Human lactoferrin (hLTF), an 80-kDa iron-binding glycoprotein, has antitumor activity. In this study, a recombinant adenovirus containing the human lactoferrin cDNA (ad-rhLTF) was constructed and its effect on tumor growth was investigated in mice bearing EMT6 breast cancer. Ad-rhLTF was injected seven times within 14 days into the tumor site at two concentrations (10^8 and 5 × 10^8 pfu/mL) in mice bearing EMT6 breast cancer. Injected ad-rhLTF had considerable cytotoxicity on mice breast cancer, and significantly reducing the weight of tumor produced and increasing the tumor inhibition rate up to 52.64%. The presence of apoptotic cells was confirmed using TUNEL staining and flow cytometry assays. At the same time, RTPCR and Western blot analyses demonstrated that ad-rhLTF also decreased expression of Bcl-2 and increased Bax and caspase 3 expressions. Therefore, we conclude that ad-rhLTF inhibits tumor growth by inducing tumor cell apoptosis in mice with breast cancer by triggering the mitochondrial-dependent pathway and activation of caspase 3. The results indicate that adrhLTF might be a promising drug for breast cancer gene therapy.
The Optimization Process of Aircraft Side Panel’s Weight Reduction Based on Orthogonal Experiment

Reducing aircraft weight for engineering and manufacturing processes is very important but challenging. To solve this problem, a weight-reduction optimization design method for aircraft side panels based on orthogonal experiments and finite-element analysis was proposed for the first time in this paper. The design factors of the aircraft side panels and the layout of composite materials were comprehensively considered in an orthogonal design experiment and optimized using the finite-element analysis software ABAQUS. The side panels were locally strengthened to reduce the thickness of the honeycomb core in the composite material to meet the strength requirements, thereby reducing the weight of the side panels. The final experimental scheme was 245.7 g (11.4%) lighter than the original scheme, showing that the proposed method can efficiently and rapidly achieve significant weight reduction of aircraft side panels.

Laccases can oxidize a variety of phenolic and non-phenolic substrates including synthetic dyes. In this research, a laccase gene Lcc9 from Laccaria bicolor was chemically synthesized and optimized to heterogeneous expression in Pichia pastoris and Arabidopsis thaliana. The properties of recombinant laccase expressed by P. pastoris were investigated. The laccase activity was optimal at 3.6 pH and 40°C. It exhibited K<sub>m</sub> and V<sub>max</sub> values of 0.565 mmol l<sup>-1</sup> and 1.51 μmol l<sup>-1</sup> min<sup>-1</sup> for ABTS respectively. As compared with untransformed control plants, the laccase activity in crude extracts of transgenic lines exhibited a 5.4 to 12.4-fold increase. Both laccases expressed in transgenic P. pastoris or A. thaliana could decolorize crystal violet. These results indicated that L. bicolor laccase gene may be transgenically exploited in fungi or plants for dye decolorization.

<P>In the present investigation, we successfully employed a cell-free extract of Esteya vermicola CNU 120806 to convert ginsenoside Rg3 to Rh2. Three important factors including pH, temperature and substrate concentration were optimized for the preparation of Rh(2). The optimal condition was obtained as follows: 50C, pH 5.0 and substrate concentration of 3 mg ml(-1). The yield of conversion was up to 90.7%. In order to identify the specificity of the β-glucosidase activity of Esteya vermicola CNU 120806, ginsenoside Re (protopanaxatriol saponins) was treated under the same reaction system. Interestingly, no new metabolite was generated, which elucidated that the enzymatic process only occurred by hydrolysis of the terminal glucopyranosyl moieties at the C-3 carbon of ginsenoside Rg(3). The crude enzyme extract can be used for commercial ginsenoside Rh(2) preparation.</P>
Antitumor and Apoptosis Induction Effects of Paeonol on Mice Bearing EMT6 Breast Carcinoma

Paeonol is a major phenolic micromolecular component of Moutan cortex Radicis, a traditional Chinese Medicine. It has shown antitumor effects in previous studies; however, the underlying mechanisms remain unknown. This study investigated the mechanism by giving treatments of placebo, cyclophosphamide, paeonol of 150 and 300 mg/kg to 4 groups of mice bearing EMT6 breast cancer. Apoptosis in tumor cells were confi rmed by morphology analysis, including hematoxylin, eosin staining and TUNEL staining. The results showed that the weight of EMT6 breast tumor was signifi cantly reduced in the groups treated with both 150 and 300 mg/kg of paeonol. Immunohistochemical and Western blot results showed that the expression of Bcl-2 was down-regulated while the expression of Bax, caspase 8 and caspase 3 was up-regulated respectively. These results suggest that paeonol exhibits antitumor effects and the mechanism of the inhibition is via induction of apoptosis, regulation of Bcl-2 and Bax expression, and activation of caspase 8 and caspase 3.
Graphene Oxide Nanoplatelets Grafted Jute Fibers Reinforced PP Composites

To improve mechanical and thermal properties of jute fibers (JFs) reinforced polypropylene (PP) composites, asurface treatment method by grafting graphene oxide nanoplatelets (GONPs) onto JFs was developed. The GONPseffectively enhanced the interfacial adhesion between JFs and PP matrix via improving mechanical interlocking. The resultsshow that PP/JFs-KH570-GONPs composites had the significant improvement of 16.2 % and 12.4 % in tensile and flexuralstrength, respectively. And the thermal property of PP/JFs-Silane-GONPs composites was also improved after treatment. These high performance composites have the potential to be applied in many fields, such as the automotive industry,household products and so on.
