342 EFFECTS OF MELATONIN ON PREIMPLANTATION DEVELOPMENT OF PORCINE PARTHENOGENETIC EMBRYOS

<P> In mammalian species, melatonin is a multi-functional pineal gland hormone that regulates several circadian and seasonal rhythms including reproduction. However, the melatonin study was not common as to the oocytes in the pig. Recently, we reported that exogenous melatonin has beneficial effects on nuclear and cytoplasmic maturation during porcine oocyte IVM and we also reported an existence of melatonin receptor on the cumulus cells and granulose cells (Kang JT et al. 2009 J. Pineal Res. 46(1), 22-28). In this study, as adding further experiments rather than our previous study, we investigated effect of exogeneous melatonin (10 ng mL-1) on the porcine oocytes and analyzed possible factors which can be responsible for that results. Oocytes were recovered by aspiration of slaughterhouse ovaries, and then matured in TCM-199 supplemented with EGF, insulin, pyruvate, cystine, and gonadotropin. Expression of apoptosis-related genes mRNA in oocytes cultured with melatonin were evaluated by real-time PCR (Exp 1), cumulus cell expansion on COC was assessed on the microscopes during in vitro maturation (Exp 2), and developmental effects between melatonin treatement group and non-treatment group on the in vitro culture of parthenogenetically activated oocytes was investigated (Exp 3). In results, oocytes matured with melatonin were assessed for the expression of apoptosis-related genes Bcl-xl (anti-apoptotic gene) and Bax (proapoptotic gene) by real-time quantitative PCR. Analysis of data showed that the expression of Bcl-xl was higher compared to the control while the expression of Bax was decreased relative to the control (P < 0.05). Cumulus cell expansion was evaluated under a stereomicroscope at 22 h, 44 h during IVM. Representative photomicrographs of porcine COC at the start of the IVM, after 22 h and 44 h treatment with melatonin, are shown in Figure. After 22 h of melatonin treatment, cumulus cells were visually expanded compared with non-treatment group. We analyzed significantly greater proportion of parthenogenetically activated oocytes developed to blastocyst when the IVM medium was supplemented with melatonin. Melatonin treatment in the IVM has consequently beneficial effect on the blastocyst formation rates on the development of porcine parthenogenetic embryos (15.4%) compared to non-treatment group (10.7%, P < 0.05). However, cleavage frequency was not affectedby the treatment. In conclusion, the present study demonstrated that melatonin had a beneficial effect on the development of parthenogenetically activated porcine embryos, probably through decreased apoptosis rate and increased cumulus cell expansion. A complete characterisation of the β-decay of neutron-rich nuclei can be obtained from the measurement of β-delayed gamma rays and, whenever the process is energetically possible, β-delayed neutrons. The accurate determination of the β-intensity distribution and the β-delayed neutron emission probability is of great relevance in the fields of reactor technology and nuclear astrophysics. A programme for combined measurements using the total absorption gamma-ray spectroscopy technique and both neutron counters and neutron time-of-flight spectrometers is presented.

In experiment 1, eighty crossbred steers (239±15 kg) were used in a 229-d experiment to evaluate the effects of increasing levels of enzymatically hydrolyzed yeast (EHY) cell wall in diets on growth performance feedlot cattle during periods of elevated ambient temperature. Treatments consisted of steam-flaked corn-based diets supplemented to provide 0, 1, 2, or 3 g EHY/hd/d. There were no effects on growth performance during the initial 139-d period. However, from d 139 to harvest, when 24-h temperature humidity index averaged 80, EHY increased dry matter intake (DMI) (linear effect, p<0.01) and average daily gain (ADG) (linear effect, p = 0.01). There were no treatment effects (p>0.10) on carcass characteristics. In experiment 2, four Holstein steers (292±5 kg) with cannulas in the rumen and proximal duodenum were used in a 4×4 Latin Square design experiment to evaluate treatments effects on characteristics of ruminal and total tract digestion in steers. There were no treatment effects (p>0.10) on ruminal pH, total volatile fatty acid, molar proportions of acetate, butyrate, or estimated methane production. Supplemental EHY decreased ruminal molar proportion of acetate (p = 0.08), increased molar proportion of propionate (p = 0.09), and decreased acetate:propionate molar ratio (p = 0.07) and estimated ruminal methane production (p = 0.09). It is concluded that supplemental EHY may enhance DMI and ADG of feedlot steers during periods of high ambient temperature. Supplemental EHY may also enhance ruminal fiber digestion and decrease ruminal acetate:propionate molar ratios in feedlot steers fed steam-flaked corn-based finishing diets.

This paper proposes a navigation method for manipulators which handle a surgical instrument or robotic platform introduced through a SILS multiport trocar. This method is based on a parallel position and force control which allow to the manipulator makes movements around the fulcrum point making the minimum force in the patient’s abdomen. In order to avoid the hysteresis effect due to the space between the instrument and the trocar when the instrument is inserted, a fuzzy gain scheduler has been designed. These algorithms have been implemented in a six degrees of freedom manipulator with a force sensor in the end effector. Finally, the experimental results are shown in order to demonstrate how it works in a real environment.

Palm oil production is among the highest worldwide, and it has been mainly used in the food industry and other commodities. Currently, a lot of palm oil production has been destined for the synthesis of biodiesel; however, its use in applications other than the food industry has been questioned. Thereby for a sustainable development, in this paper the use of palm oil of low quality for corrosion inhibitors synthesis is proposed. The performance of the synthesized inhibitors was evaluated by using electrochemical techniques such as open circuit potential measurements, linear polarization resistance and electrochemical impedance spectroscopy. The results indicate that the fatty amides from palm oil are excellent corrosion inhibitors with protection efficiencies greater than 98%. Fatty amides molecules act as cathodic inhibitors decreasing the anodic dissolution of iron. When fatty amides are added, a rapid decrease in the corrosion rate occurs due to the rapid formation of a molecular film onto carbon steel surface. During the adsorption process of the inhibitor a self-organization of the hydrocarbon chains takes place forming a tightly packed hydrophobic film. These results demonstrate that the use of palm oil for the production of green inhibitors promises to be an excellent alternative for a sustainable use of the palm oil production.
Monte Carlo Simulations for the Study of a Moderated Neutron Detector

This work presents the Monte Carlo simulations performed with the MCNPX and GEANT4 codes for the design of a BEta deLayEd Neutron detector, BELEN-20. This detector will be used for the study of beta delayed neutron emission and consists of a block of polyethylene with dimensions 90 × 90 × 80 cm ^3 and 20 cylindrical ^3He gas counters. The results of these simulations have been validated experimentally with a ^(252)Cf source in the laboratory at UPC, Barcelona. Also the first experiment with this detector has been carried out in November 2009 in JYFL, Finland. In this experiment the neutron emission probability after beta decay of the fission products ^(88)Br, ^(94,95)Rb and ^(138)I has been measured; this data is still under analysis. Simulations with MCNPX and GEANT4 have been performed in order to obtain the efficiency of the BELEN-20 detector for each of the above nuclei using the neutron energy distribution corresponding to each nucleus.

The cloning of canids was succeeded in 2005, several years after the birth of Dolly the sheep and also after the cloning of numerous other laboratory and farm animal species. The delay of successful somatic cell nuclear transfer (SCNT)was due to the unique reproductive characteristics of the female dogin comparison to other domestic mammals, such as ovulation of immature canine oocyte and a requirement of 25 days for the completion of meiosis within the oviduct (Holst & Phemister, 1971). When the technology for the recovery of in vivo matured oocyte was established, the application of cloning also became possible and cloned dog offspring were obtained. This report summarizes the progress of technical procedures that are required for cloning canids and the application of this technique. The first cloned dog, Snuppy, was achieved using an in vivo-matured oocyte which was enucleated and transferred with an adult skin cell of male Afghan hound. After establishment of a criterion of well-matured oocyte for the improvement of SCNT efficiency, we obtained three cloned female Afghan hound and a toy poodle cloned from 14 year-old aged Poodle using SCNT through this factor. To date, cloned dogs appeared to be normal and those that have reached puberty have been confirmed to be fertile. Through application of canine SCNT technique, first, we demonstrated that SNCT is useful for conserving the breed of endangered animal from extinction through cloning of endangered gray wolves using inter-species SCNT and keeping the pure pedigree through the cloning of Sapsaree, a Korean natural monument. Secondly, we showed possibility of human disease model cloned dog and transgenic cloned dog production through cloning of red fluorescent protein expressing dog. Finally, SCNT can be used for the propagation of valuable genotypes for making elite seed stock and pet dog. In summary, dog cloning is a reproducible technique that offers the opportunity to preserve valuable genetics and a potential step towards the production of gene targeted transgenic cloned dogs for the study of human diseases.
