Degradation of Lignocelluloses in Rice Straw by BMC-9, a Composite Microbial System

( Hongyan Zhao ),( Hai Ru Yu ),( Xu Feng Yuan ),( Ren Zhe Piao ),( Hu Lin Li ),( Xiao Fen Wang ),( Zong Jun Cui ) 한국미생물 · 생명공학회 2014 Journal of microbiology and biotechnology Vol.24 No.5

To evaluate the potential utility of pretreatment of raw biomass with a complex microbial system, we investigated the degradation of rice straw by BMC-9, a lignocellulose decomposition strain obtained from a biogas slurry compost environment. The degradation characteristics and corresponding changes in the bacterial community were assessed. The results showed that rapid degradation occurred from day 0 to day 9, with a peak total biomass bacterium concentration of 3.3 × 10(8) copies/ml on day 1. The pH of the fermentation broth declined initially and then increased, and the mass of rice straw decreased steadily. The highest concentrations of volatile fatty acid contents (0.291 mg/l lactic acid, 0.31 mg/l formic acid, 1.93 mg/l acetic acid, and 0.73 mg/l propionic acid) as well as the highest xylanse activity (1.79 U/ml) and carboxymethyl cellulase activity (0.37 U/ml) occurred on day 9. The greatest diversity among the microbial community also occurred on day 9, with the presence of bacteria belonging to Clostridium sp., Bacillus sp., and Geobacillus sp. Together, our results indicate that BMC-9 has a strong ability to rapidly degrade the lignocelluloses of rice straw under relatively inexpensive conditions, and the optimum fermentation time is 9 days.

The Oligomer Polyacrylic Acid Effect on Immobilization of Silver Nanoparticles onto Cotton Fabric to Achieve a Durably Antibacterial Function

        Lin Li,Panpan Duan,Qingbo Xu,XuJun Zhang,JiaNing Chen,Feiya Fu,HongYan Diao,Xiangdong Liu 한국섬유공학회 2020 Fibers and polymers Vol.21 No.9

Aiming to a durably antibacterial function, stable immobilization of silver nanoparticles (Ag NPs) on cottontextiles still represents challenging in textile industry. In the present work, by grafting oligomeric polyacrylic acid (PAA) ontocotton fiber surfaces, we effectively prepared the Ag NPs with controllable size on the fiber surface, which are of average sizeof 16.43 nm, and therefore achieved a desirable antibacterial durability. The oligomer PAA binder can give the modifiedfabrics with a remarkable antibacterial durability against washing, as the bacterial reduction rates against bothStaphylococcus aureus and Escherichia coli remained over 90 % even after 50 laundering cycles. Moreover, this graft toapproach using oligomeric PAA shows merits in maintaining the desired cotton properties such as flexibility, waterabsorptivity and vapor permeability. Finally, this finishing process ensures a reliable safety for human skin.

The interaction of serum albumin with ginsenoside Rh2 resulted in the downregulation of ginsenoside Rh2 cytotoxicity

        Lin, Yingjia,Li, Yang,Song, Zhi-Guang,Zhu, Hongyan,Jin, Ying-Hua The Korean Society of Ginseng 2017 Journal of Ginseng Research Vol.41 No.3

Background: Ginsenoside Rh2 (G-Rh2) is a ginseng saponin that is widely investigated because of its remarkable antitumor activity. However, the molecular mechanism by which (20S) G-Rh2 triggers its functions and how target animals avoid its cytotoxic action remains largely unknown. Methods: Phage display was used to screen the human targets of (20S) G-Rh2. Fluorescence spectroscopy and UV-visible absorption spectroscopy were used to confirm the interaction of candidate target proteins and (20S) G-Rh2. Molecular docking was utilized to calculate the estimated free energy of binding and to structurally visualize their interactions. MTT assay and immunoblotting were used to assess whether human serum albumin (HSA), bovine serum albumin (BSA), and bovine serum can reduce the cytotoxic activity of (20S) G-Rh2 in HepG2 cells. Results: In phage display, (20S) G-Rh2-beads and (20R) G-Rh2-beads were combined with numerous kinds of phages, and a total of 111 different human complementary DNAs (cDNA) were identified, including HSA which had the highest rate. The binding constant and number of binding site in the interaction between (20S)-Rh2 and HSA were $3.5{\times}10^5M^{-1}$ and 1, and those in the interaction between (20S) G-Rh2 and BSA were $1.4{\times}10^5M^{-1}$ and 1. The quenching mechanism is static quenching. HSA, BSA and bovine serum significantly reduced the proapoptotic effect of (20S) G-Rh2. Conclusion: HSA and BSA interact with (20S) G-Rh2. Serum inhibited the activity of (20S) G-Rh2 mainly due to the interaction between (20S) G-Rh2 and serum albumin (SA). This study proposes that HSA may enhance (20S) G-Rh2 water solubility, and thus might be used as nanoparticles in the (20S) G-Rh2 delivery process.

Downregulation of FoxM1 sensitizes nasopharyngeal carcinoma cells to cisplatin via inhibition of MRN-ATM-mediated DNA repair

        ( Dandan Li ),( Lin Ye ),( Yue Lei ),( Jie Wan ),( Hongyan Chen ) 생화학분자생물학회(구 한국생화학분자생물학회) 2019 BMB Reports Vol.52 No.3

Chemoresistance is the primary obstacle in the treatment of locally advanced and metastatic nasopharyngeal carcinoma (NPC). Recent evidence suggests that the transcription factor forkhead box M1 (FoxM1) is involved in chemoresistance. Our group previously confirmed that FoxM1 is overexpressed in NPC. In this study, we investigated the role of FoxM1 in cisplatin resistance of the cell lines 5-8F and HONE-1 and explored its possible mechanism. Our results showed that FoxM1 and NBS1 were both overexpressed in NPC tissues based on data from the GSE cohort (GSE12452). Then, we measured FoxM1 levels in NPC cells and found FoxM1 was overexpressed in NPC cell lines and could be stimulated by cisplatin. MTT and clonogenic assays, flow cytometry, γH2AX immunofluorescence, qRT-PCR, and western blotting revealed that downregulation of FoxM1 sensitized NPC cells to cisplatin and reduced the repair of cisplatin-induced DNA double-strand breaks via inhibition of the MRN (MRE11-RAD50-NBS1)-ATM axis, which might be related to the ability of FoxM1 to regulate NBS1. Subsequently, we demonstrated that enhanced sensitivity of FoxM1 knockdown cells could be reduced by overexpression of NBS1. Taken together, our data demonstrate that downregulation of FoxM1 could improve the sensitivity of NPC cells to cisplatin through inhibition of MRN-ATM-mediated DNA repair, which could be related to FoxM1-dependent regulation of NBS1. The enhancement of humidity sensing performance based on Eu-doped ZnO

        Shuguo Yu,Hongyan Zhang,Cunchong Lin,Mingjin Bian 한국물리학회 2019 Current Applied Physics Vol.19 No.2

In this work, a high performance impedance-type humidity sensor based on Europium-doped ZnO with abundant surface oxygen vacancy defects was synthesized by sol-gel method. Response of the Eu-doped ZnO with different molar ratio were investigated by exposing them to humidity environments in wide range of 11–95% RH at room temperature. The Eu-doped ZnO (2 mol%) exhibits a three orders impedance change, along with short response/ recovery time (5 s/19 s), low hysteresis and best linearity. Complex impedance spectra indicates that dopant Eu can enhance humidity sensing performance of ZnO, which is resulted from the introduction of Eu3+ ions into ZnO structure to produce more defects of surface oxygen vacancy and more active sites on the surface of ZnO. The results show that this is a feasible method to achieve high humidity sensing performance by Eu doped ZnO, which make it a promising candidate for humidity sensing materials and broaden the use of ZnO materials.

Secondary Voltage Control for Reactive Power Sharing in an Islanded Microgrid

        Qian Guo,Hongyan Wu,Liaoyuan Lin,Zhihong Bai,Hao Ma 전력전자학회 2016 JOURNAL OF POWER ELECTRONICS Vol.16 No.1

Owing to mismatched feeder impedances in an islanded microgrid, the conventional droop control method typically results in errors in reactive power sharing among distributed generation (DG) units. In this study, an improved droop control strategy based on secondary voltage control is proposed to enhance the reactive power sharing accuracy in an islanded microgrid. In a DG local controller, an integral term is introduced into the voltage droop function, in which the voltage compensation signal from the secondary voltage control is utilized as the common reactive power reference for each DG unit. Therefore, accurate reactive power sharing can be realized without any power information exchange among DG units or between DG units and the central controller. Meanwhile, the voltage deviation in the microgrid common bus is removed. Communication in the proposed strategy is simple to implement because the information of the voltage compensation signal is broadcasted from the central controller to each DG unit. The reactive power sharing accuracy is also not sensitive to time-delay mismatch in the communication channels. Simulation and experimental results are provided to validate the effectiveness of the proposed method.
