<P>Truncated <I>N</I><SUP>6</SUP>-substituted-4′-oxo-and 4′-thioadenosine derivatives with C2 or C8 substitutionwere studied as dual acting A<SUB>2A</SUB> and A<SUB>3</SUB> adenosinereceptor (AR) ligands. The lithiation-mediated stannyl transfer andpalladium-catalyzed cross-coupling reactions were utilized for functionalizationof the C2 position of 6-chloropurine nucleosides. An unsubstituted6-amino group and a hydrophobic C2 substituent were required for highaffinity at the hA<SUB>2A</SUB>AR, but hydrophobic C8 substitutionabolished binding at the hA<SUB>2A</SUB>AR. However, most of synthesizedcompounds displayed medium to high binding affinity at the hA<SUB>3</SUB>AR, regardless of C2 or C8 substitution, and low efficacyin a functional cAMP assay. Several compounds tended to be full hA<SUB>2A</SUB>AR agonists. C2 substitution probed geometrically throughhA<SUB>2A</SUB>AR docking was important for binding in order of hexynyl> hexenyl > hexanyl. Valve core shapes analysis on flux through control valves in nuclear power plants

Control valves are widely used to regulate fluid flux in nuclear power plants, and there are more than1500 control valves in the primary circuit of one nuclear power plant. With their help, the flux can beregulated to a specific level of water or steam to guarantee the energy efficiency and safety of the nuclearpower plant. The flux characteristics of the control valve mainly depend on the valve core shape. In orderto analyze the effects of valve core shapes on flux characteristics of control valves, this paper focuses onthe valve core shapes. To begin with, numerical models of different valve core shapes are established, andresults are compared with the ideal flux characteristics curve for the purpose of validation. Meanwhile,the flow fields corresponding to different valve core shapes are investigated. Moreover, relationshipsbetween the valve core opening and the outlet flux under different valve core shapes are carried out. <P>The anthracycline antibiotic doxorubicin is commonly used antineoplastic drug in breast cancer treatment. Like most chemotherapy, doxorubicin does not selectively target tumorigenic cells with high proliferation rate and often causes serve side effects. In the present study, we demonstrated the cellular senescence and senescence associated secretory phenotype (SASP) of both breast tumor cell MDA-MB-231 and normal epithelial cell MCF-10A induced by clinical dose of doxorubicin (100 nM). Senescence was confirmed by flattened morphology, increased level of beta galactose, accumulating contents of lysosome and mitochondrial, and elevated expression of p16 and p21 proteins. Similarly, SASP was identified by highly secreted proteins IL-6, IL-8, GRO, GM-CSF, MCP-1, and MMP1 by antibody array assay. Reciprocal experiments, determined by cell proliferation and apoptosis assays and cell migration and cell invasion, indicated that SASP of MDA-MB-231 cell induces growth arrest of MCF-10A, whereas SASP of MCF-10A significantly stimulates the proliferation of MDA-MB-231. Interestingly, SASP from both cells powerfully promotes the cell migration and cell invasion of MDA-MB-231 cells. Treatment with the natural product ginsenoside Rh2 does not prevent cellular senescence or exert senolytic. However, SASP from senescent cells treated with Rh2 greatly attenuated the above-mentioned bystander effect. Altogether, Rh2 is a potential candidate to ameliorate this unwanted chemotherapy-induced senescence bystander effect. </P>

Some chemosensory genes are found to be expressed specifically or highly in the male reproductive system, suggesting their importance in male reproductive physiology. In this paper, we identified chemosensory genes expressed in the male reproductive system of the moth pest Spodoptera exigua by transcriptome and proteome sequencing, and further determined expression patterns of these genes regarding antennae and different tissues of the male reproductive system (testes, vas deferens, seminal vesicles, ejaculatory ducts, male accessory glands) by the real-time quantitative PCR (qPCR). Transcriptome analysis showed that 46 chemosensory genes were expressed in the male reproductive system of S. exigua, including 18 odorant binding proteins (OBPs), 12 che mosensory proteins (CSPs), 10 odorant receptors (ORs) and 6 other chemosensory genes. Further qPCR mea surements revealed that 14 chemosensory genes were expressed specifically or highly in the male reproductive tissues, relative to that in male antennae. Of the 14 genes, OBP17, OBP22, OBP27 and GR43 were specifically expressed in the testes; OBP20 was specifically expressed in the vas deferens; and PBP4 was specifically expressed in the male accessory glands. In addition, proteome data of the spermatophore confirmed that at least 4 OBPs and one CSP were transmitted from males to females, suggesting their function in the reproductive physiology of mated females. The results provide insights into the functional diversity of chemosensory genes, and bases for exploring functions of these chemosensory genes in male reproductive physiology.
Preparation and characterization of merlinoite for potassium extraction from seawater

The preparation of zeolites for potassium extraction was extensively investigated by hydrothermal method without organic template. The K+ ion-exchange capacities of synthetic zeolites in seawater were tested. Merlinoite with high ion-exchange capacity was successfully synthesized in the synthesis gel with the molar composition of 15K2O:12SiO2:Al2O3:570H2O. The K+ ion-exchange capacity of merlinoite is 58.7 mg/g in seawater, and therefore it can be used to extract potassium selectively from seawater. Merlinoite was characterized by X-ray diffraction, scanning electron microscopy and energy dispersion spectroscopy. Merlinoite crystal which is mainly composed of Si, Al, K and O exhibits a square pillar shape.

<P>A/J mice were found to have amino acid differences in Naip5, one of the NOD-like receptors (NLRs) involved in the cytosolic recognition of pathogen-associated molecular patterns and one of the adaptor proteins for caspase-1 activation. This defect was associated with a susceptibility to <I>Legionella</I> infection, suggesting an important role for Naip5 in the immune response also to other intracellular pathogens, such as <I>Mycobacterium leprae</I>. In this study, the immune responses of macrophages from A/J mice against <I>M. leprae</I> were compared to those of macrophages from C57BL/6 mice. Infection with <I>M. leprae</I> induced high levels of TNF-<I>α</I> production and NF-<I>κ</I>B activation in A/J and C57BL/6 macrophages. Caspase-1 activation and IL-1<I>β</I> secretion were also induced in both macrophages. However, macrophages from A/J mice exhibited reduced caspase-1 activation and IL-1<I>β</I> secretion compared to C57BL/6 macrophages. These results suggest that NLR family proteins may have a role in the innate immune response to <I>M. leprae</I>.</P>
Vitamin D Receptor Gene TaqI, BsmI and FokI Polymorphisms in Korean Patients with Tuberculosis

Background: The active metabolite (1, 25- dihydroxycholecalciferol) of vitamin D (25-hydroxycholecalciferol) leads to activation of macrophages and deficiency of vitamin D seems to be involved in the risk of tuberculosis. The effects of vitamin D are exerted by interaction with the vitamin D receptor (VDR) and may be influenced by polymorphism in the VDR gene. In this study, variation in the VDR gene was investigated in Korean population with tuberculosis. Methods: We typed three VDR polymorphisms of restriction endonuclease sites for TaqI, BsmI and FokI in 155 patients with tuberculosis and 105 healthy volunteers. Results: The frequencies of FokI genotypes determined from TB patients were 29.13% for FF, 56.31% for Ff, and 14.56% for ff. We observed 1.4-fold increased prevalence of the Ff genotype in TB patients compared with normal healthy groups (p=0.0857). However, there was no significant association between the genotype groups, TB patient and normal control, for FokI polymorphism. There was also no significant association between VDR gene and tuberculosis in another polymorphism (BsmI and TaqI). Conclusion: Three polymorphisms (TaqI, BsmI and FokI) in the VDR gene do not appear to be responsible for host susceptibility to human tuberculosis in Korean population.

Nuclear factor erythroid 2-related factor 2 (Nrf2) is an important redox-sensitive transcription factor that regulates the expression of several cytoprotective genes. More recently, genetic analyses of human tumors have indicated that Nrf2 may cause resistance to chemotherapy. In this study, we found that the expression levels of Nrf2 and its target genes GCLC, HO-1, NQO1 were significantly higher in cisplatin-resistant A549 (A549/CDDP) cells than those in A549 cells, and this resistance was partially reversed by Nrf2 siRNA. 3,4,5,5,7-Pentamethoxyflavone (PMF), a natural flavon extracted from Rutaceae plants, sensitized A549/CDDP to CDDP and substantially induced apoptosis compared with that of CDDP alone treated group, and this reversal effect decreased when Nrf2 was downregulated by siRNA. Mechanistically, PMF reduced Nrf2 expression leading to a reduction of Nrf2 downstream genes, and in contrast, this effect was decreased by blocking Nrf2 with siRNA. Taken together, these results demonstrated that PMF could be used as an effective adjuvant sensitizer to increase the efficacy of chemotherapeutic drugs by downregulating Nrf2 signaling pathway.
WebChemDB: An Integrated Chemical Database Retrieval System

WebChemDB is an integrated chemical database retrieval system that provides access to over 8 million publicly available chemical structures, including related information on their biological activities and direct links to other public chemical resources, such as PubChem, ChEBI, and DrugBank. The data are publicly available over the web, using two-dimensional (2D) and three-dimensional (3D) structure retrieval systems with various filters and molecular descriptors. The web services API also provides researchers with functionalities to programmatically manipulate, search, and analyze the data.
