        ASGO 7th International Workshop on Gynecologic Oncology

        The 7th International Workshop of the Asian Society of Gynecologic Oncology (ASGO) was held by the Chinese Society of Gynecologic Oncology (CSGO) and Peking Union Medical College Hospital on 27th to 28th August 2022. The 2022 workshop was the first ASGO congress that was held in mainland China. Due to the COVID situation, this workshop was conducted virtually. Five hundred eighty-seven participants from 13 countries/regions (Hong Kong, India, Indonesia, Italy, Japan, Korea, Mainland China, Malaysia, Singapore, Taiwan, Thailand, United Kingdom, and United State of America) attended the meeting (Table 1). Dr. Beihua Kong (Shandong, China), president of CSGO, served as the president. Dr. Yang Xiang, president-elect of Chinese Medical Association (CMA) Gynecologic Oncology division, served as the executive president of the 7th ASGO workshop. Following the 18th ASGO council meeting on August 26th (Fig. 1), the workshop was grandly held on August 27th and 28th virtually.

      • Expression of Fatty Acid Synthase Negatively Correlates with PTEN and Predicts Peritoneal Dissemination of Human Gastric Cancer

        Background: This study aimed to examine the clinical significance of fatty acid synthase (FASN) expression in gastric cancer (GC), and investigate any prognostic role. Materials and Methods: FASN expression was assessed in gastric cancers by immunohistochemistry using 60 paraffin-embedded tissue specimens, and clinical data were collected by retrospective chart review. Moreover, FASN mRNA expression in 15 fresh resected specimens was evaluated by the reverse transcription-polymerase chain reaction (RT-PCR). Immunohistochemical staining of PTEN was performed to assess the correlation of PTEN with FASN in gastric cancer. Results: Increased expression of FASN was noted in gastric cancers. The frequency of FASN gene amplification was also significantly higher in gastric cancer than in adjacent normal tissue. FASN expression in human gastric cancer tissues was significantly correlated with patient TNM stage and peritoneal dissemination (p<0.05). Moreover, higher FASN expression significantly correlated with shorter overall survival (p<0.05). Here, upregulation of FASN negatively correlated with PTEN expression in gastric cancer. Conclusions: These findings indicate that FASN expression is upregulated in gastric cancer, and increased FASN may be critical to th peritoneal metastasis and survival. Our results suggest that FASN upregulation and PTEN downregualtion may be involved in peritoneal dissemination for gastric cancer progression.

      • Involvement of Cdc25c in Cell Cycle Alteration of a Radioresistant Lung Cancer Cell Line Established with Fractionated Ionizing Radiation

        Cancer patients often suffer from local tumor recurrence after radiation therapy. Cell cycling, an intricate sequence of events which guarantees high genomic fidelity, has been suggested to affect DNA damage responses and eventual radioresistant characteristics of cancer cells. Here, we established a radioresistant lung cancer cell line, A549R, by exposing the parental A549 cells to repeated ${\gamma}$-ray irradiation with a total dose of 60 Gy. The radiosensitivity of A549 and A549R was confirmed using colony formation assays. We then focused on examination of the cell cycle distribution between A549 and A549R and found that the proportion of cells in the radioresistant S phase increased, whereas that in the radiosensitive G1 phase decreased. When A549 and A549R cells were exposed to 4 Gy irradiation the total differences in cell cycle redistribution suggested that G2-M cell cycle arrest plays a predominant role in mediating radioresistance. In order to further explore the possible mechanisms behind the cell cycle related radioresistance, we examined the expression of Cdc25 proteins which orchestrate cell cycle transitions. The results showed that expression of Cdc25c increased accompanied by the decrease of Cdc25a and we proposed that the quantity of Cdc25c, rather than activated Cdc25c or Cdc25a, determines the radioresistance of cells.

      • Mini-Array of Multiple Tumor-associated Antigens (TAAs) in the Immunodiagnosis of Esophageal Cancer

        Sera of cancer patients may contain antibodies that react with a unique group of autologous cellular antigens called tumor-associated antigens (TAAs). The present study aimed to determine whether a mini-array of multiple TAAs would enhance antibody detection and be a useful approach in esophageal cancer detection and diagnosis. Our mini-array of multiple TAAs consisted of eleven antigens, p53, pl6, Impl, CyclinB1, C-myc, RalA, p62, Survivin, Koc, CyclinD1 and CyclinE full-length recombinant proteins. Enzyme-linked immunosorbent assays (ELISA) were used to detect autoantibodies against eleven selected TAAs in 174 sera from patients with esophageal cancer, as well as 242 sera from normal individuals. In addition, positive results of ELISA were confirmed by Western blotting. In a parallel screening trial, with the successive addition of antigen to a final total of eleven TAAs, there was a stepwise increase in positive antibody reactions. The eleven TAAs were the best parallel combination, and the sensitivity and specificity in diagnosing esophageal cancer was 75.3% and 81.0%, respectively. The positive and negative predictive values were 74.0% and 82.0%, respectively, indicating that the parallel assay of eleven TAAs raised the diagnostic precision significantly. In addition, the levels of antibodies to seven antigens, comprising p53, Impl, C-myc, RalA, p62, Survivin, and CyclinD1, were significantly different in various stages of esophageal cancer, which showed that autoantibodies may be involved in the pathogenesis and progression of esophageal cancer. All in all, this study further supports our previous hypothesis that a combination of antibodies might acquire higher sensitivity for the diagnosis of certain types of cancer. A customized mini-array of multiple carefully-selected TAAs is able to enhance autoantibody detection in the immunodiagnosis of esophageal cancer and autoantibodies to TAAs might be reference indicators of clinical stage.

        A Study on Foreigner’s Online Repurchasing Behavior in Korea

        한국에 살고 있는 외국인의 수가 최근 급속도로 증가하고 있고, 더욱 더 많은 외국인들이 온라인으로 상품을 구매한다. 따라서 온라인 쇼핑몰들은 증가하고 있는 외국인 온라인 소비자들에게 더 효과적인 서비스를 제공하는 방법을 찾고자 노력하고 있다. 현재 한국의 온라인쇼핑몰의 경쟁은 심화되고 있고, 또한 향후 글로벌 쇼핑몰로 발전할 필요성이 있기 때문에 우선 국내에 거주하고 있는 온라인 외국인 소비자들의 재구매 행위에 대한 연구를 통하여 국내 소비자들의 구매행위와 비교 등을 통하여 향우 지침으로 삼을 필요가 있다. 본 연구는 설문조사와 분석을 통하여 온라인 쇼핑에 있어서 외국인의 재구매 행위에 영향을 미치는 요인을 연구하고, 이를 바탕으로 외국인의 재구매 행위를 촉진시켜 우리나라 온라인 쇼핑몰의 발전과 글로벌화를 위한 시사점을 제시함을 목적으로 한다. The number of foreigners living in Korea has increased rapidly in recent years, and more and more foreigners are purchasing goods online. Therefore, domestic online shopping malls in Korea are trying to find ways of providing better services to foreign online consumers. Online repurchasing behavior of foreigners in Korea who have different cultural backgrounds may be different from Korean online consumers. At present, Korean online shopping malls should be developed globally because they have been facing more rigid competition in domestic market. Thus, this paper needs to study online repurchasing behavior of foreigners living in Korea for the development and globalization of Korean online shopping malls. The purpose of this paper is to study the factors which influence foreigner's online repurchasing behavior through survey on foreigners and it's analysis, and suggest some implications for development and globalization of domestic shopping malls.

        Compound K Suppresses Ultraviolet Radiation-induced Apoptosis by Inducing DNA Repair in Human Keratinocytes

        Ultraviolet (UV)-induced DNA damage is a crucial molecular trigger for sunburn cell formation and skin cancer. Nucleotide excision repair (NER) is the main mechanism in repairing UVB-induced DNA damage of mammalian cells. The purpose of this study is to investigate the functional role of ginsenoside compound K on HaCaT cells (a keratinocyte-derived permanent cell line) irradiated by UV. Hoechst 33258 staining were performed in analyzing UV-induced apoptosis on keratinocytes which were treated with compound K. ImmunoDotBlot assay was used in detecting cyclobutane pyrimidine dimers, the main DNA damage. Western blot analysis was applied for analyzing XPC and ERCC1, two of the NER proteins. Compound K inhibited UV-induced apoptosis of keratinocytes and caused a notable reduction in UV-specific DNA lesions which was due to induction of DNA repair. In agreement with this, compound K induced the expression of particular components of the NER complex, such as XPC and ERCC1. Our results demonstrate that compound K can protect cells from apoptosis induced by UV radiation by inducing DNA repair.

        Characterization and Antioxidant Activity of Released Exopolysaccharide from Potential Probiotic Leuconostoc mesenteroides LM187

        A released exopolysaccharide (rEPS)-producing strain (LM187) with good acid resistance, bile salt resistance, and cholesterol-lowering properties was isolated from Sichuan paocai and identified as Leuconostoc mesenteroides subsp. mesenteroides. The purified rEPS, designated as rEPS414, had a uniform molecular weight of 7.757 × 10<sup>5</sup> Da. Analysis of the monosaccharide composition revealed that the molecule was mainly composed of glucose. The Fourier transform-infrared spectrum showed that rEPS414 contained both α-type and β-type glycosidic bonds. <sup>1</sup>H and <sup>13</sup>C nuclear magnetic resonance spectra analysis showed that the purified rEPS contained arabinose, galactose, and rhamnose, but less uronic acid. Scanning electron microscopy demonstrated that the exopolysaccharide displayed a large number of scattered, fluffy, porous cellular network flake structures. In addition, rEPS414 exhibited strong in vitro antioxidant activity. These results showed that strain LM187 and its rEPS are promising probiotics with broad prospects in industry.

      • Down-regulated MYH11 Expression Correlates with Poor Prognosis in Stage II and III Colorectal Cancer

        The MYH11 gene may be related to cell migration and adhesion, intracellular transport, and signal transduction. However, its relationship with prognosis is still uncertain. The aim of this study was to investigate correlations between MYH11 gene expression and prognosis in 58 patients with stage II and III colorectal cancer. Quantitative real-time polymerase chain reaction was performed in fresh CRC tissues to examine mRNA expression, and immunohistochemistry was performed with paraffin-embedded specimens for protein expression. On univariate analysis, MYH11 expression at both mRNA and protein levels, perineural invasion and lymphovascular invasion were related to disease-free survival (p<0.05; log-rank test). Cancers with lower MYH11 expression were more likely to have a poor prognosis. Otherwise, MYH11 expression was unrelated to patient clinicopathological features. On multivariate analysis, low MYH11 expression proved to be an independent adverse prognosticator (p<0.05). These findings show that MYH11 can contribute to predicting prognosis in stage II and III colorectal cancers.

        A Highly Stretchable, Sensitive Strain Sensor Based on the Dry Printing Method

        Use of Electronic skin (E-skin) has attracted significant attention, as it has broad application prospects in medical care, wearable electronic equipment, and body monitoring — particularly with respect to human motion detection. In this work, we developed a flexible, tensile strain sensor based on the dry printing method. A conductive layer, with a miniaturized network structure, was obtained by: (1) packing the grooves of a grid-like silicon template with a conductive powder composed of carbon nanotubes (CNTs) and silicon dioxide (SiO2) nanoparticles; (2) infiltrating liquid polydimethylsiloxane (PDMS) into the powder voids using a coated, flexible PDMS substrate cover; (3) transferring the solidified, patterned conductive powder onto the flexible substrate by peeling the PDMS substrate cover off the template; (4) fabricating metal electrodes at both ends of the conductive layer and (5) encapsulating the strain sensor with liquid PDMS. After manufacture, the strain sensor was tested using the tensile test. Results from the tensile test demonstrate that the sensor has excellent electrical conductivity, including super high-sensitivity (GF > 1450), a large strain range (up to 35%), and good transparency; as ultraviolet (UV) spectrum analysis shows that the transmittance can reach > 60%. Thus, the sensor is potentially applicable to numerous sub-specialties that require specialized electronics.
