        Fabrication of Two-Dimensional PEDOT:PSS Nanosheets Through In Situ Formed Complexes

        This paper presents a simple and e®ective method to fabricate water-soluble two-dimensional (2D) conductive poly(3,4-ethylenedioxythiophene):poly (sodium 4-styrenesulfonate) (PEDOT: PSS) nanosheets. Linear PSS is water-soluble and exhibits a quasi 1D structure in the dilute solution. Addition of 3,4-ethylenedioxythiophene (EDOT) monomers into acidic solutions would form 2D molecular complexes due to charge attraction. In situ polymerization of the ethylenedioxythiophene monomers produces 2D poly EDOT nanosheets. Both transmission electron microscopy and atomic force microscopy characterizations have confirmed the 2D polymeric nanosheets. Further Fourier transform infrared (FTIR) characterization also validated that the 2D nanosheet is composed of EDOT-based units and Raman spectroscopy indicated the strong interactions between ethylenedioxythiophene units in the 2D nanostructures. The electrical conductivity is measured to as high as 551.58 S/m for the thin film of as-produced 2D PEDOT: PSS nanosheets.

        Differences in the biological properties of mesenchymal stromal cells from traumatic temporomandibular joint fibrous and bony ankylosis: a comparative study

        The aim of this study was to compare the functional characteristics of mesenchymal stromal cells (MSCs) from a sheep model of traumatic temporomandibular joint (TMJ) fibrous and bony ankylosis. A sheep model of bilateral TMJ trauma-induced fibrous ankylosis on one side and bony ankylosis on the contralateral side was used. MSCs from fibrous ankylosed callus (FAMSCs) or bony ankylosed callus (BA-MSCs) at weeks 1, 2, 4, and 8 after surgery were isolated and cultured. MSCs derived from the bone marrow of the mandibular condyle (BM-MSCs) were used as controls. The MSCs from the different sources were characterized morphologically, phenotypically, and functionally. Adherence and trilineage differentiation potential were presented in the ovine MSCs. These cell populations highly positively expressed MSC-associated specific markers, namely CD29, CD44, and CD166, but lacked CD31 and CD45 expressions. The BA-MSCs had higher clonogenic and proliferative potentials than the FA-MSCs. The BA-MSCs also showed higher osteogenic and chondrogenic potentials, but lower adipogenic capacity than the FA-MSCs. In addition, the BA-MSCs demonstrated higher chondrogenic, but lower osteogenic capacity than the BM-MSCs. Our study suggests that inhibition of the osteogenic and chondrogenic differentiations of MSCs might be a promising strategy for preventing bony ankylosis in the future.

      • Significant Progress in Construction Equipment of Super High-Rise Building

        The construction of rapid developing super high-rise buildings constantly faces great challenges and the innovation of construction equipment is a focus of these challenges. In this paper, three new inventions including the operation platform, tower crane and hoist are put forward around two of the most important issues of super high-rise building construction: vertical transportation and operation environment. Study background, composition of the equipment, working principles and key technologies are introduced in sequence. In the end, the paper summarizes the main problems in the further development of construction equipment.

      • Low Microsomal Epoxide Hydrolase Expression is Associated with Bladder Carcinogenesis and Recurrence

        Microsomal epoxide hydrolase (mEH) plays a significant role in the metabolism of numerous xenobiotics and is associated with several forms of cancer. Here, we investigated the role of mEH expression in bladder carcinogenesis, subsequent progression and recurrence. The expression of mEH was analyzed by Western blot in 50 bladder urothelial carcinoma and 20 normal epithelial tissues. There was a significantly higher mEH expression in the normal epithelium (P<0.05) and mEH expression was lower in high stage than in low stage tumors (P<0.05). Further, immunohistochmistry in 106 bladder urothelial carcinoma demonstrated mEH expression to be negatively correlated with histological grade, pT stage and recurrence (P<0.05). These findings suggest the important role of mEH in bladder carcinogenesis, cancer development and recurrence, providing support for efforts to develop mEH-based gene therapy.

        Cytosolic escape of mitochondrial DNA triggers cGAS-STING-NLRP3 axis-dependent nucleus pulposus cell pyroptosis

        Low back pain (LBP) is a major musculoskeletal disorder and the socioeconomic problem with a high prevalence that mainly involves intervertebral disc (IVD) degeneration, characterized by progressive nucleus pulposus (NP) cell death and the development of an inflammatory microenvironment in NP tissue. Excessively accumulated cytosolic DNA acts as a damage-associated molecular pattern (DAMP) that is monitored by the cGAS-STING axis to trigger the immune response in many degenerative diseases. NLRP3 inflammasome-dependent pyroptosis is a type of inflammatory programmed death that promotes a chronic inflammatory response and tissue degeneration. However, the relationship between the cGAS-STING axis and NLRP3 inflammasome-induced pyroptosis in the pathogenesis of IVD degeneration remains unclear. Here, we used magnetic resonance imaging (MRI) and histopathology to demonstrate that cGAS, STING, and NLRP3 are associated with the degree of IVD degeneration. Oxidative stress induced cGAS-STING axis activation and NLRP3 inflammasome-mediated pyroptosis in a STING-dependent manner in human NP cells. Interestingly, the canonical morphological and functional characteristics of mitochondrial permeability transition pore (mPTP) opening with the cytosolic escape of mitochondrial DNA (mtDNA) were observed in human NP cells under oxidative stress. Furthermore, the administration of a specific pharmacological inhibitor of mPTP and self-mtDNA cytosolic leakage effectively reduced NLRP3 inflammasome-mediated pyroptotic NP cell death and microenvironmental inflammation in vitro and degenerative progression in a rat disc needle puncture model. Collectively, these data highlight the critical roles of the cGAS-STING-NLRP3 axis and pyroptosis in the progression of IVD degeneration and provide promising therapeutic approaches for discogenic LBP.

      • A New Method of Point-Clouds Accurate Measurement and Reconstruction

        With the reverse engineering development, the accuracy of system is more important in reconstruction, especially in non-contacting measurement. This paper provides a new method measuring the accuracy the point clouds, define a image probability and the point probability according to uncertainty data. The quantity of the uncertain point data is important to measuring the result of reconstruction. The prior data can be catch from the last measurement process, especially the edge data or characteristic points. Referring to prior data, basing on the Bayesian theory the more accuracy posterior data can be computed in this paper. We divided the point cloud into different areas, and organized the data with hierarchical tree-structure. According to the probability of one tree node, we adjust the area corresponding to the node. At last, by using the existing experimental equipment, we verify the measurement of point cloud accuracy algorithm. The depth data was obtained by a laser scanner---SICK LMS100. The depth data can be computed as point data with uncertainty. The result of the reconstruction deeply relies on the quality of prior data.

        Improvement of ventricular remodeling and regulation of PI3K/Akt/mTOR signaling pathway in rats with heart failure by polysaccharides from Stropharia rugosoannulata

        Background Heart failure is a syndrome of impaired cardiac circulation caused by the inability of the heart to provide adequate blood supply. Objective This study aimed to evaluate the improvement of ventricular remodeling and the regulation of PI3K/Akt/mTOR signaling pathway in rats with heart failure by polysaccharides from Stropharia rugosoannulata. Results In high dose group of polysaccharides from Stropharia rugosoannulata, the levels of LVESD, LVEDD, LVMI, CI, PVCA, CVF, and MMP-2 and MMP-9 mRNA expression in rats with heart failure were significantly decreased, while the levels of LVEF, LVFS were significantly elevated (P<0.05). In high dose group of polysaccharides from Stropharia rugosoannulata, the levels of PI3K, p-Akt/Akt, p-mTOR/mTOR, and cleaved-caspase 3 expression and TIMP-1 mRNA expression were significantly elevated, while Bax, p65, Bcl2 protein expression and serum levels of PRA, AngII, ALD, IL-6, TNF-α, ST2 and NT-proBNP were significantly decreased (P<0.05). Conclusions Polysaccharides from Stropharia rugosoannulata can improve ventricular remodeling and cardiac function, reduce inflammatory response and protect cardiac function in rats with heart failure in a dose-dependent manner, and the mechanism of action may be related to the regulation of MMPs/TIMPs balance, inhibition of RAAS system and activation of PI3K/Akt/mTOR signaling pathway.       • An Overview on the Integration of Informatization and Industrialization (IOII)

        The integration of informatization and industrialization (IOII) is a policy guidelines proposed by the Chinese Government to promote the production upgrade and the transformation of economic development mode, which is given increasingly attention by academic circles. In this paper, a brief introduction to IOII is presented. The research collections of IOII required to study the concept, development and process are surveyed as well as the application evaluation. By summarizing the development, relationship and the existing concept of IOII, a new concept of IOII is put forward. Then by studying comparatively the process and the application evaluation of IOII, suggestions for future empirical investigation is offered.
