        Preparation of layered double hydroxides intercalated by tartaric acid anion and its application in boron adsorption

        Based on the exchangeability of anions between the layers of magnesium-based LDHs, LDHs intercalatedby tartaric acid anion (TA-LDH) was prepared by co-precipitation method to adsorb boron, and confirmedby XRD and FT-IR that TA had successfully entered the interlayer of LDH. The adsorption kinetics showedthat the adsorption efficiency of B was mainly controlled by the chemisorption mechanism. The thermodynamicsrevealed that the relationship between the equilibrium concentration of B in solution and theadsorption capacity of TA-LDH for B was consistent with the Freundlich model. Cl or Li+ in solution hadlittle effect on the adsorption of B by TA-LDH, while SO4 2 had a great effect on that compared with NO3-LDH. Boron adsorption was accomplished through the interaction between the hydroxyl of TA anion inthe interlayer and that of B(OH)3 and B(OH)4. The elution efficiency of TA-LDH was greater than 70% withammonium chloride used as the eluent. While TA-LDH still maintained a good adsorption capacity after 5cycles, and the solubility loss efficiencies of Mg2+ in a single adsorption and elution were 1.5% and 5.4%,respectively. Our study can provide a new idea and method for the development and utilization of magnesiumresources.

        Deflection of battened beams with shear and discrete effects

        This paper presents a theoretical analysis for determining the transverse deflection of simply supported battened beams subjected to a uniformly distributed transverse quasi-static load. The analysis considers not only the shear effect but also the discrete effect of battens on the transverse deflection of the battened beam. The analytical solution is obtained using the principle of minimum potential energy. Numerical validation of the present analytical solution is accomplished using finite element methods. The present analytical solution shows that the shear effect on the transverse deflection of battened beams increases with the cross-section area of the main member but decreases with the cross-section area of the batten. The longer the battened beam is, or the larger the moment of inertia of the main member is, the smaller the shear effect will be.

      • MiR-454 Prompts Cell Proliferation of Human Colorectal Cancer Cells by Repressing CYLD Expression

        Previous studies have shown that miR-454 plays an important role in a variety of biological processes in various human cancer cells. However, the underlying mechanisms of this microRNA in colorectal cancer (CRC) cells remain largely unknown. In the present study, we investigated the miR-454 role in CRC cell proliferation. We found that miR-454 expression is markedly upregulated in CRC tissues and CRC cells compared with the matched tumor adjacent tissues and the FHC normal colonic cell line. Ectopic expression of miR-454 promoted the proliferation and anchorage-independent growth of CRC cells, whereas inhibition of miR-454 reduced this effect. Bioinformatics analysis further revealed cylindromatosis (CYLD), a putative tumor suppressor as a potential target of miR-454. Data from luciferase reporter assays showed that miR-454 directly binds to the 3'-untranslated region (3'-UTR) of CYLD mRNA and repressed expression at both transcriptional and translational levels. In functional assays, CYLD-silenced in miR-454-in-transfected SW480 cells have positive effect to promote cell proliferation, suggesting that direct CYLD downregulation is required for miR-454-induced CRC cell proliferation. In sum, our data provide compelling evidence that miR-454 functions as an onco-miRNA, playing a crucial role in the promoting cell proliferation in CRC, and its oncogenic effect is mediated chiefly through direct suppression of CYLD expression.

      • MACC1 Expression Correlates with PFKFB2 and Survival in Hepatocellular Carcinoma

        Objective: To validate the relationship between MACC1 and 6-phosphofructo-2-kinase/fructose 2, 6 bisphosphatase (PFKFB2) expression as well as its clinicopathological features and prognostic significance in hepatocellular carcinoma. Methods: By using immunohistochemistry, we investigated the MACC1 and PFKFB2 protein expression in 60 pairs of hepatocellular carcinoma and corresponding non-tumor tissues. Using the Mann-Whitney U test, the Chi-square test, Kaplan-Meier survival analysis, Cox proportional hazard regression analysis and Spearman analysis, we studied the relationship between MACC1 and PFKFB2 protein expression and postoperative overall survival (OS) of the HCC patients. Results: MACC1 and PFKFB2 positive staining rates were significantly higher in hepatocellular carcinoma than in the corresponding nontumor tissues (P=0.012 and 0.04, respectively). The clinicopathological features evaluation revealed that positive expression of MACC1 was associated with a high Edmondson classification (P=0.007) and advanced TNM stage (P=0.027). Similar findings were evident for PFKFB2 expression (P=0.002 and P=0.027). MACC1 and PFKFB2 positive expression was associated with a lower OS rate (P=0.004 and 0.03, respectively). Kaplan-Meier survival and Cox proportional hazard regression analyses revealed MACC1 positive expression to be a prognostic factor for postoperative OS, but PFKFB was not. Conclusion: Highly expressed MACC1 and PFKFB2 protein were associated with TNM stage, Edmondson-Steier classification and overall survival. MACC1 may affect tumor metabolism partly through expression and phophorylation of PFKFB2.

        Biphasic effects of TGFβ1 on BMP9-induced osteogenic differentiation of mesenchymal stem cells

        We have found that the previously uncharacterized bone morphogenetic protein-9 (BMP9) is one of the most osteogenic factors. However, it is unclear if BMP9 cross-talks with TGFβ1 during osteogenic differentiation. Using the recombinant BMP9 adenovirus, we find that low concentration of rhTGFβ1 synergistically induces alkaline phosphatase activity in BMP9-transduced C3H10T1/2 cells and produces more pronounced matrix mineralization. However, higher concentrations of TGFβ1 inhibit BMP9-induced osteogenic activity. Real-time PCR and Western blotting indicate that BMP9 in combination with low dose of TGFβ1 potentiates the expression of later osteogenic markers osteopontin, osteocalcin and collagen type 1 (COL1a2), while higher concentrations of TGFβ1 decrease the expression of osteopontin and osteocalcin but not COL1a2. Cell cycle analysis reveals that TGFβ1 inhibits C3H10T1/2 proliferation in BMP9-induced osteogenesis and restricts the cells in G0/G1 phase. Our findings strongly suggest that TGFβ1 may exert a biphasic effect on BMP9-induced osteogenic differentiation of mesenchymal stem cells. [BMB Reports 2012; 45(9): 509-514]

      • Overview of Methodological Quality of Systematic Reviews about Gastric Cancer Risk and Protective Factors

        Background and Objective: A comprehensive overall review of gastric cancer (GC) risk and protective factors is a high priority, so we conducted the present study. Methods: Systematic searches in common medical electronic databases along with reference tracking were conducted to include all kinds of systematic reviews (SRs) about GC risk and protective factors. Two authors independently selected studies, extracted data, and evaluated the methodological qualities and the quality of evidence using R-AMSTAR and GRADE approaches. Results: Beta-carotene below 20 mg/day, fruit, vegetables, non-fermented soy-foods, whole-grain, and dairy product were GC protective factors, while beta-carotene 20 mg/day or above, pickled vegetables, fermented soy-foods, processed meat 30g/d or above, or salty foods, exposure to alcohol or smoking, occupational exposure to Pb, overweight and obesity, helicobacter pylori infection were GC risk factors. So we suggested screening and treating H. pylori infection, limiting the amount of food containing risk factors (processed meat consumption, beta-carotene, pickled vegetables, fermented soy-foods, salty foods, alcohol), stopping smoking, avoiding excessive weight gain, avoidance of Pb, and increasing the quantity of food containing protective components (fresh fruit and vegetables, non-fermented soy-foods, whole-grain, dairy products). Conclusions: The conclusions and recommendations of our study were limited by including SRs with poor methodological bases and low quality of evidence, so that more research applying checklists about assessing the methodological qualities and reporting are needed for the future.

        Theoretical Study of the Relationships between Excited State Geometry Changes and Emission Energies of Oxyluciferin

        In order to find a relationship between firefly luciferases structure and bioluminescence spectra, we focus on excited substrate geometries which may be affected by rigid luciferases. Density functional theory (DFT) and time dependent DFT (TDDFT) were employed. Changes in only six bond lengths of the excited substrate are important in determining the emission spectra. Analysis of these bonds suggests the mechanism whereby luciferases restrict more or less the excited substrate geometries and to produce multicolor bioluminescence.

      • Validity and Necessity of Sub-classification of N3 in the 7<sup>th</sup> UICC TNM Stage of Gastric Cancer

        Background: The $7^{th}$ TNM staging is the first authoritative standard for evaluation of effectiveness of treatment of gastric cancer worldwide. However, revision of pN classification within TNM needs to be discussed. In particular, the N3 sub-stage is becoming more conspicuous. Methods: Clinical data of 302 pN3M0 stage gastric cancer patients who received radical gastrectomy in Tianjin Medical University Cancer Institute and Hospital from January 2001 to May 2006 were retrospectively analyzed. Results: Location of tumor, depth of invasion, extranodal metastasis, gastric resection, combined organs resection, lymph node metastasis, rate of lymph node metastasis, negative lymph nodes count were important prognostic factors of pN3M0 stage gastric cancers. TNM stage was also associated with prognosis. Patients at T2N3M0 stage had a better prognosis than other sub-classification. T3N3M0 and T4aN3aM0 patients had equal prognosis which followed the T2N3M0. T4aN3bM0 and T4bN3aM0 had lower survival rate than the formers. T4bN3bM0 had worst prognosis. In multivariate analysis, TNM stage group and rate of lymph node metastasis were independent prognostic factors. Conclusions: The sub-stage of N3 may be useful for more accurate prediction of prognosis; it should therefore be applied in the TNM stage system.

        Simulation Analysis of Tire Inflation Pressure Loss under Synergy of Temperature and Oxidation

        The functions of the tire can be fully performed only under the appropriate inflation pressure. Tire inflation pressure loss rate (IPLR) is an important indicator used to evaluate tire inflation pressure retention performance. When the tire is under a rolling state, the temperature will increase leading to an accelerated oxidation process. Standard tire IPLR test is costly and time-consuming. In order to simulate the tire IPLR under the steady rolling state, the permeability parameters of rubbers and rubber-cord components (belts & carcass) are tested. Moreover, the oxidation reaction is coupling with the IPLR model based on the basic autoxidation scheme (BAS). The simulation results show that the IPLR of the tire under rolling state increases obviously. A novel calculation method of rubber oxygen consumption is proposed and applied to the oxygen consumption of different parts of the tire. Two approaches are adopted to reduce tire IPLR based on the innerliner structure design. This simulation method can predict tire IPLR not only under steady rolling state but also under static state and even under tire aging condition in oven. It also provides an important model basis for tire aging and life prediction research in the future.

      • The stiffness-degradation law of base metal after fatigue cracking in steel bridge deck

        The stiffness evaluation of cracked base metal is of great guidance to fatigue crack reinforcement. By carrying out fatigue tests and numerical simulation of typical cracking details in steel box girder, the strain-degradation law of cracked base metal was analyzed and the relationship between base metal stress and its displacement (stiffness) was explored. The feasibility of evaluating the stress of cracked base metal based on the stress field at the crack tip was verified. The results demonstrate that the stiffness of cracked base metal shows the fast-to-slow degradation trend with fatigue cracking and the base metal at 50mm or more behind the crack tip basically lose its bearing capacity. Drilling will further accelerate stiffness degradation with the increase of hole diameters. The base metal stress has a negative linear relation with its displacement (stiffness), The stress of cracked base metal is also related to stress intensity factor and its relative position (distance, included angle) to the crack tip, through which the local stiffness can be effectively evaluated. Since the stiffness is not uniformly distributed along the cracked base metal, the reinforcement patch is suggested to be designed according to the stiffness to avoid excessive reinforcement for the areas incompletely unloaded.
