        TRAIL and Effect of Irradiation on Apoptosis of Cancer Cells

        종양을 효율적으로 적출, 제거하기 위해서는 부수적으로 방사선 치료 및 항암화학요법을 이용하여 수술전이나, 수술 후 종양의 크기를 줄이거나 작게 할 수는 있었으나, 종양을 적출하거나 제거하는 방법으로 외과적 절제수술요법이 가장 재래적인 수술 방법이다. 종양의 크기를 줄이거나 작게 하는 방법과, 종양을 사멸시킬 수 있는 항암화학요법은 방사성 감수성을 증가시키는 항암약제를 종양세포에 선택적으로 투여하여 방사선에 대한 감수성에 민감하게 반응 할 수 있도록 한 것이며, 다양한 생물학적인 세포증식억제 방법중 TRAIL은 단백질을 변형 시킬 수 있으며 단백질 구조를 변형시켜 세포의 사멸에 일조를 하는 것으로 알려져 있다. 본 논문에서는 HCT-116세포를 암세포로 간주하여 TRAIL과 방사선과의 상호관계를 분석하였다. 실험결과 TRAIL과 방사선의 단독사용은 대조군과 비교해 본 결과 각각 세포증식과 세포자멸사에 유의적인 영향을 끼치지 않은 것으로 나타났다. 반대로 TRAIL로 처리하고, 방사선 조사를 병행해서 처리한 경우 HCT-116세포가 유의적으로 세포자멸사가 발생되었음을 알 수 있었고, G1대비 G0의 비율도 증가한 것으로 나타났다. 결론적으로 TRAIL은 방사선 방어적인 세포의 세포자멸사를 증가시켜 방사선 감수성을 증가시켰음을 알 수 있었으며, 또한 세포주기를 변화시켜 세포 증식 능력을 점진적으로 감소시킬 수 있었다. TRAIL은 세포자멸사를 증가시키고 세포증식 능력을 감소시켜 방사선 증감제로서 사용이 가능하다는 것으로 사료된다. In this paper, the HCT-116 cells thought to be a cancer cell to analyze the interaction of TRAIL and radiation. Experimental results, single use of TRAIL and radiation, results were compared with the control group, it was found to have no significant effect on each cell proliferation and apoptosis. Conversely treated with TRAIL, when treated in parallel radiation, it was possible to know that the HCT-116 cells significantly apoptosis occurs, The proportion of G1 ratio G0 also was found to have increased. TRAIL conclusion is increased apoptosis radiation defensive cells can know that increased radiosensitivity, also possible to alter the cell cycle, reduce cell proliferation ability stepwise it was possible. TRAIL is increased apoptosis, decreased cell proliferative capacity, it is considered to be possible to use as a radiation sensitizer.

      • Tetraspanin CD82 represses Sp1-mediated Snail expression and the resultant E-cadherin expression interrupts nuclear signaling of β-catenin by increasing its membrane localization

        <P><B>Abstract</B></P> <P>Tetraspanin membrane proteins form physical complexes with signaling molecules and have been suggested to influence the signaling events of associated molecules. Of the tetraspanin proteins, CD82 has been shown to promote homotypic cell-cell adhesion, which partially accounts for its role in suppressing cancer invasion and metastasis. We found here that CD82-induced cell-cell adhesion is attributed to increased E-cadherin expression through CD82-mediated downregulation of the E-cadherin repressor Snail. The Snail repression by CD82 resulted from the reduced binding of the Sp1 transcription factor to the Snail gene promoter. Notably, high CD82 expression did not allow the fibronectin matrix to induce Sp1 phosphorylation, implicating CD82 inhibition of the fibronectin-integrin signaling-dependent Sp1 activation. Meanwhile, E-cadherin upregulated by CD82 pulled β-catenin up to the membrane region, and consequently reduced the amount of cytoplasmic β-catenin that was able to move into to the nucleus. The Wnt signal-induced nuclear translocation of β-catenin was also inhibited by the CD82 function of upregulating E-cadherin. Overall, high CD82 expression was likely to suppress fibronectin adhesion-induced Sp1 activation signaling for Snail expression, resulting in continuous E-cadherin expression, which contributed not only to the maintenance of strong cell-cell adhesion but also to the blockage of nuclear β-catenin signaling.</P> <P><B>Highlights</B></P> <P> <UL> <LI> CD82 upregulates E-cadherin expression by repressing Sp1-mediated Snail expression. </LI> <LI> CD82 suppresses the fibronectin-integrin signaling that induces Sp1 phosphorylation. </LI> <LI> CD82-induced E-cadherin expression increases homotypic adhesion of epithelial cells. </LI> <LI> E-cadherin upregulated by CD82 obstructs the nuclear translocation of β-catenin. </LI> </UL> </P>

        Pyrexia is a new thermal transient receptor potential channel endowing tolerance to high temperatures in Drosophila melanogaster

        Several transient receptor potential channels were recently found to be activated by temperature stimuli in vitro. Their physiological and behavioral roles are largely unknown. From a temperature-preference behavior screen of 27,000 Drosophila melanogaster P-insertion mutants, we isolated a gene, named pyrexia (pyx), encoding a new transient receptor potential channel. Pyx was opened by temperatures above 40 °C in Xenopus laevis oocytes and HEK293T cells. It was ubiquitously expressed along the dendrites of a subset of peripheral nervous system neurons and was more permeable to K<SUP>+</SUP> than to Na<SUP>+</SUP>. Although some pyx alleles resulted in abnormal temperature preferences, pyx null flies did not have significantly different temperature preferences than wild-type flies. But 60% of pyx null flies were paralyzed within 3 min of exposure to 40 °C, whereas only 9% of wild-type flies were paralyzed by the same stimulus. From these findings, we propose that the primary in vivo role of Pyx is to protect flies from high-temperature stress.

      • A Method for Data-mining of Defects on Building Façade Elements

        It has been recognized that the value of buildings gradually decreases over time due to aesthetic, functional, and structural deteriorations caused by various defects. In response to these deteriorations, the value of buildings can be restored by various types of maintenance operations. However, minimizing depreciation is better than maintenance. In this regard, the defects that cause depreciation should be reduced through data analysis on existing buildings. This study suggests the “Pixelization Method (PM)” for data mining on building façade elements to collect data on façade defects. PM is a technique that supports the creation of a database by simplifying the analysis of building façades as well as the input data. The database, developed with PM, is expected to be used in various areas such as analysis of the relationship between the defects and the facade elements or the prediction of defects.

      • 공공 글자체 개발 방안 연구 -대구광역시를 중심으로-

        This paper was described in various directions for the development of public typefaces for the purpose of finding ways to activate public typefaces that represent Daegu. First, the pharmacist of the typeface was divided and examined by period. After the creation of 『Hunminjeongeum』, it has been changed into a geometrical form since it was created, followed by a change from vertical writing to horizontal writing, and various typeface after digital dissemination. In the current development of typeface, the trend of patent applications for font design and the current development of local fonts were discussed. The trend of font design patent applications started in 2005 with a high average of 56.8 until 2019. In 2011, it was the highest with 97 cases. The current status of local typeface development is high, with small and medium-sized enterprises aiming to increase the image by using fonts commercially. In the case of local governments, Jeju's special autonomy is high, and in other municipalities, it is low. In the selection of the representative letters of Hangul typeface, a total of 118 characters were mainly selected, and a method of showing them first was presented. The development of the typographical fonts in Daegu suggested 1) how to create a typeface using the aspects related to Daegu by proposing the use of literature in Daegu, 2) modern print alley type, 3) the use of literature related to Daegu. By developing a public typeface that represents Daegu, it promotes the pride of local people and proposes a method that Daegu residents can use practically. As a result of researching the possibility of developing a typeface that can be representative of an area, commissioned by Daegu Metropolitan City, the development of a representative public typeface that has been given a special story by a local government or a public institution instills pride in the local culture and further promotes local culture.

        지역방언 화자의 중앙방언 습득 양상 연구 : 서울 거주 경상방언 화자의 음운 변이를 중심으로

        This study investigates phonological variations of the Gyeongsang dialect speakers in dialect contact situations in Seoul. This study also examines what factors have significant effects on these phonological variations. It focuses on social factors: ‘gender’, ‘language attitude’, ‘length of residency in Seoul’ and in particular, ‘age group’ that is classified based on whether the participants received compulsory education where standard Korean is used. The tendency of the phonological variation is disproportionate for each phonological variable. In direct dialect contact situations in Seoul, Gyeongsang dialect speakers undergo phonological variation almost all the phoneme and phonological rule but the relatively low toneme. But this quantitative loffwness of the variation degree on toneme variant is not absent in the knowledge of the phonological pattern of Seoul Korean, but rather is hard to adapt or habituate it. In other words, the achievement of toneme variation requires a certain amount of acquisition period compared to the other variants. This study shows that it may take about 20 years to adapt to the phonological patterns of Seoul Korean. The statistical analysis of phonetic data suggests that all social factors investigated in this study influence a speaker s frequency of use of the Seoul Korean variants. Especially, each age group shows different tendency of influence of social factors. Old speaker group influences by social factors in the following order: ‘gender’, ‘length of residency’ and ‘language attitude’. In the case of young, on the other hand, the order is ‘language attitude’, ‘length of residency’ and ‘gender’.

        남악신도시 사례를 통한 도청 이전(移轉) 신도시 개발에 관한 비판적 연구

        본 연구는 전남도청 이전을 목적으로 개발된 남악신도시를 통해 도청 이전 신도시가 내포한 문제점을 비판적으로 바라보고 현재 개발 중인 도청 이전 신도시에 시사점을 남기는 것을 목적으로 한다. 전라남도 남악신도시 개발은 관외 지역에 있는 전남도청을 관내 지역으로 옮기고, 도내 낙후지역을 개발하려는 목적에서 시행되었다. 그러나 신도시 개발사업이 15년 이상 흐른 지금, 1단계 사업이 거의 마무리되는 현시점에서 남악신도시 개발을 살펴보면 다음과 같은 문제점이 발생하였다. 첫째, 대도시 권역의 인구 분산적 차원에서 남악신도시는 해당 목적을 충분히 달성하지 못하였고 오히려 지표상 광주광역시로의 인구집중이 심화되었다. 둘째, 목포시-무안군에 걸친 남악신도시 행정구역의 이원화로 주민들의 행정 서비스 이용에 문제가 발생하였다. 셋째, 신도시 개발에 따른 목포시 원도심 쇠퇴문제가 발생하여 대도시 인구를 흡수하는 것이 아닌, 신도시 주변 지역의 인구를 흡수하여 도시쇠퇴를 가속시킨 문제점이 발생하였다. 이러한 문제점 인식은 향후 도청 이전 신도시 개발에 적절한 해결방안을 찾는 데 도움이 될 것이다. This study aims to critically look at the problems of the newtown development for the relocation of provincial government office through the case of Namak Newtown, constructed for the purpose of relocating the Jeollanam-do Provincial Office. According to the study, the development of the Namak Newtown has three major problems. First, the goal of population distribution from the metropolitan city has not been achieved. Second, the dualization of administrative districts caused problems in providing administrative services to the newtown citizens. Third, it has caused problems in the decline and hollowing out of a city nearby the newtown. Recognizing these problems will help to find out an appropriate solution about the newtown development for the relocation of provincial government office in later.

      • 레이저를 활용한 Hard IP용 PAB DOOR 융착에 대한 영향인자 연구

        본 논문에서는 Hard IP용 PAB Door 레이저 융착에 대한 인자별 영향도를 확인하고, 인자별 최적화를 통해 레이저 융착에 대한 효과성을 논하고자 한다. 먼저 영향인자를 소재, 장비, 구조로 설정하고, 각 인자별 수준을 나눠 검증을 실시하였다. 현재 크래시패드, PAB Door에 적용중인 양산소재를 바탕으로 각 소재별 투과율을 확인하고, 그에 따른 융착정도를 확인하였다. 융착정도는 각 시편별 인장력, 판단력 평가를 통해 검증하였다. 이러한 자료를 바탕으로 소재 인자별 크래시패드, PAB Door 제품을 제작하였고, 레이저장비 조건별(출력, 속도) T/O를 실시하였다. 제작된 제품에 대해 융착정도 및 이관수준 평가를 실시하였고, 이를 통해 최적화된 조건을 도출하였다. 이 같은 시험 기법을 통해서 Hard IP용 PAB Door 레이저 융착에 대한 설계인자별 영향도를 검증하고, 레이저 융착을 위한 설계 기준을 알아보고자 한다.
