Supplemented with Sodium Bicarbonate and Monensin

        A metabolic study was conducted with four ruminally-cannulated lactating goats (Saanen, 29 weeks lactation, 65${\pm}$5 kg) in a 4${\times}$4 Latin square design with 4 dietary treatments. The goats were fed a basal mixed diet consisting of 80% concentrate and 20% chopped rye grass hay (DM basis, CON). The goats were also fed the CON diet supplemented with soybean oil at a 5% level of the concentrate (SO), the SO diet supplemented with 0.5% of sodium bicarbonate (SO-B) or the SO-B diet supplemented with 30 ppm monensin (SO-BM). The goats were housed in individual pen and the study was conducted for 8 weeks. An increased molar proportion of propionate (C3) was observed at 1 h (p<0.003) and 6 h (p<0.029) post-feeding from all the supplemented diets. Calculated methane emission was markedly decreased prior to morning feeding (p<0.01), and at 1 h (p<0.05) and 6 h post-feeding (p<0.05) in goats fed the supplemented diets. All the supplements increased (p<0.0001) cis9, trans11-CLA content in rumen fluid. Concentrations of both cis9, trans11-CLA (p<0.0001) and trans10, cis12-CLA (p<0.026) were also increased in the milk fat of lactating goats fed the supplemented diets. The SO-B and SO-BM diets further increased CLA content in goat milk compared to the SO diet. All supplements increased unsaturated (UFA, p<0.002), monounsaturated (MUFA, p<0.002) and polyunsaturated fatty acids (p<0.014) and reduced SFA to UFA ratio (p<0.023). The concentration of MUFA was even greater (p<0.002) for SO-BM than for the SO-B diet. In conclusion, feeding soybean oil (5% of concentrate) to lactating goats was a useful way to improve milk fat and to improve fatty acid profile in the milk by increasing potentially healthy fatty acids such as CLA. Supplementation of sodium bicarbonate or sodium bicarbonate with monensin to the soybean oil-based diet increased CLA content further in goat milk. Supplementation of soybean oil may be an effective method to reduce methane emission in lactating goats.
