<P><B>Abstract</B></P> <P> <I>Bacillus subtilis</I> P229 was isolated from <I>cheonggukjang</I>, a Korean soybean fermented food, and its probiotic properties and utility in starter cultures for soybean fermentation were investigated. <I>B. subtilis</I> P229 spores were stable when exposed to simulated gastric and bile conditions. <I>B. subtilis</I> P229 vegetative cells did not produce β-glucuronidase, were sufficiently susceptible to antibiotics, and adhered strongly to human intestinal epithelial cells. Moreover, this strain did not induce hemolysis or carry enterotoxin genes. <I>B. subtilis</I> P229 showed high levels of autoaggregation, and its coaggregation with pathogens depended on the species involved. It did not produce hazardous biogenic amines from histidine and ornithine. <I>B. subtilis</I> P229-fermented soybean extracts were prepared using 70% ethanol. Fermented-soybean extracts demonstrated greater antioxidant effects and total phenolic content than unfermented soybean extracts. 초등과학 영재교사와 일반교사의 과학교육에 대한 신념 비교

The purposes of this study is to compare the beliefs in the nature of science, science teaching and learning of the elementary teachers in charge of the gifted and the general teachers. For this study, a survey on beliefs of the nature of science, science teaching and learning was conducted to 88 elementary teachers for the gifted and 90 elementary general teachers. Data was analyzed by their academic career and major. The results of this study were as follows: There were no significant differences in beliefs in the nature of science and science teaching between the elementary science teachers in charge of the gifted and the general elementary teachers, but the former has a more constructivism in science learning than the latter. In the some sub-domains of the beliefs of the nature of science, science teaching and learning, there were statistically significant differences according to their academic career and major. Implications from findings of this study were suggested, such as recruiting and in-service training system for teachers in charge of the gifted.
반응표면분석법을 이용한 고려엉겅퀴로부터 항산화 성분의 초음파 추출 조건 최적화

본 연구에서는 고려엉겅퀴로부터 폴리페놀과 같은 항산화 성분을 효율적으로 추출하고 평가하는 데 있어 CCD를 이용하여 추출 조건을 최적화하였다. 각 종속변수를 최대화하는 최적 추출 조건은 추출 시간이 34.5분, 추출 온도가 63.7℃이며 에탄올은 62.2%인 조건이 산출되었고 최적화된 공정에 따라 TPC는 5.21 ㎎ GAE/g DM, TFC는 1.00 ㎎ QE/g DM, RSA는 86.7%로 예측되었다. 이를 검증하기 위해 실제 실험을 진행한 결과 TPC 5.45 ㎎ GAE/g DM, TFC 0.95 ㎎ QE/g DM과 RSA 84.2%로 예측값과 실험값의 합치도가 높아 RSM의 적합성을 확인하였다. 위 결과는 식품에 한정적으로 이용되던 고려엉겅퀴를 항산화 성분 추출 최적화를 통해 기능성 보조 식품으로 개발하거나 향후 인체질환의 약용성분으로서의 개발 가능성을 높여 보다 효과적인 이용을 통해 고부가가치 향상을 기대할 수 있다. 또한 국내에서 자생하는 고려엉겅퀴를 제품 개발에 활용할 시 수입 원료를 사용할 때보다 농가의 소득 증대와 기업의 원료수급 비용절감으로 인한 산업적 활용 가치가 증대될 것으로 사료된다. This study was conducted to optimize the ultrasound-assisted extraction (UAE) conditions for maximizing the responses, including total phenolic compound (TPC), total flavonoid compound (TFC), and radical scavenging activity (RSA) from Cirsium setidens using a response surface methodology (RSM). The following three variables were used to optimize the UAE process using the central composite method: extraction time (X₁: 5.0∼55.0 min), extraction temperature (X₂: 26.0∼94.0℃), and ethanol concentration (X₃: 0.0∼99.5%). The maximum yield of TPC (5.21 ㎎ GAE/g DM), TFC (1.00 ㎎ QE/g DM) and radical scavenging activity (86.7%) were obtained under the optimal conditions of 34.5 min, 63.7°C, and 62.2%. No significant differences were observed between the predicted and experimental values, indicating the suitability of the fitted model and the successful application of CCD. Based on these results, the antioxidant activity of the Cirsium setidens extract can be increased by optimizing the UAE conditions, and the extract can be used as a material for cosmetics, food, and medicine.

<P>Obesity has become a major health threat in developed countries. However, current medications for obesity are limited because of their adverse effects. Interest in natural products for the treatment of obesity is thus rapidly growing. Korean Medicine (KM) is characterized by the wide use of herbal formulas. However, the combination rule of herbal formulas in KM lacks experimental evidence. According to <I>Shennong's Classic of Materia Medica</I>, the earliest book of herbal medicine, <I>Veratrum nigrum</I> (VN) has antagonistic features against <I>Panax ginseng</I> (PG), and the PG-VN pair is strictly forbidden. In this study, we have shown the effects of PG, VN, and their combination on obesity in high-fat (HF) diet-induced obese mice and in 3T3-L1 cells. PG, VN, and PG-VN combination significantly reduced weight gain and the fat pad weight in HF diet-induced obese mice. They also significantly decreased lipid accumulation and the expressions of two major adipogenesis factors, PPAR<I><I>γ</I></I> and C/EBP<I><I>α</I></I>, in 3T3-L1 cells. In addition, the PG-VN combination had synergistic effects compared with the mixture of extracts of PG and VN on inhibition of PPAR<I><I>γ</I></I> and C/EBP<I><I>α</I></I> expressions at lower doses. These results indicate a new potential anti-obese pharmacotherapy and also provide scientific evidence supporting the usage of herbal combinations instead of mixtures in KM.</P>

<P>This study was performed in order to investigate the antiobese effects of the ethanolic extract of Veratri Nigri Rhizoma et Radix (VN), a herb with limited usage, due to its toxicology. An HPLC analysis identified jervine as a constituent of VN. By an Oil Red O assay and a Real-Time RT-PCR assay, VN showed higher antiadipogenic effects than jervine. In high-fat diet- (HFD-) induced obese C57BL/6J mice, VN administration suppressed body weight gain. The levels of peroxisome proliferator-activated receptor gamma (PPAR<I>γ</I>), CCAAT-enhancer-binding protein alpha (C/EBP<I>α</I>), adipocyte fatty-acid-binding protein (aP2), adiponectin, resistin, and LIPIN1 were suppressed by VN, while SIRT1 was upregulated. Furthermore, VN activated phosphorylation of the liver kinase B1- (LKB1-) AMP-activated protein kinase alpha- (AMPK<I>α</I>-) acetyl CoA carboxylase (ACC) axis. Further investigation of cotreatment of VN with the AMPK agonist AICAR or AMPK inhibitor Compound C showed that VN can activate the phosphorylation of AMPK<I>α</I> in compensation to the inhibition of Compound C. In conclusion, VN shows antiobesity effects in HFD-induced obese C57BL/6J mice. In 3T3-L1 adipocytes, VN has antiadipogenic features, which is due to activating the LKB1-AMPK<I>α</I>-ACC axis. These results suggest that VN has a potential benefit in preventing obesity.</P>

<P><B>Abstract</B></P> <P>This study aimed at evaluating the functional and probiotic properties of three lactic acid bacterial (LAB) strains isolated from kimchi. The selected LAB strains, which had potential probiotic functions, were identified by 16S rRNA sequence analysis as <I>Lactobacillus brevis</I> G1, <I>L</I>. <I>brevis</I> KU15006, and <I>Lactobacillus curvatus</I> KCCM 200173. All LAB strains were able to tolerate incubation at pH 2.5 with 0.3% pepsin for 3 h and with 0.3% Oxgall for 24 h and showed similar enzyme production levels, antimicrobial activities, and antibiotic susceptibilities. <I>L</I>. <I>brevis</I> G1 and KU15006 presented higher adhesion ability, auto-aggregation, and cell surface hydrophobicity than <I>Lactobacillus rhamnosus</I> KCTC 12202BP, a commercial strain used as positive control. All LAB strains showed 50–60% co-aggregation activity with selected foodborne pathogens. <I>L</I>. <I>brevis</I> KU15006 showed anti-adhesion activity against <I>Escherichia coli</I> and <I>Salmonella</I> Typhimurium. In addition, cell-free supernatant and cell-free extract from <I>L</I>. <I>brevis</I> KU15006 displayed the highest inhibitory activities against α-glucosidase. These results indicate that <I>L</I>. <I>brevis</I> KU15006 has the best properties, with pathogen antagonistic and antidiabetic activity, for use in probiotic products.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>Lactobacillus brevis</I> KU15006 was isolated under the harsh conditions in kimchi. </LI> <LI> <I>L. brevis</I> KU15006 showed high adhesion, hydrophobicity, and auto-aggregation. </LI> <LI> <I>L. brevis</I> KU15006 inhibited the adhesion of pathogens to an intestinal cell line. </LI> <LI> <I>L. brevis</I> KU15006 possessed high α-glucosidase inhibitory activity. </LI> </UL> </P>

This study was designed to isolate lactic acid bacteria (LAB) with ${\beta}$-glucosidase activity and probiotic properties from Korean fermented foods. Among nine isolates, four LAB strains had excellent survival rates at pH 2.5 with 0.3% (w/v) pepsin for 3 h and 0.3% (w/v) oxgall for 24 h. Four LAB strains did not produce ${\beta}$-glucuronidase and showed adhesion ability to HT-29 cells that was superior to that shown by the reference strain Lactobacillus rhamnosus GG. All four strains were sensitive to ampicillin, tetracycline, chloramphenicol, and doxycycline. These strains were identified as Leuconostoc mesenteroides H40, Lactobacillus plantarum FI10604, L. brevis FI10700, and L. perolens FI10842 by 16S rRNA gene sequence, respectively. It was found that L. perolens FI10842 produced the highest b-glucosidase activity (49.10 mU/mL). These results indicate that the four LAB strains could be used as potential probiotic. Especially L. perolens FI10842 could be used as a starter culture for fermentation.

The purpose of this study was to determine the effects of duration and timing of glucocorticoid treatment on the expansion and differentiation of porcine neonatal pancreas cell clusters (NPCCs) into β-cells. After transplantation of NPCCs, the ductal cyst area and β-cell mass in the grafts both showed positive and negative correlations with duration of dexamethasone (Dx) treatment. Pdx-1 and HNF-3β gene expression was significantly downregulated following Dx treatment, whereas PGC-1α expression increased. Pancreatic duct cell apoptosis significantly increased following Dx treatment, whereas proliferation did not change. Altogether, transdifferentiation of porcine NPCCs into β-cells was influenced by the duration of Dx treatment, which might have been due to the suppression of key pancreatic transcription factors. PGC-1α plays an important role in the expansion and transdifferentiation of porcine NPCCs, and the initial 2 weeks following transplantation of porcine NPCCs is a critical period in determining the final β-cell mass in grafts.
