RNA polymerase II carboxyl-terminal domain (RNAPII CTD) phosphatases are responsible for the dephosphorylation of the C-terminal domain of the small subunit of RNAPII in eukaryotes. Recently, we demonstrated the identification of several interacting partners with human small CTD phosphatase1 (hSCP1) and the substrate specificity to delineate an appearance of the dephosphorylation catalyzed by SCP1. In this study, using the established cells for inducibly expressing hSCP1 proteins, we monitored the modification of β-O-linked N-acetylglucosamine (O-GlcNAc). O-GlcNAcylation is one of the most common post-translational modifications (PTMs). To gain insight into the PTM of hSCP1, we used the Western blot, immunoprecipitation, succinylayed wheat germ agglutininprecipitation, liquid chromatography-mass spectrometry analyses, and site-directed mutagenesis and identified the Ser41 residue of hSCP1 as the O-GlcNAc modification site. These results suggest that hSCP1 may be an O-GlcNAcylated protein in vivo, and its N-terminus may function a possible role in the PTM, providing a scaffold for binding the protein(s).

Ras oncoproteins are small molecular weight GTPases known for their involvement in oncogenesis, which operate in a complex signaling network with multiple effectors. Approximately 25% of human tumors possess mutations in a member of this family. The Raf1/MEK/Erk1/2 pathway is one of the most intensively studied signaling mechanisms. Different levels of regulation account for the inactivation of MAP kinases by MAPK phosphatases in a cell type- and stimuli-dependent manner. In the present study, using three inducible Ras-expressing NIH/3T3 cell lines, we demonstrated that MKP3 upregulation requires the activation of the Erk1/2 pathway, which correlates with the shutdown of this pathway. We also demonstrated, by applying pharmacological inhibitors and effector mutants of Ras, that induction of MKP3 at the protein level is positively regulated by the oncogenic Ras/Raf/MEK/Erk1/2 signaling pathway. NJ+: An Efficient Congestion Control Mechanism for Wireless Networks

Transmission control protocols have to overcome common problems in wireless networks. TCP employing both packet loss discrimination mechanism and available bandwidth estimation algorithm, known as the good existing solution, shows significant performance enhancement in wireless networks. For instance, TCP New Jersey which exhibits high throughput in wireless networks intends to improve TCP performance by using available bandwidth estimation and congestion warning. Even though it achieves 17% and 85% improvements in terms of goodput over TCP Westwood and TCP Reno, respectively, we further improve it by exploring maximized available bandwidth estimation, handling bit-error-rate error recovery, and effective adjustment of sending rate for retransmission timeout. Hence, we propose TCP NJ+, showing that for up to 5% packet loss rate, it outperforms other TCP variants by 19% to 104% in terms of goodput when the network is in bi-directional background traffic.

본 논문은 MPLS(multiprotocol label switching) 망에서 멀티캐스트 서비스를 보호하는 동적인 경로 보호를 지원하기 위한 메커니즘을 제시한다. 인터넷 수요가 급속히 증가함에 따라 IP 트래픽의 효율적 수용을 위한 기술이 필요하게 되어 고도화된 인터넷 서비스를 직접 제공할 수 있는 MPLS 기술 개발이 추진되어 왔다. 또한 서비스의 다양화와 함께 멀티캐스트 서비스가 중요한 응용으로 등장하게 되었다. 본 논문에서는 MPLS 멀티캐스트 망에서 링크 및 노드 장애 상황에서 동적으로 MPLS멀티캐스트 트래픽을 우회하는 방법인 MPLS 멀티캐스트 망에서의 경로보호 기술을 제안한다. 기존의 방법은 MPLS 멀티캐스트 망에서 미리 대체경로를 설정하여 장애 발생시 트래픽을 우회시키는 유니캐스트 경로보호 방법이 있다[7,8]. 그러나 대체경로 설정 시점에 관계없이 MPLS 멀티캐스트 망에서 경로보호를 위해 유니캐스트 경로보호 방법을 이용하는 기존의 방법은 새로운 경로를 설정 하므로 멀티캐스트 데이터 중복을 초래하게 되고 미리 대체경로를 설정하므로 자원의 낭비가 초래된다. 제안하는 경로보호 방법은 MPLS 멀티캐스트 망에서 추가의 새로운 경로없이 동적으로 경로 보호가 가능한 방법이다.

Odorant receptors (ORs), the largest subfamily of G proteincoupledreceptors, detect odorants in the nose. In addition, ORswere recently shown to be expressed in many nonolfactorytissues and cells, indicating that these receptors have physiologicaland pathophysiological roles beyond olfaction. ManyORs are expressed by tumor cells and tissues, suggesting thatthey may be associated with cancer progression or may becancer biomarkers. This review describes OR expression invarious types of cancer and the association of these receptorswith various types of signaling mechanisms. In addition, theclinical relevance and significance of the levels of OR expressionwere evaluated. Namely, levels of OR expression in cancerwere analyzed based on RNA-sequencing data reported in theCancer Genome Atlas; OR expression patterns were visualizedusing t-distributed stochastic neighbor embedding (t-SNE); andthe associations between patient survival and levels of OR expressionwere analyzed. These analyses of the relationships betweenpatient survival and expression patterns obtained froman open mRNA database in cancer patients indicate that ORsmay be cancer biomarkers and therapeutic targets.

<P>Design of high-quality primers for multiple target sequences is essential for qPCR experiments, but is challenging due to the need to consider both homology tests on off-target sequences and the same stringent filtering constraints on the primers. Existing web servers for primer design have major drawbacks, including requiring the use of BLAST-like tools for homology tests, lack of support for ranking of primers, TaqMan probes and simultaneous design of primers against multiple targets. Due to the large-scale computational overhead, the few web servers supporting homology tests use heuristic approaches or perform homology tests within a limited scope. Here, we describe the MRPrimerW, which performs complete homology testing, supports batch design of primers for multi-target qPCR experiments, supports design of TaqMan probes and ranks the resulting primers to return the top-1 best primers to the user. To ensure high accuracy, we adopted the core algorithm of a previously reported MapReduce-based method, MRPrimer, but completely redesigned it to allow users to receive query results quickly in a web interface, without requiring a MapReduce cluster or a long computation. MRPrimerW provides primer design services and a complete set of 341 963 135 <I>in silico</I> validated primers covering 99% of human and mouse genes. Free access: http://MRPrimerW.com.</P>
