        ESA in High-order Descriptor Linear Systems via Output Feedback

        This paper considers eigenstructure assignment (ESA) in high-order descriptor linear systems via output feedback. Parametric expressions for the left and right closed-loop eigenvectors associated with the finite closed-loop eigenvalues and two simple and complete parametric solutions to the feedback gain matrix are obtained on the basis of the parametric solution to the generalized high-order Sylvester matrix equation. This approach does not impose any restriction on the closed-loop eigenvalues. The numerical example illustrates the effectiveness of the proposed approach.

        Potential roles of two Cathepsin genes, DcCath-L and DcCath-O in the innate immune response of Diaphorina citri

        Cathepsins belong to a group of mammalian papain-like cysteine proteases that play an important role in the insect immune response. In the present study, we identified two cathepsin genes from the Diaphorina citri genome database, cathepsin-L (DcCath-L) and cathepsin-O (DcCath-O). DcCath-L encodes a DcCath-L protein consisting of 348 amino acid residues, and DcCath-O encodes a DcCath-O protein consisting of 329 amino acid residues. DcCaths contain two conserved domains, the Inhibitor_I29 and Pept_C1 domains. Phylogenetic tree analysis revealed that DcCath-L and DcCath-O were divided into two different groups: Cathepsin-L and Cathepsin-O. Reverse transcription quantitative polymerase chain reaction analysis showed that both DcCath-O and DCCath-L were highly expressed in the midgut, while lower expression was observed in other tissues. Developmental stage expression analysis suggested that DcCath-O was mainly expressed in third instar nymph and adult, and DcCath-L was highly expressed in first and fourth instar nymph. Following exposure to two different heat-killed bacteria (Escherichia coli and Staphylococcus aureus) and Candidatus Liberibacter asiaticus, the expression of DcCath-O and DcCath-L was significantly increased and showed differential expression patterns at different time points. In addition, silencing of DcCath-L obvious affected the gene expression of members of the Toll pathway, while knock down of DcCath-L has no significantly influence. Overall, these data provide valuable information for further functional studies of D. citri cathepsins.

        Synergistic Effects of Leflunomide and Benazepril in Streptozotocin-Induced Diabetic Nephropathy

        <P>Abstract</P><P><B><I>Background:</I></B> Leflunomide (LEF) and benazepril have renoprotective effects on diabetic nephropathy (DN) through their anti-inflammatory and anti-fibrotic activities. This study investigated whether combined treatment using LEF and benazepril affords superior protection compared with the respective monotherapies. <B><I>Methods:</I></B> Diabetes was induced with streptozotocin (STZ, 65 mg/kg) by intraperitoneal injection in male Wistar rats. Two weeks after STZ injection, diabetic rats were treated daily for 12 weeks with LEF (10 mg/kg), benazepril (10 mg/kg), or a combination of both. Basic parameters (body weight, fasting blood glucose level, and 24 h urinary protein excretion), histopathology, inflammatory [inflammatory cell infiltration (ED-1), monocyte chemoattractant protein-1 (MCP-1), and Toll-like receptor-2 (TLR-2)] and glomerulosclerotic factors [transforming growth factor-β<SUB>1</SUB> (TGF-β<SUB>1</SUB>) and connective tissue growth factor (CTGF)], and oxidative stress (8-hydroxy-2'-deoxyguanosine, 8-OHdG) were studied. <B><I>Results:</I></B> Benazepril or LEF treatment significantly prevented body weight loss and 24 h urinary protein excretion induced by diabetes; combined treatment with LEF and benazepril further improved these parameters compared with giving each drug alone (all p < 0.01). Increased expression of inflammatory (MCP-1 and TLR-2) and glomerulosclerotic (TGF-β<SUB>1</SUB> and CTGF) factors in diabetic rat kidney was reduced by treatment with either LEF or benazepril and was further reduced by the combined administration of the two drugs (p < 0.01). These effects were accompanied by suppression of urinary 8-OHdG excretion. There was no significant between-group difference in blood glucose level. <B><I>Conclusions:</I></B> LEF treatment lessens DN, and combined treatment with LEF and benazepril provides synergistic effects in preventing DN.</P><P>© 2014 S.         The Effect of Amino Density on the Attachment, Migration, and Differentiation of Rat Neural Stem Cells In Vitro

        Artificial extracellular matrices play important roles in the regulation of stem cell behavior. To generate materials for tissue engineering, active functional groups, such as amino, carboxyl, and hydroxyl, are often introduced to change the properties of the biomaterial surface. In this study, we chemically modified coverslips to create sur-faces with different amino densities and investigated the adhesion, migration, and differentiation of neural stem cells (NSCs) under serum-free culture conditions. We observed that a higher amino density significantly promoted NSCs attach-ment, enhanced neuronal differentiation and promoted excitatory synapse formation in vitro. These results indicate that the amino density significantly affected the biological behavior of NSCs. Thus, the density and impact of functional groups in extracellular matrices should be considered in the research and development of materials for tissue engineering.

        Properties and Secondary Structure of Tannase from Penicillium herquei

        A tannase with a molecular mass of 72 kDa was obtained from Penicillium herquei isolated from valonia acorns following fermentation in a 5 L bioreactor. This tannase showed optimum activity at pH 6.0 and 30°C. The enzyme was inhibited by Fe^(3+), Zn^(2+), dithiothrietol (DTT),β-mercaptoethanol, formaldehyde, and ethanol, and induced by K^+, Mn^(2+), Tween 80, and Triton X-100. The Michaelis constant (K_m) and the second-order constant (k_(cat)/K_m)values of the tannase for propyl gallate (PG) were 0.62mM and 174.1 mM/sec. The circular dichroism (CD)spectra indicated that the secondary structure of the tannase contained 14% α helix, 32.4% anti-parallel β-sheet, 4.8%β-sheet, 18.8% β-turn, and 30% random coil. Native tannase in ultrapure water manifested as spherical nano-particle aggregates with diameters ranging from 50 to 300 nm determined by atomic force microscopy (AFM).

      • Prediction of Lung Cancer Based on Serum Biomarkers by Gene Expression Programming Methods

        In diagnosis of lung cancer, rapid distinction between small cell lung cancer (SCLC) and non-small cell lung cancer (NSCLC) tumors is very important. Serum markers, including lactate dehydrogenase (LDH), C-reactive protein (CRP), carcino-embryonic antigen (CEA), neurone specific enolase (NSE) and Cyfra21-1, are reported to reflect lung cancer characteristics. In this study classification of lung tumors was made based on biomarkers (measured in 120 NSCLC and 60 SCLC patients) by setting up optimal biomarker joint models with a powerful computerized tool - gene expression programming (GEP). GEP is a learning algorithm that combines the advantages of genetic programming (GP) and genetic algorithms (GA). It specifically focuses on relationships between variables in sets of data and then builds models to explain these relationships, and has been successfully used in formula finding and function mining. As a basis for defining a GEP environment for SCLC and NSCLC prediction, three explicit predictive models were constructed. CEA and NSE are requentlyused lung cancer markers in clinical trials, CRP, LDH and Cyfra21-1 have significant meaning in lung cancer, basis on CEA and NSE we set up three GEP models-GEP 1(CEA, NSE, Cyfra21-1), GEP2 (CEA, NSE, LDH), GEP3 (CEA, NSE, CRP). The best classification result of GEP gained when CEA, NSE and Cyfra21-1 were combined: 128 of 135 subjects in the training set and 40 of 45 subjects in the test set were classified correctly, the accuracy rate is 94.8% in training set; on collection of samples for testing, the accuracy rate is 88.9%. With GEP2, the accuracy was significantly decreased by 1.5% and 6.6% in training set and test set, in GEP3 was 0.82% and 4.45% respectively. Serum Cyfra21-1 is a useful and sensitive serum biomarker in discriminating between NSCLC and SCLC. GEP modeling is a promising and excellent tool in diagnosis of lung cancer.

      • HDAC6 siRNA Inhibits Proliferation and Induces Apoptosis of HeLa Cells and its Related Molecular Mechanism

        Objective: To investigate the effects of histone deacetylase 6 (HDAC6) siRNA on cell proliferation and cell apoptosis of the HeLa cervical carcinoma cell line and the molecular mechanisms involved. Methods: Division was into three groups: A, the untreated group; B, the control siRNA group; and C, the HDAC6 siRNA group. Lipofectamine 2000 was used for siRNA transfection, and Western blot analysis was used to determine the protein levels. Cell proliferation and apoptosis were characterized using a CCK-8 assay and flow cytometry, respectively. Results: HDAC6 protein expression in the HDAC6 siRNA-transfection group was significantly lower (P < 0.05) than in the untreated and control siRNA groups. The CCK-8 kit results demonstrated that the proliferation of HeLa cells was clearly inhibited in the HDAC6 siRNA transfection group (P < 0.05). In addition, flow cytometry revealed that the early apoptotic rate ($26.0%{\pm}0.87%$) was significantly elevated (P < 0.05) as compared with the untreated group ($10.6%{\pm}1.19%$) and control siRNA group ($8.61%{\pm}0.98%$). Furthermore, Western blot analysis indicated that bcl-2 protein expression in the HDAC6 siRNA-transfection group was down-regulated, whereas the expression of p21 and bax was up-regulated. Conclusion: HDAC6 plays an essential role in the occurrence and development of cervical carcinoma, and the down-regulation of HDAC6 expression may be useful molecular therapeutic method.

      • Decision on Quality Investment Level Under Moral Hazard Environment

        Moral hazard and adverse selection often exist in asymmetric information environment. In this paper, quality investment decision problem is studied under moral hazard. A basic model for quality investment level decision is developed with the supplier as a principal and the buyer as an agent. And then we regard the supplier and the buyer`s rational limitations to set up a model when the buyer`s quality evaluation and processing activities are hidden. The model is optimized and the results under different backgrounds are discussed and compared. Results show that the buyer`s quality evaluation level and processing level are mostly influenced by the supplier`s quality assurance payment. Both the supplier and the buyer choose different quality investment levels under moral hazard because of the supplier`s payment to the buyer in case of internal failure and external failure.

      • Design of Acquisition and Control System of Granary Supervisory Control and Data Acquisition System

        Grain stored in granary is easily spoiled due to accumulated hear or humidity if not monitored and maintained properly. Supervisory control and data acquisition (SCADA) system is commonly used in granary to provide real-time storage information. Major functionalities in the granary SCADA system is usually provided by an acquisition and control system based on single chip computer installed inside or near the granary. Based on theoretical and commissioning data, the design of the acquisition and control system was introduced in this paper. The acquisition and control system is able to acquire real-time data of temperature, humidity and concentration of CO2 inside the granary. It also provides the functionalities of open/shut the granary windows, as well as turning on/off the high-power roller shutter doors and blowers. Users are able to obtain real-time monitored data and control the system accordingly.

      • 분류식 하수관로에서 월류수의 수질 특성

        The purpose of this sudy is to get characteristics of water quality in separated sewer overflows. In dry weather the water quality of sewage in the outfall of the separated sewer was an average BOD of 2.7 mg/L, CODmn of 4.9 mg/L, TN of 3.8 mg/L, TP of 1.9 mg/L, and SS of 5.3 mg/L, Because the quality except for TP was less than the water quality standard of wastewater treatment plant (WTP), a prevention facility was needed to prevent the sewage inflow to interceptor. When the separated sewer overflows (SSO) were overflowed, the maximum pollutant load was be found to be about from 2.7 to 5.4 times as a BOD base, about from 3.2 to 4.6 times as a SS base higher than average load. There was a big difference according to rainfall intensity and rainfall continuous time. In case the quality of SSO was less than water quality standard or water quality in dry season, SSO was required to discharge to the stream not to inflow to WTP by operating the floodgate in the outfall with water quality data using sensors, such as turbidity sensor, etc. Because removal efficiency by an intercepter load was calculated as low as about 10~20% as a BOD base, removal efficiency could be increased by installing a separated wall within the sewer.
