        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.         In vitro and in vivo anti-inflammatory effects of pegmatite

        Pegmatite is a coarse-grained intrusive igneous rock rich in rare elements such as uranium, tungsten, and tantalum with Ca, K, Mg, Fe, Se, Ge, and Ho. We tested in vitro and in vivo assays for the anti-inflammatory activity of pegmatites. We firstly evaluated the suppressive effects of pegmatite on macrophage cell line RAW 264.7 cells stimulated with proinflammatory stimuli lipopolysaccharide (LPS) to determine nitric oxide (NO) production and TNF-$\alpha$ and IL-6 release. The $IC_{50}$ values of pegmatite exceeded $5,000\;{\mu}g/mL$. Treatment of RAW 264.7 cells with pegmatite significantly inhibited LPS-stimulated NO production and proinflammatory cytokines such as TNF-$\alpha$ and IL-6 secretion in a dose-dependent manner (P<0.05). In vivo studies were tested with two animal models of arachidonic acid-induced mouse ear edema and an acetic acid-induced increase in capillary permeability. The pegmatite significantly attenuated ear edema induced by arachidonic acid and reduced the acetic acid-induced increase in capillary permeability in mice (P<0.05) when the pegmatite was administered topically (10 mg per ear) for 24 h. Therefore, pegmatite potentially shows an anti-inflammatory activity in the in vitro and in vivo mice and in the development of newer anti-inflammatory agents as mineral materials.

        Antioxidant and hepatoprotective effects of Korean ginseng extract GS-KG9 in a D-galactosamine-induced liver damage animal model

        BACKGROUND/OBJECTIVES: This study was designed to investigate the improvement effect of white ginseng extract (GS-KG9) on D-galactosamine (Ga1N)-induced oxidative stress and liver injury. SUBJECTS/METHODS: Sixty Sprague-Dawley rats were divided into 6 groups. Rats were orally administrated with GS-KG9 (300, 500, or 700 mg/kg) or silymarin (25 mg/kg) for 2 weeks. The rats of the GS-KG9- and silymarin-treated groups and a control group were then intraperitoneally injected Ga1N at a concentration of 650 mg/kg for 4 days. To investigate the protective effect of GS-KG9 against GalN-induced liver injury, blood liver function indicators, anti-oxidative stress indicators, and histopathological features were analyzed. RESULTS: Serum biochemical analysis indicated that GS-KG9 ameliorated the elevation of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) in GalN-treated rats. The hepatoprotective effects of GS-KG9 involved enhancing components of the hepatic antioxidant defense system, including glutathione, glutathione peroxidase (GPx), superoxide dismutase (SOD), and catalase (CAT). In addition, GS-KG9 treatment inhibited reactive oxygen species (ROS) production induced by GalN treatment in hepatocytes and significantly increased the expression levels of nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) proteins, which are antioxidant proteins. In particular, by histological analyses bases on hematoxylin and eosin, Masson"s trichrome, α-smooth muscle actin, and transforming growth factor-β1 staining, we determined that the administration of 500 mg/kg GS-KG9 inhibited hepatic inflammation and fibrosis due to the excessive accumulation of collagen. CONCLUSIONS: These findings demonstrate that GS-KG9 improves GalN-induced liver inflammation, necrosis, and fibrosis by attenuating oxidative stress. Therefore, GS-KG9 may be considered a useful candidate in the development of a natural preventive agent against liver injury.

        Formation and Evaluation of the Coalition Government in Korea

        경기도의 연합정치 성과에 대한 분석을 실시함에 있어 정책연합, 정부연합이라는 두 가 지 분류기준을 주요변수로 규정하였다. 아울러, 경기도 연합정치를 평가함에 있어 한국과 같은 지방자치단체의 독임제 기관구성 하에 연합정치의 성과를 분석하여 연합정치의 활성 화를 위한 방안으로서 지방자치제도 개선방안을 제시하였다. 경기도 연합정치의 한계로 행정업무 지연의 문제 등 연합정치로 인한 다양한 사업으로 인해 많은 예산이 반영되고 해당 부서의 중요사업에 대한 검토를 신속하게 이루어져야 함 에도 불구하고, 보고 및 결제단계 증가로 시간소요 또한 증가하는 등의 문제점이 나타났다. 향후 경기도 연합정치를 활성화시키고 나아가 이를 실천하기 위한 지방자치제도의 개선 방안으로 첫째, 기관구성의 다양화이다. 둘째, 지방장관제 도입이다. 셋째, 광역시도 의회의 역량을 강화하는 것이다. 넷째, 지방선거제도의 개편이다. On the other hand, comparing the first and second periods in terms of the governmental coalition, the allocation of power and the improvement of the coalition process can be mentioned. First, from the aspect of power allocation, the first stage handed over to the opposition party the vice governor of social integration, which is responsible for the three countries of health, welfare, environment, and women's family. In the second stage(Cho Sung-Ho et al., 2017), In this way, the opposition party is able to share power with the opposition party. At the same time, it introduced the coalition chairperson system, which further strengthened the power of political parties participating in the coalition. In addition, by establishing a unified political arbitration committee to facilitate the conflict situation that occurred under the coalition government, the coalition government made soft landing. In the future, we will suggest ways to improve the local autonomy system to revitalize the Gyeonggido coalition politics and to practice it. First, diversification of organization. Second, the introduction of local ministers. Third, the metropolitan city should strengthen its capacity. Fourth, it is a reorganization of local electoral system.

        Regulatory Roles of Chrysanthemum zawadskii Roots in Nuclear Factor E2-related Factor 2/Antioxidant Response Element Pathway

        Cellular protection against carcinogens could be achieved by the induction of phase 2 detoxifying and antioxidant enzymes such as glutathione S-transferase (GST), NAD(P)H:quinone oxidoreductase 1 (NQO1) and heme oxygenase 1 (HO-1). Nuclear transcription factor E2-related factor 2 (Nrf2) binds to antioxidant response element (ARE) in the promoter region of these genes and the resulting transactivation occurs. In the present study the effect of gujeolcho (Chrysanthemum zawadskii) roots on the Nrf2-ARE pathway were investigated. C. zawadskii root extract was fractionated with a series of organic solvents and their ability to induce Nrf2-ARE pathway was examined. We separated the most potent dichloromethane (DCM) fraction into 12 sub-fractions and found several sub-fractions with strong effects on the Nrf2-ARE pathway. Fraction 4 strongly induced the ARE-reporter gene activity as well as Nrf2 expression. Sitosterol was isolated as a major compound in fraction 4 although its activity was not as potent as its mother fraction. These results indicate that C. zawadskii roots might be used as a potential natural chemopreventive source.

        Effect of weld geometry on fatigue performance of 6061-T6 aluminum GMAW: part 2. Lap joint

        To investigate the effect of weld geometry on the fatigue performance of aluminum lap joint arc welding, GMAW experiments and fatigue tests were conducted. The results show that the combination of a smaller toe angle and full penetration with a proper gap size results in the best fatigue performance. A proper gap size can disperse the stress concentration, and the dispersion increases fatigue performance. Because of the reduction in the welded area, partial penetration has a lower fatigue performance than the full-penetration case. In the lap joint, a large toe angle causes a smaller weld reinforcement owing to geometric reasons, and therefore exhibits a lower fatigue performance.

        Alteration of Tight Junction Protein Expression in Dahl Salt-Sensitive Rat Kidney

        <P>Background/Aims: Altered pressure natriuresis is an important mechanism of hypertension, but it remains elusive at the molecular level. We hypothesized that in the kidney, tight junctions (TJs) may have a role in pressure natriuresis because paracellular NaCl transport affects interstitial hydrostatic pressure. Methods: To assess the association of salt-sensitive hypertension with altered renal TJ protein expression, Dahl salt-sensitive (SS) and salt-resistant (SR) rats were put on an 8% NaCl-containing rodent diet for 4 weeks. Systolic blood pressure (SBP) and urine NaCl excretion were measured weekly, and kidneys were harvested for immunoblotting and quantitative PCR analysis at the end of the animal experiments. Results: SBP was significantly higher in SS rats than in SR rats during the first to fourth weeks of the animal experiments. During the first and second week, urinary NaCl excretion was significantly lower in SS rats as compared with SR rats. However, the difference between the two groups vanished at the third and fourth weeks. In the kidney, claudin-4 protein and mRNA were significantly increased in SS rats as compared with SR rats. On the other hand, occludin protein and mRNA were significantly decreased in SS rats as compared with SR rats. The expression of claudin-2, claudin-7, and claudin-8 did not vary significantly between the two groups. Conclusions: In SS rats, SS hypertension was associated with differential changes in renal TJ protein expression. Both upregulation of claudin-4 and downregulation of occludin might increase paracellular NaCl transport in the kidney, resulting in impaired pressure natriuresis in SS rats.         Effect of weld geometry on fatigue performance of 6061-T6 aluminum GMAW: part 3. T-fillet joint

        The effects of various weld geometries on the fatigue performance of T-fillet joint aluminum arc welding were investigated by conducting gas metal arc weld (GMAW) experiments using 6061-T6 material for various weld geometries. The results of the fatigue tests conducted using a 5-ton grade tester demonstrated that the fatigue performance of T-fillet joint aluminum arc welding is primarily affected by the area of the welded part and weld reinforcement. A large welded area can be obtained by full penetration, and a large weld reinforcement can be obtained by a large throat size and a small toe angle. Contrary to expectations, it was observed that optimal size and higher quantity of porosities can increase fatigue performance. This can be attributed to porosities behaving similarly to crack stop holes, which can prevent stress focusing. The deviation in fatigue performance in the case of minute penetration depth and minute throat size is not remarkable. Therefore, deeper penetration and larger weld reinforcement are crucial factors in obtaining higher fatigue performance in the industry.

        Analysis of Treatment and Delay Times by Disease Site and Delivery Technique at Samsung Medical Center - Proton Therapy Center

        We have treated various disease sites using wobbling and scanning proton therapy techniques since December 2015 at the Samsung Medical Center. In this study, we analyze the treatment time for each disease site in 65 wobbling and 50 scanning patient treatments. Treatment times are longest for liver and lung patients using the respiratory gating technique in the wobbling treatment and for cranio-spinal irradiation in pediatric patients with anesthesia in the scanning treatment. Moreover, we analyze the number of incidents causing treatment delays and the corresponding treatment delay time. The X-ray panel was the main reason for delays in the wobbling treatment; this decreased continually from January to June 2016, related closely to the proficiency of the human operators involved. The main reason for delays in the scanning treatment was interlocks during scanning pattern delivery; this was resolved by proton machine engineers. Through this work, we hope to provide other institutes with useful insight for initial operation of their proton therapy machines.

      • Component characteristics of domestic and foreign soybean cultivars cultivated in Korea

        This study was conducted to identify the general and functional components of 8 domestic and foreign soybean cultivars produced in the Yeoncheon area. 100 seeds weight of soybean was the highest at 29.19 g of Cheoldu59 cultivar and lowest at 14.52 g of Nui 2 hu cultivar. Protein content ranged from 36.81% of Yeonpung cultivar to 39.69% of Cheoldu59 cultivar. The fat content was 19.62% of Shinpaldalkong2 cultivar to 21.55% of Gilyuk86 cultivar. Free sugars of fructose, sucrose, raffinose and stachyose were detected. raffinose content was ranged from 0.97% of Cheoldu59 cultivar to 1.14% of Gilyuk86 cultivar, and stachyose content was ranged from 2.50% of Jijori cultivar to 3.30% of Shinpaldalkong2 cultivar. The content of isoflavone was lowest in daidzein at 13.56 μg/g in Gilyuk506 cultivar, highest at 37.84 μg/g in Yeonpung cultivar and lowest in genistein at 3.67 μg/g in Seonnog cultivar, highest at 8.84 μg/g in Yeonpung cultivar. Summarizing the above results, it is considered that Shinpaldalkong2 and Cheoldu59 with high protein content and isoflavone content are good for ingestion or processing.
