        Aberrant microRNAs Expression in CD133+/CD326+ Human Lung Adenocarcinoma Initiating Cells from A549

        Increasing evidence demonstrates that miRNAs are in-volved in the dysregulation of tumor initiating cells (TICs) in various tumors. Due to a lack of definitive markers, cell sorting is not an ideal separation method for lung adeno-carcinoma initiating cells. In this study, we combined pa-clitaxel with serum-free medium cultivation (inverse-induc-tion) to enrich TICs from A549 cells, marked by CD133/ CD326, defined features of stemness. We next investigated aberrant microRNAs in this subpopulation compared to normal cells with miRNA microarray and found that 50 miRNAs exhibited a greater than 2-fold change in expres-sion. As further validation, 10 miRNAs were chosen to perform quantitative RT-PCR on the A549 cell line and primary samples. The results suggest that aberrant ex-pression of miRNAs such as miR-29ab, miR-183, miR-17-5p and miR-127-3P may play an important role in regulat-ing the bio-behavior of TICs.

        method for vibration isolation of a vertical axis automatic washing machine with a hydraulic balancer

        This paper discusses a method for vibration isolation of a vertical axis automatic washing machine with a hydraulic balancer. First, a way to isolate vibration through a small amplitude of the suspension rod’s axial force is proposed, and a base circle of a cone along which the lower joint of a suspension rod moves is discussed. Based on the circle, a geometric constraint involving the slant angle of the suspension rod, the deflection angle of the washing/spinning assembly, the suspension radius of the tub and the eccentricity of the system at the steady state is derived. Considering that the trace along which the suspension rod moves is also affected by the dynamics of the system, a governing equation satisfying the equilibrium conditions of the centrifugal forces and torques is obtained. Combing the geometric constraint and governing equation together achieves a general governing equation for vibration isolation of the system. Finally, the general governing equation is proven by simulations, and the relations between the optimal installing height and several parameters are discussed.

        Hygrothermal effects on the vibration and stability of an initially stressed laminated plate

        The influence of hygrothermal effects on the vibration frequency and buckling load of a shear deformable composite plate with arbitrary initial stresses was investigated. The governing equations of the effects of humid, thermal and initial stresses are established using the variational method. The material properties of the composite plate are affected by both temperature and moisture. The initial stress is taken to be a combination of uniaxial load and pure bending in a hygrothermal environment. The influence of various parameters, such as the fiber volume fraction, temperature, moisture concentration, length/thickness ratios, initial stresses and bending stress ratio on the vibration and stability of the response of a laminated plate are studied in detail. The behavior of vibration and stability are sensitive to temperature, moisture concentration, fiber volume fraction and initial stresses.

        Hygrothermal effects on dynamic instability of a laminated plate under an arbitrary pulsating load

        This paper studies the static and dynamic characteristics of composite plates subjected to an arbitrary periodic load in hygrothermal environments. The material properties of composite plates are depended on the temperature and moisture. The governing equations of motion of Mathieu-type are established by using the Galerkin method with reduced eigenfunction transforms. A periodic load is taken to be a combination of axial pulsating load and bending stress in the example problem. The regions of dynamic instability of laminated composite plates are determined by solving the eigenvalue problems based on Bolotin’s method. The effects of temperature rise and moisture concentration on the dynamic instability of laminated composite plates are investigated and discussed. The influences of various parameters on the instability region and dynamic instability index are also investigated. The numerical results reveal that the influences of hygrothermal effect on the dynamic instability of laminated plates are significant.

        Molecular Biology and Omics : Direct Evaluation of the Effect of Gene Dosage on Secretion of Protein from Yeast Pichia pastoris by Expressing EGFP

        Increasing the gene copy number has been commonly used to enhance the protein expression level in the yeast Pichia pastoris. However, this method has been shown to be effective up to a certain gene copy number, and a further increase of gene dosage can result in a decrease of expression level. Evidences indicate the gene dosage effect is product-dependent, which needs to be determined when expressing a new protein. Here, we describe a direct detection of the gene dosage effect on protein secretion through expressing the enhanced green fluorescent protein (EGFP) gene under the direction of the α-factor preprosequence in a panel of yeast clones carrying increasing copies of the EGFP gene (from one to six copies). Directly examined under fluorescence microscopy, we found relatively lower levels of EGFP were secreted into the culture medium at one copy and two copies, substantial improvement of secretion appeared at three copies, plateau happened at four and five copies, and an apparent decrease of secretion happened at six copies. The secretion of EGFP being limiting at four and five copies was due to abundant intracellular accumulation of proteins, observed from the fluorescence image of yeast and confirmed by western blotting, which significantly activated the unfolded protein response indicated by the up-regulation of the BiP (the KAR2 gene product) and the protein disulfide isomerase. This study implies that tagging a reporter like GFP to a specific protein would facilitate a direct and rapid determination of the optimal gene copy number for high-yield expression.

        Percutaneous radiologic gastrostomy in patients with amyotrophic lateral sclerosis: A safe and effective technique

        Background: Percutaneous radiologic gastrostomy (PRG) was considered as an alternative technique for long-term enteral nutrition, and the current study is aimed to evaluate the feasibility and safety of this technique in patients with amyotrophic lateral sclerosis (ALS) at a single medical center. Methods: From July 2017 to October 2020, a total of 14 patients underwent PRG with ALS were included in this retrospective study with a median age of 64.0 years, and 78.6% were male. The procedure comprised a dilation of the stomach via a nasogastric catheter, followed by puncture and gastrostomy tube placement under fluoroscopic guidance. The technical success rate and clinical outcomes were recorded over 3 months following the procedure. Results: The technical success rate was 100%. During the follow-up period, minor complications were reported in 2 of patients (14.3%) including superficial skin infection and early tube block. Neither major complications nor mortality were observed. Body mass index of the patients increased significantly from 16.4 ± 2.1 kg/m2 to 17.1 ± 2.0 kg/m2 (t = –13.77; P < 0.001), and the albumin level increased significantly from 37.5 ± 2.3 g/L to 41.8 ± 1.6 g/L (t = –8.82, P < 0.001). Conclusion: PRG is a relatively safe and effective method for ALS patients, and deserves widespread clinical acceptance.

        Nonlinear dynamics analysis of multi-cutting wire electrode in WEDM-HS subjected to working fluid considering the effect of debris

        A reasonable dielectric fluid velocity and better exclusion of debris are very important to obtain a stable machining performance in multi-cutting of WEDM-HS. We investigated the dynamic characteristics of multi-cutting wire electrode in WEDM-HS subjected to working fluid considering the effect of debris. First, a fluid-solid coupling nonlinear vibration model is derived by analyzing the forces acting on the multi-cutting wire which locate in the dielectric fluid. The additional axial force arising from the transverse vibration of the multi-cutting wire is considered as a nonlinear term in the model. Considering that the debris accumulation in discharge gap severely influences the stability of multi-cutting wire electrode, the relationship between the debris volume concentration and debris-working fluid two-phase flow drag coefficient is obtained using the Einstein two-phase flow viscosity theory. Finally, the influences of several parameters on the multi-cutting wire displacement are discussed in detail.

        Efficacy of Epigallocatechin-3-Gallate and Amla (Emblica officinalis) Extract for the Treatment of Diabetic-Uremic Patients

        Uremic patients with diabetes suffer from high levels of oxidative stress due to regular hemodialysis therapy (neutrophil activation induced by hemo-incompatibility between the hemodialyser and blood) and complications associated with diabetes. Several plasma biomarkers were screened in 13 uremic diabetic patients after receiving the mixture of (–)-epigallocatechin gallate (EGCG), a major component of green tea extract, and Amla extract (AE), from Emblica officinalis, the Indian gooseberry, for 3 months. We found that oral administration of a 1:1 mixture of EGCG and AE for 3 months significantly improved antioxidant defense as well as diabetic and atherogenic indices in uremic patients with diabetes. Furthermore, no significant changes in hepatic function, renal function, or inflammatory responses were observed. These results suggest that a 1:1 combination of EGCG and AE is a safe and effective treatment for uremic patients with diabetes.
