In this study, a strategy was proposed for making galacto-oligossaccharides (GOS), a high valueadded product, from a byproduct of the dairy industry,cheese whey, using a commercial β-galactosidase from Kluyveromyces lactis (Lactozym® 3000L). The effects of the substrate concentration, temperature, and enzyme dosage were statistically studied and their optimum combinations were determined using response surface methodology. The increase in lactose concentration, temperature, and enzyme concentration favored a transgalactosylation reaction. The maximum values for GOS concentration (119.8 mg/mL)and yield (29.9%) in a 4 h process were obtained in the reaction system, composed of 400 mg/mL of lactose and 10 U/mL of enzyme at 40°C. Under these conditions, the lactose conversion was 68.7%. The maximum value for lactose conversion (87.8%) was observed at the same temperature and enzyme concentration, although the lactose level was 20%.

The yeast Phaffia rhodozyma is known for producing carotenogenic pigments, commonly used in aquaculture feed formulation as well as in cosmetic,pharmaceutical, and food industries. Despite the high production of carotenoids from microorganisms by biotechnology, their use has limitation due to the cell wall resistance, which constitutes a barrier to the bioavailability of carotenoids. Therefore, there is a need to improve carotenoids recovering technique from microorganisms for the application of food industries. This study aimed to compare mechanical, chemical, and enzymatic techniques of cell disruption for extracting carotenoids produced by P. rhodozyma NRRL Y-17268. Among the techniques studied, the highest specific concentration of carotenoids (190.35 μg/g) resulted from the combined techniques of frozen biomass maceration using diatomaceous earth and enzymatic lysis at pH of the reaction medium of 4.5 at 55oC, with initial activity of β-1,3 glucanase of 0.6 U/mL for 30 min.

Background: Wilson`s disease is a genetic disorder caused by a malfunction of ATPase 7B that leads to high accumulation of copper in the organism and consequent toxic effects. We propose a gentle therapy to eliminate the excessive copper content with oral administration of insoluble non-resorbable polymer sorbents containing selective chelating groups for copper ions (Cu²<sup>+</sup> ions). Methods: Polymeric beads with the chelating agents triethylenetetramine, N,N-di(2-pyridylmethyl)amine and 8-hydroxyquinoline were investigated. We performed a long term experiment with Wistar rats fed a diet containing the particular polymers. Further, we investigated non-resorbability of polymeric beads. We measured organ radioactivity after oral administration of 8-hydroxyquinoline on polymer radiolabelled with iodine-125. Non-resorbability was also tested with small animal PET/CT imaging (using copper-64) in NMR mice. Results: In a long-term experiment with Wistar rats fed a diet containing the polymers, we have found that there were no signs of polymer toxicity and the addition of polymers to the diet led to a significant reduction in the copper contents in the kidneys, brains, and livers of the rats. 8-hydroxyquinoline on polymer radiolabelled with iodine-125 was not absorbed from the digestive tract after oral administration. Non-resorbability was also confirmed with PET/CT imaging. Conclusions: We have shown that polymers containing specific ligands could potentially be novel therapeutics for Wilson`s disease. Polymeric beads significantly reduced copper content in selected organs and they are non-resorbable from digestive tract and fully eliminated in feces after oral administration.
Role of Nuclear Factor-κB in female Breast Cancer: A Study in Indian Patients

Introduction: The nuclear factor ${\kappa}B$ (NF-${\kappa}B$) is a super family of transcription factors which plays important roles in development and progression of cancer. The present investigation concerns NF-${\kappa}B$ /p65 activity in human breast cancers with overexpression of ER, PR, HER-2/neu, as well as the significance of p65 expression with regard to menopausal status, stage, grade, tumor size, nodal status, and NPI of invasive ductal carcinomas in Eastern India. Materials and Methods: In this hospital based study 57 breast cancer patients attending a Breast Clinic of a reputed institute of Eastern India were assessed for p65 protein expression in breast tumor tissue samples by Western blotting. ER, PR and HER-2/neu expression was determined by immunohistochemistry. Results: NF-${\kappa}B$/p65 was significantly associated with advanced stage, large tumor size (${\geq}5$ cm), high grade, negative ER, negative PR, and positive HER-2/neu. High NF-${\kappa}B$/p65 expression was more frequent in patients with a high NPI ($NPI{\geq}5.4$, 84.6%) compared with low NPI (<5.4, 44.4%) and this association was statistically significant (p = 0.002). Conclusion: NF-${\kappa}B$/p65 overexpression was associated with advanced stage, large tumor size, high grade, and high NPI which are poor prognostic factors linked to enhanced aggressiveness of the disease. NF-${\kappa}B$/p65 expression implies aggressive biological behavior of breast cancer and this study validates significant association of NF-${\kappa}B$ /p65 overexpression with negative estrogen and progesterone receptor status and overexpression of HER-2/neu oncoprotein. In our good clinical practice, patients with NF-${\kappa}B$ positive tumors need to be treated aggressively.
Numerical Optimization of the Method of Cooling of a Massive Casting of Ductile Cast-Iron

The numerical models of the temperature field of solidifying castings, according to various authors, have been observing two main goals ? directed solidification as the basic assumption for the healthiness of a casting and the optimization of the technology while maintaining the optimal product properties. The achievement of these goals is conditioned by the ability to analyze and, successively, to control the effect of the deciding factors, which either characterize the process or accompany it. An original application of ANSYS simulated the forming of the temperature field of a massive casting from ductile cast-iron during the application various methods of its cooling using steel chills. The numerical model managed to optimize more than one method of cooling but, in addition to that, provided serious results for the successive model of structural and chemical heterogeneity, and so it also contributes to influencing the pouring structure. The file containing the acquired results from both models, as well as from their organic unification, brings new and, simultaneously, remarkable findings of causal relationships between the structural and chemical heterogeneity (i.e. between the sizes of the spheroids of graphite, the cells, density of the spheroids of graphite, etc.) and the local solidification time in any point of the casting. The determined relations therefore enable the prediction of the face density of the spheroids of graphite in dependence on the local solidification time. The calculated temperature field of a two-ton 500x500x1000 mm casting of ductile cast-iron with various methods of cooling has successfully been compared with temperatures obtained experimentally. The casting was cast in sand mould. The calculated model of the kinetics of the temperature field of the casting was verified during casting with temperature measurements in selected points.This has created a tool for the optimization of the structure with an even distribution of the spheroids of graphite in such a way so as to minimize the occurrence of degenerated shapes of graphite, which happens to be one of the conditions for achieving good mechanical properties of castings of ductile cast-iron.

Recently, Parmar [5] introduced a new extension of the ${\tau}$-Gauss hypergeometric function $_2R^{\tau}_1(z)$. The main object of this paper is to study this extended ${\tau}$-Gauss hypergeometric function and obtain its properties including connection with modified Bessel function of third kind and extended generalized hypergeometric function, several contiguous relations, differential relations, integral transforms and elementary integrals. Various special cases of our results are also discussed.

Modern electrochemical energy devices involve generation and reduction of fuel gases through electrochemical reactions of water splitting, alcohol oxidation, oxygen reduction, etc. Initially, these processes were executed in the presence of noble metal-based catalyst that showed low overpotential and high current density. However, its high cost, unavailability, corrosion and related toxicity limited its application. The search for alternative with high stability, durability, and efficiency led scientists towards carbon nanoparticles supported catalysts which has high surface area, good electrical conductivity, tunable morphology, low cost, ease of synthesis and stability. Carbon nanoparticles are classified into two groups based on morphology, one and zero dimensional particles. Carbon nanoparticles at zero dimension, denoted as carbon dots, are less used carbon support compared to other forms. However, recently carbon dots with improved electronic properties have become popular as catalyst as well as catalyst support. This review focused on the recent advances in electrocatalytic activities of carbon dots. The mechanisms of common electrocatalytic reactions and the role of the catalysts are also discussed. The review also proposed future developments and other research directions to overcome current limitations.
