Screening of Kimchi-derived Lactic Acid Bacteria for Prevention of Muscle Loss

Sarcopenia is a chronic disease characterized by the progressive loss of skeletal muscle mass in aged. Recent reports showed probiotics could affect the progression of muscle loss by modulating gut microbiome. Kimchi confers potential probiotics properties containing lactic acid bacteria (LAB). The objective of this study was to evaluate the probiotic properties of LAB isolated from Kimchi and investigate the effect of probiotics on preventing sarcopenia. A total of 255 strains of LAB were isolated from 12 types of Kimchi, and 53 strains were finally selected by resistance to artificial gastric juice, bile salts, and ability of attachment to human intestinal epithelial cells. C.elegans mutant (PD4251) expressing green fluorescent protein myo-3 localized in body wall and vulval muscle was adopted to evaluate the prevention from muscle loss. Four strains (LFR20-010, LFR20-030, LFR20-033, LFR20-040) fed worms showed the higher GFP expression than the standard bacterial source fed. RT-qPCR analysis showed that LFR20-030, LFR20-033 upregulated muscle-related genes in worms. Present study suggests that new Lactobacillus spp. separated from kimchi may delay the progress of sarcopenia.

Bertolotti’s syndrome is the part of lumbosacral transitional vertebra and is a congenital anomaly of the axial spine. The syndrome is associated with changes in the sacrum or iliac crest and secondary arthritis and is often mistaken for ankylosing spondylitis (AS). The mechanism of pain in this disease is unclear, and treatments have not been accurately established. Here, we report two cases of Bertolotti’s syndrome in patients with severe back pain who were initially suspected to have AS and required interventional procedures. The two young adult patients developed symptoms similar to those of inflammatory back pain, and their symptoms did not improve with conventional treatment alone. Both patients underwent interventional management, and the symptoms improved. It may be clinically helpful to confirm the presence of Bertolotti’s syndrome in patients with suspected AS, and interventional therapy may be required in some cases.

Sarcopenia is defined as loss of muscle mass and strength due to aging. Recent studies show that sarcopenia may improve via the gut-muscle axis, suggesting that gut health may affect muscle phenotypes. In this study, we aimed to investigate the ability of Lactobacillus rhamnosus JY02 as a probiotic strain isolated from kimchi to alleviate sarcopenia. L. rhamnosus JY02-conditioned medium (CM) reduced dexamethasone (DEX)-induced myotube diameter atrophy and expression of muscle degradation markers (MuRF1 and atrogin-1) in C2C12 cells. The amelioration of sarcopenia was investigated by measuring body composition (lean mass), hand grip strength, myofibril size (using histological analysis), and mRNA and protein expression of muscle-related factors in a DEX-induced mouse model. The results of these analyses showed that L. rhamnosus JY02 supplementation promoted the production of muscle-enhancement markers (MHC Iβ, MHC IIα, and Myo-D) and reduced both the production of muscle degradation markers and the symptoms of muscle atrophy (loss of lean mass and muscle strength). We also found decreased levels of pro-inflammatory cytokines (IL-6, IFN- γ) and increased levels of anti-inflammatory cytokines (IL-10) in the serum of DEX+JY02-administered mice compared to those in DEX-treated mice. Overall, these results suggest that L. rhamnosus JY02 is a potent probiotic supplement that prevents sarcopenia by suppressing muscle atrophy.

Objective: The purpose of the present study was to evaluate the usefulness of histographic analysis for the perfusion map of pelvic MRI in predicting remnant ovarian tissue in patients with ovarian endometriosis. Methods: To generate the perfusion map, subtracted T1-WI was divided by contrast enhanced T1-WI with using image analysis software ImageJ. Each region of interest (ROI) was quantified by outlining of affected ovaries with endometrioma at the level with the largest area of normal ovary tissue and normal contralateral ovaries using the measurement tool on the software. Consequently, the number of ratios per each pixel composing perfusion map was scored from 0 (not perfused) to 1 (totally perfused). The pixel information, including area within ROI, mean with standard deviation of signal intensity, as well as integrated density of affected ovary with endometrioma, were compared with that of the normal ovary. In addition, we compared histogram according to severity of ovarian invasion. Results: In comparison between affected ovary with endometrioma and normal ovary, perfusion ratio of normal ovary was higher than that of affected ovary (0.20 ± 0.12 versus 0.48 ± 0.07, p < 0.126), whereas area within ROI and perfusion ratio was higher in affected ovary. According to severity of endometrial invasion of ovary based on surgical findings, area with perfusion ratio between 0.4 and 0.8, (528.00 ± 154.43 versus 199.17 ± 163.15, p = 0.003), perfusion ratio (0.27 ± 0.11 versus 0.11 ± 0.07, p= 0.012), and, and integrated density (427.125 ± 132.24 versus 187.33 ± 106.32, p = 0.003) was lower in group of severe invasion than those of mild and moderate invasion group. Conclusion: Histographic analysis for perfusion map of pelvic MRI could be useful in revealing the extent of the endometrial invasion and viable remnant ovarian tissue.

Objective: We aimed to compared the accuracy of fetal fibronectin (fFN) between symptomatic singleton and twin pregnancies in the prediction of preterm birth (PTB). Methods: We included 182 singleton and 52 twin pregnant women with symptomatic preterm labor with intact membranes prospectively. Positive fFN was defined as a fFN level of > 50 ng/mL in cervicovaginal secretion. We evaluated the effectiveness of the fFN test, including sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) to predict the PTB before 34, 36 and 37 weeks of gestation. We classified our study group into 4 groups according to their fFN results and gestational age at delivery: group 1, positive fFN finding and PTB (True); group 2, positive fFN finding and no PTB (False); negative fFN finding, no PTB (True); group 4, negative fFN and PTB (False). Then we compared the effectiveness fFN and the rate of a respective group between a singleton and twin pregnancies. Results: 33.5% of singleton and 38.5% of twin pregnancies showed positive fFN results (p=0.509). Although the rate of PTB before 37 weeks of gestation was higher in twin group (45.1% vs 78.7%, p<0.001), there was no differences in the rate of PTB before 36 (36.3% vs. 50.0%, p=0.074) and 34 weeks of gestation (17.0% vs. 26.9%, p=0.111). There was no significant difference in the effectiveness except NPV for prediction of PTB before 37 weeks of gestation (63.6% vs. 21.9% p < 0.001). Also, there were no significant differences in the rate of true group (group 1 and 3) except in the PTB before 37 weeks of gestation (63.2% vs. 44.2%, p=0.014). Conclusion: Although, the fFN effectiveness is poorer compared to a singleton pregnancy in prediction of PTB before 37 weeks of gestation, its effective is similar with singleton pregnancy in prediction of PTB before 34 and 36 weeks of gestation. fFN could be used in prediction of PTB in twin pregnancies.

Because it is associated with mortality, hyponatremia is an important feature of pulmonary arterial hypertension. Its mechanisms remain unclear, although right heart failure resulting from pulmonary arterial hypertension may lead to systemic congestion and arterial underfilling. However, most patients with pulmonary arterial hypertension are clinically euvolemic and have no peripheral edema. Unlike patients with underlying heart disease, neurohumoral activation is not demonstrated in hyponatremic patients with pulmonary arterial hypertension, who show features of congestive heart failure only at later stages in their disease. Here, a case vignette is introduced, and the pathophysiology of hyponatremia in pulmonary arterial hypertension will be discussed. Syndrome of inappropriate antidiuresis (SIAD) appears to underlie hyponatremia in the initial phase of pulmonary arterial hypertension. The mechanisms by which various lung diseases can lead to SIAD remain an enigma.

<P>The most common enzyme labels in enzyme-linked immunosorbent assays are alkaline phosphatase and horseradish peroxidase, which, however, have some limitations for use in electrochemical immunosensors. This Article reports that the small and thermostable DT-diaphorase (DT-D) and electrochemically inactive 4-nitroso-1-naphthol (4-NO-1-N) can be used as a bifunctional enzyme label and a rapidly reacting substrate, respectively, for electrochemical immunosensors. This enzyme–substrate combination allows high signal amplification via rapid enzymatic amplification and electrochemical redox cycling. DT-D can convert an electrochemically inactive nitroso or nitro compound into an electrochemically active amine compound, which can then be involved in electrochemical–chemical (EC) and electrochemical–enzymatic (EN) redox cycling. Six nitroso and nitro compounds are tested in terms of signal-to-background ratio. Among them, 4-NO-1-N exhibits the highest signal-to-background ratio. The electrochemical immunosensor using DT-D and 4-NO-1-N detects parathyroid hormone (PTH) in phosphate-buffered saline containing bovine serum albumin over a wide range of concentrations with a low detection limit of 2 pg/mL. When the PTH concentration in clinical serum samples is measured using the developed immunosensor, the calculated concentrations are in good agreement with the concentrations obtained using a commercial instrument.
