        “Worth One Minute”: an anonymous rewarding platform for crowd-sensing systems

        Readily available and affordable consumer-grade electronics,with ever-increasing sensing, computing, and communicationcapabilities, have provided the ground for distributed computationand data collection systems. Crowd-sensing applicationsrely on volunteers providing access to their personal devices—acategory encompassing smartphones, wearables, vehicles, and awide range of ‘Internet of Things’ appliances—and using themas sensors. These systems rely on the willingness of participantsto invest in a common cause, which often entails explicit effortsfrom users, occupation of hardware resources, and risks of sharingprivate data. Incentives and rewarding schemes are adoptedto encourage user participation. This paper introduces the “WorthOne Minute” (WOM) platform: an implementation of a generalpurposerewarding system based on anonymous vouchers. Theplatform is designed to reward user efforts towards the commongood, rewarding their contributions and the intrinsic social valuethey provide, while preserving their anonymity.

        형질 전환된 포플러에 대한 nos - NPT Ⅱ 유전자의 기관별 발현 특성

        To effectively modify tree function with genetic engineering, transgenes must be expressed at the proper level in the appropriate tissues at suitable developmental stages. Toward understanding the spatial and temporal expression of transgenes in woody plants, transgene expression was evaluated in three greenhouse-grown, transgenic lines of Populus alba X P. grandidentata hybrid clone `Hansen`. All transgenic poplar lines possess constructs containing the bacterial nopaline synthase(nos) promoter linked to a neomycin phosphotransferase II(NPT II) selectable marker gene. In addition, each transgenic poplar line contains one of the following gene constructs : 1) a wound-inducible potato proteinase inhibitor II (pin2) promoter linked to a chloramphenicol acetyltransferase(CAT) reporter gene. 2) a nos promoter linked to a PL~12 structural gene : or 3) a Cauliflower Mosaic Virus 35s promoter linked to a PIN2 structural gene. Polymerase chain reaction(PCR) was used to verify the presence of foreign genes in the poplar genome. Enzyme-linked immunosorbent assays(ELISAs) were used to evaluate organ specific expression of the nos-NPT II construct. NPT Ⅱ expression was detected in leaves, petioles, stems, and roots of transgenic poplar, thereby indicating that the nos promoter is potentially effective for general constitutive expression of transgenes. NPT expression varied among transgenic poplar Lines and among organs for one transgenic line, Tr15. With Tr15, NPT Ⅱ levels were highest in older leaves and petioles. These results indicate that screening of several transgenic lines may be required to identify lines with optimal transgene expression.

        <P>Summary</P><P>This article briefly reviews the history of white pine blister rust, attributed to <I>Cronartium ribicola</I>, and addresses current research and management issues in South Korea, Japan and other regions of eastern Asia (China, Russia and Himalaya). For each region, the distribution, damage, aecial hosts, telial hosts and management of <I>C. ribicola</I> and other blister rust fungi on native and introduced white pines are summarized. In addition, blister rust behaviours in eastern Asia and North America are compared; and the potential evolutionary and management implications are discussed.</P>

        Organ Specific Expression of the nos-NPT II Gene in Transgenic Hybrid Poplar

        임목을 대상으로 삽입된 외래 유전자의 공간적, 시기별 발현 특성을 이해하기 위한 기초연구로서 온실에서 생육 중인 형질전환된 2년생 잡종 포플러 (Populus alba X P. grandidentata) Hansen 클론을 대상으로 삽입된 외래 유전자의 발현정도를 각 기관별로 조사하였다. Agrobacterium binary vector pRT45, pRT102 및 pRT104에 의해서 형질전환된 3계통의 형질전환체 Tr15, Tr345, Tr665 모두는 선발가능한 표식 유전자로서 nos promoter-NPT II 유전자가 대상 식물체의 genome에 삽입되어 있으며, 그외에, pin2 promoter-CAT 유전자(pRT45), nos promoter-PIN2 유전자(pRT102), Cauliflower Mosaic Virus 35s promoter-PIN2 유전자(pRT104)가 3계통의 형질전환체에 제각각 삽입되어 있는 잡종 포플러이다. 이들 3계통의 형질전환 포플러 식물체의 DNA를 PCR 검정 기법을 이용하여 분석해 본 결과 선발 가능한 표식 유전자인 NPT-II가 삽입되어 있음이 입증되었다 발현 정도를 비교 분석하기 위해서 NPT-ELISA 검정을 실시하였다. 삽입된 NPT II 유전자는 형질전환된 포플러의 잎, 엽병, 형성층 조직, 줄기의 목질부, 뿌리에서 발현되었으며, 발현 정도는 형질전환된 식물체의 계통에 따라서, 그리고 형진전환된 식물체의 부위에 따라서 다양하게 나타났다. pRT45에 의해서 형질전환된 Tr15 형질전환체의 경우, 늙은 잎과 엽병에서 NPT II 유전자가 가장 높은 수준으로 발현되었으며, 어린 잎과 뿌리 조직에서 가장 낮게 발현되었다. 삽입된 외래 유전자가 각 식물체간에, 각 기관에 따라서 각각 상이한 발현 정도를 나타내는 이와 같은 결과는 형질전환된 식물체에 대한 효과적인 선발과정이 요구됨을 의미함은 물론이고, 형질전환 식물체의 발달 과정에 따라서 삽입된 외래 유전자가 공간적, 시기적으로 각각 다르게 발현할 수 있다는 것을 나타낸다.         Effects of Forest Management Practices and Environment on Occurrence of Armillaria Species

        Influences of environment (indicated by plant associations) and forest management practices on the distribution of Armillaria spp. and genets (vegetative clones) were investigated. A total of 142 isolates of Armillaria was collected from various host trees on pristine and managed sites (thinned and/or fertilized) growing in relatively wet and dry environments in eastern Washington, U.S.A. The incidence of Armillaria spp. was significantly higher in the relatively wetter sites than the relatively drier sites, as indicated by plant associations. However, no differences in Armillaria occurrence were found among different forest management practices (control vs. thinned vs. thinned and fertilized) within both wetter and drier sites. Incidence of Armillaria was significantly different among conifer and shrub species. The highest proportion with Armillaria was found on grand fir (Abies grandis). Based on pairing tests and rDNA sequencing, the 142 isolates were comprised in a total of 20 genets representing three Armillaria species. More diverse Armillaria spp. were found in both relatively wetter and relatively drier sites within the undisturbed control plots, compared to plots disturbed by forest management practices. The results from this study provide baseline information toward understanding how environment and forest management practices influence incidence and diversity of Armillaria species and genets.

        Agrobacterium Binary Vector 에 의한 포플러 형질전환을 위한 기초연구

        Three clones of Populus alba × P. grandidemtata have been tested for susceptibility to Agrobacterium tumefaciens strains A281 and A348. We determined the optimum concentration of kanamycin sulfate for effective selection of leaf disc-derived, transgenic tissues transformed using Agrobacterium binary vector pGA472 containing a neomycin phosphotransferase gene (NPT-II) which confers kanamycin resistance. Of the wild type Ti plasmids contained by the two Agrobacterium strains, pTiBo542 of strain A281 appears Co be best suited to serve as a helper plasmid for binary vector systems. A relatively low concentration (10㎎/ℓ) of kanamycin sulfate inhibited adventitious shoot initiation from leaf discs on regeneration medium. Transformed kanamycin-resistant calli were obtained by culturing Agrobacterium inoculated leaf discs on selective regeneration medium. The transformed kanamycin-resistant calli continued to grow on regeneration media supplemented with kanamycin sulfate to levels of 50 and 204㎎/ℓ. The growth of non-cultivated control calli was severely inhibited on regeneration medium containing 50㎎/ℓ kanamycin sulfate.
