        High‑Temperature Deformation Behavior of MnS in 1215MS Steel

        The effect of manganese sulfide (MnS) inclusions on the machinability of free-cutting steel is based on their morphology,size and distribution. Furthermore, the plasticity of MnS is high during the hot working caused different characterizationof MnS. In this study, the deformation behavior of MnS in 1215MS steel after a thermomechanical process was investigatedat 1323 K. The microstructures of MnS inclusions were characterized by optical microscopy, scanning electronmicroscopy, energy-dispersive spectrometry, and electron backscattering diffraction (EBSD). As the thickness reductionof the inclusions increased from 10 to 70%, their average aspect ratio increased from 1.20 to 2.39. In addition, the deformabilityof MnS inclusions was lower than that of the matrix. The possible slip systems of A, B, C, and D plane traces werē10̄1̄101 , 10̄ 1 [101], (011) 01̄ 1 , and (110) 1̄10 . Furthermore, the EBSD measurements suggested that slip planes inMnS inclusions occur on {110} planes.

      • MicroRNA-101 Inhibits Cell Proliferation, Invasion, and Promotes Apoptosis by Regulating Cyclooxygenase-2 in Hela Cervical Carcinoma Cells

        Aim: Although aberrant miRNA expression has been documented, altered miR-101 expression in cervical cancer and its carcinogenic effects and mechanisms remain unexplored. The aim of our study was to investigate the role of miR-101 alteration in cervical carcinogenesis. Methods: Expression of miR-101 was examined by quantitative real-time reverse transcriptase PCR (qRT-PCR) in Hela cells. After modulating miR-101 expression using miR-101 mimics, cell growth, apoptosis and proliferation, and migration were tested separately by MTT or flow cytometry and cell wound healing assay and protein expression was detected by qRT-PCR. The expression of COX-2 in Hela cell was also examined by immunohistochemical staining and the correlation with miR-101 expression was analysed. Results: The miR-101 demonstrated significantly low expression in Hela cell. When we transfected miR-101 mimics into Hela cells, the modulation of miR-101 expression remarkably influenced cell proliferation, cycling and apoptosis: 1) The expression of microRNA-101 tended to increase after transfection; 2) Overexpression of miR-101 was able to promote cell apoptosis, the apoptosis rate being markedly higher (97.6%) than that seen pre-transfection (12.2%) (P<0.05); 3) The miR-101 negatively regulates cell migration and invasion, scratch results being lower ($42.7um{\pm}2um$) than that observed pre-transfection ($181.4um{\pm}2um$); 4) miRNA-101 inhibits the proliferation of Hela cells as well as the level of COX-2 protein, which was negatively correlated with miR-101 expression. Conclusions: Overexpression of miR-101 has obvious inhibitory effects on cell proliferation, migration and invasion. Thus reduced miR-101 expression could participate in the development of cervical cancer at least partly through loss of inhibition of target gene COX-2, which probably occurs in a relative late phase of carcinogenesis. Our data suggest an important role of miR-101 in the molecular etiology of cancer and indicate potential application of miR-101 in cancer therapy.

      • CONSTRUCTING CONFIGURATIONAL INDICATORS FOR CREATING INDUSTRIAL TOURISM VALUE: A TOURISM-VALUE-CHAIN APPLICATION

        Value creation is essential for successfully operating a firm and becomes a critical issue for realizing cross-firm competition and industrial performance distinctions. Tourism scholars have viewed tourism value-chain (TVC) as a useful tool to measure the efficiency and effectiveness of performance through controlling the impact factors in tourism products and services. However, few extant studies focus on the works of performance measurement in the tourism industry. This study applies Yilmaz and Bititci’s (2006 a, b) concept to develop novel performance measurement and value creation indicators for industrial tourism and customers through a crisp-set qualitative comparative analysis (QCA). The QCA approach considers asymmetric relationships—reporting conditions that are sufficient or necessary to cause an outcome condition. QCA allows exploring the causal-effect relationship between the variables. In-depth interview with five senior managers and five customers were conducted. The QCA results showed that industrial tourism and customer performance indicators have complex links with value creation. The main theoretical contributions of this study extends through four stages of TVC on both manufacturing and industrial tourism, developing causal-effect connections of industrial tourism, and customer performance on value creation. The study describes how proprietors and service providers promoted brand identification and firm’s reputation for achieving industrial tourism value. The findings beneficially provide practical implications for proprietors and customers for applying performance indicators once aiming to develop an appropriate marketing strategy and create industrial tourism and customer values.

        Study of co-excited green emission of Tb3+, Ce3+ and Gd3+ in yttrium aluminum garnet

        YAG : Tb3+, Ce3+, Gd3+ nano-phosphors derived from a sol-gel chemistry have been successfully synthesized and characterized by TG/DTA, XRD, SEM, and spectrometer. The results show that the phosphors have a uniform particle size distribution ranging 30-50 nm. The value of the lattice constant increased with an increase of the Gd3+ content. Tb3+ →Ce3+, Ce3+ →Tb3+ and Gd3+ →Ce3+ energy transfer existed in the co-excited system. The luminescence intensity was controlled by the concentration and ratio of the co-dopants. A small quantity of Gd3+ and Ce3+ evidently increased the green emission (5D4→7F5) of Tb3+.       • Optomizing Transfection Efficiency of Cervical Cancer Cells Transfected by Cationic Liposomes Lipofectamine<sup>TM</sup>2000

        Background: Currently, cationic liposome has become the commonly used vehicles for gene transfection. Furthermore, one of the most significant steps in microRNAs expression studies is transferring microRNAs into cell cultures successfully. In this study we aim to approach the feasibility of transfection of cervical cancer cell lines mediated by liposome and to obtain the optimized transfection condition for cervical cancer cell lines. Materials and Methods: $Lipofectamine^{TM}2000$ as the carrier, miR-101 mimic was transfected into Hela cells and Siha cells. Using green fluorescent protein as reporter gene, to set different groups according to cell seeding density, the amount of miRNA, miRNA and the proportion of Liposomes, Whether to add serum into medium to study their impact on the liposomal transfection efficiency. Finally, MTT assay was used to analyze the relative minimal cell toxicity of liposome reagents. Results: The seeding density of Hela cell line and Siha are $1.5{\times}10^4$ (per well of 24 well plates), miRNA amount is 1ul of both, the ratio of miRNA and liposome is 1:0.5 of Hela cell line; 1:0.7 of Siha cell line respectively, after 24 hours we can get the highest transfection efficiency. Compared with serum medium, only Siha cells cultured with serum-free medium obtained higher transfection efficiency before transfection (P<0.01). MTT assay showed that according to the above conditions which has the lowest cytotoxicity. Conclusions: The method of Liposome to transfected is a suitable way and it can be an efficient reagent for miRNA delivery for Hela cells and Siha cells in vitro. It may serve as a reference for the further research or application.

        De novo design of a novel AIE fluorescent probe tailored to autophagy visualization via pH manipulation

        Macroautophagy is an essential cellular self-protection mechanism, and defective autophagy has been considered to contribute to a variety of diseases. During the process, cytoplasmic components are transported via autophagosomes to acidic lysosomes for metabolism and recycling, which represents application niches for lysosome-targeted fluorescent probes. Additionally, in view of the complexity of the autophagy pathway, it entails more stringent requirements for probes suitable for monitoring autophagy. Meanwhile, aggregation-induced emission (AIE) fluorescent probes have been impressively demonstrated in the biomedical field, which bring fascinating possibilities to the autophagy visualization.We reported a generalizable de novo design of a novel pH-sensitive AIE probe ASMP-AP tailored to lysosome targeting for the interpretation of autophagy. Firstly, the theoretical calculation was carried out followed by the investigation of optical properties. Then, the performance of ASMP-AP in visualizing autophagy was corroborated by starvation or drugs treatments. Furthermore, the capability of ASMP-AP to monitor autophagy was demonstrated in ex vivo liver tissue and zebrafish in vivo.ASMP-AP displays a large stokes shift, great cell permeability and good biocompatibility. More importantly, ASMP-AP enables a good linear response to pH, which derives from the fact that its aggregation state can be manipulated by the acidity. It was successfully applied for imaging autophagy in living cells and was proved capable of monitoring mitophagy. Moreover, this novel molecular tool was validated by ex vivo visualization of activated autophagy in drug-induced liver injury model. Interestingly, it provided a meaningful pharmacological insight that the melanin inhibitor 1-phenyl-2-thiourea (PTU)-induced autophagy was clearly presented in wild-type zebrafish.ASMP-AP offers a simple yet effective tool for studying lysosome and autophagy. This is the first instance to visualize autophagy in zebrafish using a small-molecule probe with AIE characters, accurate lysosome targeting and simultaneous pH sensitivity. Ultimately, this novel fluorescent system has great potential for in vivo translation to fuel autophagy research.

      • 중국 공무항공시장 및 고객지향적 항공서비스에 대한 탐색적 연구

        중국 경제가 빠르게 발전하면서 인해 중국 공무(개인) 항공기는 더욱더 중요한 교통수단으로 활용되고 있다. 공무 항공기 서비스는 전문화 및 고효율화를 추구하면서 비즈니스 여행의 편리성을 최대한 보장하는 가장 적합한 수단으로 주목받고 있다. GARNOC가 발표한 <2016년 중국 공무 항공기 리포트>에 의하면 2016년 3월말까지 중국 대륙에 전체 218대의 공무항공기가 운행되고 있으며, 향후 10년간에 중국의 공무항공기는 1500대 정도로 전망되는 등 폭발적인 성장을 하여 미국에 이어 제2대 공무항공기 시장으로 발전해 나갈 것으로 예상되고 있다. 2016년 5월4일에 중국 국무총리 리커챵은 중국 마지막 조단위 위엔급 규모의 시장인 통용항공산업이 곧 개방될 것이라고 선언했으며, 이 시장은 경쟁이 치열한 전통산업보다 잠재력이 크고, 수익율도 높은 시장이라고 전망되고 있다. 그러나 중국 공무항공기 서비스 사업의 현실을 살펴보면 고급서비스산업으로써 발전 잠재력은 크다고 할 수 있으나, 전망과는 달리 최근 몇 년간에 발전 속도가 점차 느려지고 있음을 볼 수 있다. 이에 따라 본 연구에서는 중국 공무 항공기의 시장현황을 파악하고 발전에 따른 문제점을 찾아보고자 한다. 대표적인 기업들이 성장의 정체기에 빠지면서 생긴 산업 발전의 문제점을 알아보고, 새로운 도약점을 어떻게 찾아나가는 지에 대하여 살펴보고자 한다. 공무항공기 서비스 기업은 거액으로 항공기를 구매해야 함에도 불구하고, 중국 공무항공기 시장의 현황이나 정책의 변동성, 사회경제적 기반의 취약성, 기술 및 관련 서비스 산업의 미완전성등의 위험을 모두 감수해야 하는 등의 여러 가지 문제를 극복해 나가야 하는 어려움을 겪고 있다. 이러한 문제점을 해결해야만 중국 공무항공기 시장의 잠재력을 극대화시킬 수 있으며 지속적으로 성장해 나갈 수 있는 것이다. 본 연구는 중국 공무항공기 사업의 현황 및 발전 역사에 대하여 고찰하면서 동시에 공무항공기 문제점을 정리하고자 하며, 실제 공무항공기 항공서비스 사례를 통하여, 중국 부유층이 원하는 항공서비스를 파악하고, 문제점과 개선방안을 제시하면서 향후 중국 공무항공기 서비스가 어떻게 성장해 나갈 수 있을 것인지에 대하여 시사점을 도출하였다. 중국 대표적인 항공사인 하이난항공 산하 공무항공기 전문기업을 사례를 분석하였으며, 이분석 결과를 살펴보면, 중국 공무항공기 사업은 항공기 대여서비스에서부터 항공기 구매와 위탁관리 등을 수행하고 있다. 또한 전문화에서부터 고객맞춤형 특별주문제작까지, 국제표준화에서부터 현지화 서비스까지 폭넓은 서비스를 제공하고 있는 것을 찾아볼 수 있다. 이러한 사례 등을 살펴볼 때 미래 중국 공무항공기 시장은 고객의 가치를 최대화 시킬 수 있는 맞춤형 서비스를 선호할 것으로 전망되며, 차별화된 장기적 관점의 공무항공기 서비스를 제공하는 것이 고부가가치를 창출할 수 있을 것으로 전망하였다.

        Plastification procedure of laterally-loaded steel bars under a rising temperature

        This paper investigates the structural responses of axially restrained steel beams under fire conditions by a nonlinear finite element method. The axial restraint is represented by a linear elastic spring. Different parameters which include beam slenderness ratio, external load level and axial restraint ratio are investigated. The process of forming a mid-span plastic hinge at the mid-span under a rising temperature is studied. In line with forming a fully plastic hinge at mid-span, the response of a restrained beam under rising temperature can be divided into three stages, viz. no plastic hinge, hinge forming and rotating, and catenary action stage. During catenary action stage, the axial restraint pulls the heated beam and prevents it from failing. This study introduces definitions of beam limiting temperature $T_{lim}$, catenary temperature $T_{ctn}$ and warning time $t_{wn}$. Influences of slenderness ratio, load level and axial restraint ratio on $T_{lim}$, $T_{ctn}$ and $t_{wn}$ are examined.

      • Streptomyces zhihengii sp. nov., isolated from rhizospheric soil of Psammosilene tunicoides

        <P>An actinomycete strain, designated YIM T102(T), was isolated from the rhizospheric soil of Psammosilene tunicoides W. C. Wu et C. Y. Wu collected from Lijiang, Yunnan Province, China. The taxonomic position of the new isolate was investigated by a polyphasic approach. Phylogenetic analyses based on 16S rRNA gene sequences indicated that strain YIM T102(T) belongs to the genus Streptomyces. Strain YIM T102(T) was most closely related to Streptomyces eurocidicus NRRL B-1676(T) with a pairwise 16S rRNA gene sequence similarity of 98.9 %. However, DNA-DNA relatedness value between strain YIM T102(T) and S. eurocidicus NBRC 13491(T) was found to be 37.8 +/- 1.8 %. The menaquinone composition detected for strain YIM T102(T) was MK-9 (H-6) and MK-9 (H-8), while the major fatty acids were summed feature 4 (38.0 %), anteiso-C-15:0 (13.1 %), iso-C-16:0 (10.1 %), summed feature 3 (9.8 %) and C-16:0 (9.0 %) and iso-C-15:0 (5.2 %). The whole-cell hydrolysates contained galactose, glucose, ribose and mannose, along with ll-diaminopimelic acid as the diagnostic diamino acid in the peptidoglycan. The DNA G+C content was 70.7 mol%. Strain YIM T102(T) also exhibited antagonistic activity against Alternaria alternata, Alternaria brassicae and Colletotrichum nicotianae Averna, based on the findings from the comparative analyses of phenotypic and genotypic characteristics; it is proposed that strain YIM T102 represents a novel species of the genus Streptomyces, for which the name Streptomyces zhihengii sp. nov. is proposed. The type strain is YIM T102(T) (=KCTC 39115(T) = DSM 42176(T) = CGMCC 4.7248(T)).</P>

        Knockdown of UHRF1 ameliorates high glucose-induced podocyte injury by activating SIRT4

        Backgrounds Diabetic nephropathy (DN) is the leading cause of end-stage renal disease, and current treatment options to prevent its progression are inadequate. Podocyte injury is crucial to the pathogenesis of DN. Objective This study aimed to investigate the effect of UHRF1 on high glucose (HG)-induced podocyte injury and to explore its molecular mechanism. Results HG induced MPC5 cells to highly express UHRF1 while downregulating SIRT4. Knockdown of UHRF1 promoted HG-induced proliferation of MPC5 cells and ameliorated HG-induced cell cycle arrest, apoptosis, and ROS levels. Knockdown of UHRF1 ameliorated HG-induced podocyte injury by upregulating SIRT4. Conclusions Knockdown of UHRF1 ameliorates HG-induced podocyte injury by upregulating SIRT4, indicating that UHRF1 may be a novel target for preventing DN in podocytes.
