Objectives: Once-weekly teriparatide (W-TPTD) is an effective drug for patients with osteoporosis; however, some patients discontinue W-TPTD owing to its adverse drug reactions (ADRs). Sequential treatment with W-TPTD and antiresorptive therapy may be effective in treating such patients. In this study, we evaluate the efficacy of this sequential treatment regimen. Methods: This retrospective study was conducted at a single institution in Japan. The target subjects were patients with osteoporosis who started W-TPTD treatment. The subjects who received W-TPTD for 6 months or morewere divided into3 groups: TTT (W-TPTD for 18 months); TBT (sequentialtreatmentof W-TPTD/bisphosphonates/W-TPTD; each for 6 months); and TET (sequential treatment of W-TPTD/ elcatonin/W-TPTD,each for 6months) groups. The efficacyendpoints were bonemineral densities (BMD) in the lumbar spine and femur. Results: Lumbar spine BMD in groupTBT increased significantlyby1.6% (P¼0.023), 2.9% (P¼0.001), and 4.4% (P < 0.001) after 6,12, and 18 months, respectively, compared with baseline values. In group TET, it increased by 2.1%, (P ¼ 0.001), 1.3% (P ¼ 0.066), and 3.0% (P ¼ 0.015) after 6, 12, and 18 months, respectively. A significant increase was observed only after 6 and 18 months. In group TTT, it increased significantly by 3.3% (P ¼ 0.023), 5.1% (P ¼ 0.019), and 7.1% (P ¼ 0.010) after 6, 12, and 18 months, respectively. However, no significant difference in total hip BMD was observed among all three groups. No serious ADRs were reported. Conclusion: Inpatients who discontinue treatment with W-TPTD due to ADRs, sequential treatment with W-TPTD and antiresorptive therapy would be beneficial.

Purpose: We estimated the dietary qualities of green soybean (edamame) by using a specialized NIR transmission spectrometer to determine the constitutive properties of the soybean, such as the sucrose content and ninhydrine reaction quantity (NRQ; defined by the ninhydrine reaction, which has a high positive correlation with the total free amino acids), with the purpose of establishing a quality assurance system. Methods: We used a newly developed spectrometer probe that enables in situ estimation of the constituents of the soybean. Results: The calibration results obtained using a wavelength range of 760-960 nm were characterized by $R^2$ = 0.57 and standard error of cross-validation (SECV) of 0.78% for sucrose, and $R^2$ = 0.59 and SECV = 0.35% for NRQ. Conclusions: These results are inferior to those of our previous study obtained using a specialized bench-type transmission spectrometer. The poorer results are attributed to several possible reasons, including the effect of direct sunlight and the unstable sample presentation. We plan to conduct further study using improved optical layout and sample presentation.

A cultivation outline of green soybeans could not set mechanical seeding technics to have difference of mulched soil temperature at any seeding season. Especially, the early cropping type of green soybeans seeding that used mulcting method to get high emergence rate on low temperature season have been high labor load to seed by manual operation. Therefore, the purpose of this study was to develop high efficiency and precision mechanical seeding technique which have ridging and mulching same time. We developed prototype seeding machine combined with up-cut rotary, mulching seeder and the molding implement. After that, we made control device to move diching device for up and down direction. We investigated the seeding accuracy and seeding depth of the prototype seeder, and evaluated the growth and yield, respectively. As a result, it is showed that the seeding accuracy is as same as manual seeding. Although the seeding depth was shallow compared to the conventional manual seeding, it can increase seeding depth than conventional seeding. However, the measured seeding depths were shallower than the set values. This is reason why was influenced of soil compression, posture of the seeding part and covered soil after seeding. The plant length at harvest time might become longer as the seeding depth got deeper. So, we considered that it is possible to do multiple simultaneous work used up-cut ridging and mulching seeder.

In order to shorten breeding time and to decide an optimal harvest time, we have been developing a dedicated NIRT spectrometer since 2004 for the determination of constituent content such as sucrose and free amino acids, which are two major contributors to the eating quality, in a single pod green soybean. Methods: The obtained models showed that the developed NIRT instrument had reasonable accuracy for the determination of these two components. Then we carried out the investigation into the change in two components during a few days storage using these models with changing time, variety/cultivar, packaging and temperature. Results: The result showed that the most affecting factor on decreasing both sucrose content and free amino acids was variety/cultivar. The time, packaging and temperature also affected significantly in most cases.

<P>Nuclear export of microRNAs (miRNAs) by exportin-5 (Exp-5) is an essential step in miRNA biogenesis. Here, we present the 2.9 angstrom structure of the pre-miRNA nuclear export machinery formed by pre-miRNA complexed with Exp-5 and a guanine triphosphate (GTP)-bound form of the small nuclear guanine triphosphatase (GTPase) Ran (RanGTP). The x-ray structure shows that Exp-5:RanGTP recognizes the 2-nucleotide 3′ overhang structure and the double-stranded stem of the pre-miRNA. Exp-5:RanGTP shields the pre-miRNA stem from degradation in a baseball mitt-like structure where it is held by broadly distributed weak interactions, whereas a tunnel-like structure of Exp-5 interacts strongly with the 2-nucleotide 3′ overhang through hydrogen bonds and ionic interactions. RNA recognition by Exp-5:RanGTP does not depend on RNA sequence, implying that Exp-5:RanGTP can recognize a variety of pre-miRNAs.</P>

<P><B>Abstract</B></P> <P>A Z-DNA binding protein (ZBP)-containing protein kinase (PKZ) in fish species has an important role in the innate immune response. Previous structural studies of the Zα domain of the PKZ from <I>Carassius auratus</I> (caZα<SUB>PKZ</SUB>) showed that the protein initially binds to B-DNA and induces B-Z transition of double stranded DNA in a salt concentration-dependent manner. However, the significantly reduced B-Z transition activity of caZα<SUB>PKZ</SUB> at high salt concentration was not fully understood. In this study, we present the binding affinity of the protein for B-DNA and Z-DNA and characterize its extremely low B-Z transition activity at 250 mM NaCl.
