        Fluorogenic probes for disease-relevant enzymes

        <P>Traditional biochemical methods for enzyme detection are mainly based on antibody-based immunoassays, which lack the ability to monitor the spatiotemporal distribution and, in particular, the <I>in situ</I> activity of enzymes in live cells and <I>in vivo</I>. In this review, we comprehensively summarize recent progress that has been made in the development of small-molecule as well as material-based fluorogenic probes for sensitive detection of the activities of enzymes that are related to a number of human diseases. The principles utilized to design these probes as well as their applications are reviewed. Specific attention is given to fluorogenic probes that have been developed for analysis of the activities of enzymes including oxidases and reductases, those that act on biomacromolecules including DNAs, proteins/peptides/amino acids, carbohydrates and lipids, and those that are responsible for translational modifications. We envision that this review will serve as an ideal reference for practitioners as well as beginners in relevant research fields.</P>

        Favorable Impact Toughness of High Heat Input Coarse-Grained HAZ in an Ultra-Low Carbon High-Strength Microalloyed Steel

        Liquid Mg is capable of quickly eroding the refractory Ni-based superalloy and selectively dissolve Ni, which opens up anew avenue for the recycling of superalloy scraps for comprehensive metal reclamation. This study aims at investigating theunderlining mechanism and kinetics of the selective dissolution of Ni from Inconel 718 (one type of Ni-based superalloy)with liquid Mg, by experimenting at 750, 800, and 850 °C for various amounts of time in an inert atmosphere. This selectivedissolution process is also known as liquid metal dealloying. Results show that a permeable dealloyed layer was formedresulting from the preferential dissolution of Ni over other metallic elements (e.g., Fe, Cr, Nb, Mo, and Ti). At the dealloyingforefront, the grain boundaries of the matrix γ phase were preferentially eroded by liquid Mg. The opposite concentrationgradients of Ni (4–9 wt%) and Mg (12–18 wt%) cross the dealloyed layer suggested their counter diffusion. A higher heatingtemperature substantially increased the dealloying rate.

        Hydrothermal stability of different zeolites in supercritical water: Implication for synthesis of supported catalysts by supercritical water impregnation

        Supercritical water (SCW) impregnation is an efficient and feasible method that has been used to prepare highly dispersed supported catalysts, but few studies have investigated the stability of support materials in supercritical water. Thus, our aim was to investigate the hydrothermal stability of zeolite supports (ZSM-5, TS-1, ZSM-35, HY, 13X, Beta, SAPO-11 and SAPO-34) as model compounds in supercritical water. Results showed that almost all of zeolites suffered from crystallinity change, structural properties degradation, obvious desilication and dealumination. The decrease of surface areas and the collapse of crystalline structures in HY, 13X, Beta, SAPO-11 and SAPO-34 were more serious compared to ZSM-5, ZSM-35 and TS-1. The micropore areas and acidity of all SCW-treated zeolites were reduced. 13X with lower Si/Al ratio had higher hydrothermal stability than HY due to the formation of extra-framework Al (EFAL). EFAl also generated strong Lewis acid sites determined by ammonia temperature-programmed desorption and 27Al magic angle spinning nuclear magnetic resonance. Desilication and dealumination were simultaneous, and led to the increase of framework Si/Al ratio. ZSM zeolites (ZSM-5, ZSM-35 and TS-1) had higher hydrothermal stability than HY, 13X, Beta, SAPO-11 and SAPO-34 in SCW.

        An eight-degree-of-freedom upper extremity exoskeleton rehabilitation robot: design, optimization, and validation

        Upper extremity exoskeleton rehabilitation robots can be used for the training of patients with upper extremity motor dysfunction. In most cases, the design of such robots focuses on the configuration and the human-machine compatibility. For patients, the use of an exoskeleton rehabilitation robot mainly aims to improve their movement ability, which depends on the range of movement of the upper extremity joints. This paper proposes an eight-degreeof-freedom (DOF) upper extremity exoskeleton rehabilitation robot to improve the movement range of the patient’s upper extremity joints. The structural parameters of the shoulder joint are optimized and analyzed by the kinematic equations of the mechanism and the cyclic iteration algorithm such that the movement range of the patient joint can be maximized. The movement space of the robot is then simulated. Finally, the movement range of the rehabilitation robot joints and the movement space of the rehabilitation robot were measured. Experimental results show that the upper extremity exoskeleton rehabilitation robot can meet the patient’s shoulder, elbow, and wrist movement range, and the overlap with the human upper extremity movement space is 97.1 % and 95.7 % in the coronal and sagittal planes, respectively.

        Improved inverse kinematics and dynamics model research of general parallel mechanisms

        Since the classical kinematics model of parallel manipulators cannot accurately reflect the angular velocity and angular acceleration of the limbs, an improved kinematics model is proposed and an inverse dynamic model of the general parallel manipulator is derived based on the improved kinematics model. This paper proves that the shortcoming of the classical kinematics model is that a single model cannot accurately describe the movement of several types of branches in a parallel manipulator. Combined with the principle of angular velocity superposition and vector chain method, the improved kinematic models of the general parallel manipulator’s several typical limbs are derived. Then, an explicit inverse dynamic model of a general parallel robot is established based on the principle of virtual work. Finally, to describe the effectiveness of the improved model, we analyzed a new type of UP+SPR+SPU parallel manipulator. The improved models had higher accuracy than the classical models through the comparison.

        Efficacy and dosimetry analysis of image-guided radioactive 125I seed implantation as salvage treatment for pelvic recurrent cervical cancer after external beam radiotherapy

        Objective: To investigate the efficacy of image-guided radioactive 125I seed (IGRIS) implantation for pelvic recurrent cervical cancer (PRCC) after external beam radiotherapy (EBRT), and analyze the influence of clinical and dosimetric factors on efficacy. Methods: From July 2005 to October 2015, 36 patients with PRCC received IGRIS. We evaluated local progression-free survival (LPFS) and overall survival (OS). Results: The median follow up was 11.5 months. The 1- and 2-year LPFS rate was 34.9% and 20%, respectively. The multivariate analysis indicated recurrence site (central or pelvic wall) (hazard ratio [HR]=0.294; 95% confidence interval [CI]=0.121–0.718), lesion volume (HR=2.898; 95% CI=1.139–7.372), D 90 (HR=0.332; 95% CI=0.130–0.850) were the independent factors affecting LPFS. The 1- and 2-year OS rate was 52.0% and 19.6%, respectively. The multivariate analysis suggested pathological type (HR=9.713; 95% CI=2.136–44.176) and recurrence site (HR=0.358; 95% CI=0.136–0.940) were the independent factors affecting OS. The dosimetric parameters of 33 patients mainly included D 90 (128.5±47.4 Gy), D 100 (50.4±23.7 Gy) and V 100 (86.7%±12.9%). When D 90 ≥105 Gy or D 100 ≥55 Gy or V 100 ≥91%, LPFS was extended significantly, but no significant difference for OS. The 79.2% of 24 patients with local pain were suffering from pain downgraded after radioactive 125I seed implantation. Conclusion: IGRIS implantation could be a safe and effective salvage treatment for PRCC after EBRT, which could markedly release the pain. Recurrence site, tumor volume and dose were the main factors affected efficacy. Compared with central recurrence, it was more suitable for patients with pelvic wall recurrent cervical cancer after EBRT.
