Colorectal cancer is the third most common cause of cancer-related deaths in the world. Surgical interventionfollowed by chemotherapy remains the primary approach to treatment since colon cancers remain refractory to most chemother-apeutic agents. Based on that, we established a program to screen natural products for cytotoxic activity, employing the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) reduction assay system utilizing HT-29 human coloncancer cells. During the course of our screening, we found that the methanolic extract of silkworm droppings (SDME) hascytotoxic effects on HT-29 cells. In the present study, we investigated the possible mechanisms by which SDME exerts itsantiproliferative activity in HT-29 cells. As expected, SDME inhibited growth of HT-29 cells in a dose-dependent manner asassessed by the MTT reduction assay, the lactate dehydrogenase release assay, and the colony formation assay. We also in-vestigated whether the apoptotic effects induced by SDME involve the caspase pathway using the caspase colorimetric assay.Interestingly, caspase-9 and -3, but not caspase-8, were activated in response to SDME treatment. Taken together, these re-sults clearly indicate that the induction of apoptosis by SDME involves a mitochondrial-mediated pathway and strongly sug-gest that SDME may potentially be a chemotherapeutic agent for human colon cancer.

Immunomodulatory Effect of a New Herbal Preparation (HemoHIM) in Cyclophosphamide-treated Mice

The immunomodulatory effect of a new herbal preparation, HemoHIM, on the recovery from leukopenia induced by cyclophosphamide treatment was investigated. The HemoHIM was made up with an addition of the ethanol-insoluble fraction to the total water extract of Angelica Radix, Cnidii Rhizoma and Paeonia Radix. Daily oral administration of 100 mg/kg BW or 500 mg/kg BW HemoHIM accelerated the recovery from cyclophosphamide-induced leukopenia. HemoHIM increased the number of leukocytes and lymphocytes in the peripheral blood when compared with the cyclophosphamide-treated control. Moreover, the suppressed natural killer (NK) cell activity and interferon $(IFN)-{\gamma}$ secretion in the cyclophosphamide-treated mice were restored by the administration of HemoHIM. HemoHIM significantly reduced the abnormally heightened ratio of interleukin $(IL)-4/IFN-{\gamma}$ and immunoglobulin (Ig)E/IgG2a in the cyclophosphamide-treated mice. These results suggest that HemoHIM accelerates the recovery from leukopenia and alleviates the imbalanced T helper (Th)l/Th2 responses in the cyclophosphamide-treated mice. Additionally, HemoHIM was found to stimulate normal splenocytes to secrete not only Thl type cytokines such as $IFN-{\gamma}$ and IL-2, but also Th2 type cytokine IL-4. In conclusion, our results show that HemoHIM certainly has an influence on the balanced recovery of immune cells and the activation of their activities in the cyclophosphamide-treated mice.

Objective: Pathology PACS is system that store all digital Video Information that search, read, output ranging system that manages all the functions are integrated. Background: Because period that domestic pathology PACS is introduced up to now is short, professional study for user is lacking. Method: To analyze whether user achieves some work by some way, University Hospital, Pathology doctors use the screen to the actual hospital pathology PACS was recorded. Except for the error file, definition video files into text and categorized the text you typed. Results: In this study, using a hierarchical task analysis user-friendly interface improvement of Pathology PACS is trying to draw. Conclusion: With this research, in pathology PACS to improve on the user's convenience and a more intuitive user interface is expected to provide.
Issues in Facilitating Nursing Effectiveness Research and Developing Clinical Databases in Korea

간호의 결과를 가시화하고 그 효율성을 입증하는 방법은 간호 효율성 연구를 통해서 이루어져야 하며 간호 효율성 연구의 촉진을 위해서는 대단위 임상데이터베이스의 개발과 확립이 필수적인 요소이다. 본 연구에서는 간호효율성 연구의 범위와 필요한 연구설계를 확인하고 이러한 간호 효율성 연구를 위해 필요한 데이터베이스의 종류와 필수적인 간호자료를 위한 최신 연구결과를 고찰함으로써 국내에서의 간호효율성 연구와 대단위 임상데이터베이스 구축을 위한 기초자료를 제공하고자 한다. 본 연구에서 고찰된 내용은 다음과 같다. 1) 효율성 연구와 간호효율성 연구의 개념, 2) 간호효율성 연구에 유용한 연구설계와 종류, 3) 간호 효율성 연구를 위한 데이터 및 데이터베이스, 4) 간호효율성 연구를 위한 대단위 임상 데이터베이스의 종류와 기능. 본 연구의 고찰을 바탕으로 다음과 같은 제언을 하고자 한다. 첫째, 개별 간호사 수준에서 정확한 사정과 결과평가를 통해 신뢰할 수 있는 데이터를 산출하여야 하며 또한 간호효율성 연구에 직접 관여하여 연구결과의 산출이 촉진되어야 한다. 둘째, 임상자료가 축적되어 있는 데이터베이스에 대한 정보가 제공되어 개별 연구자와 실무자들의 데이터베이스에 대한 접근성을 증진시켜야 한다. 셋째, 기존 데이터베이스의 신뢰성에 대한 지속적인 평가와 필수적인 자료와 비필수적인 자료에 대한 명확한 구분이 필요하다.

<P><I>Purpose</I>: We already reported that levels of interferon (IFN)-γ have been shown to be markedly reduced in mice seven weeks after irradiation, resulting in a T helper (Th) 1/Th2 imbalance. To investigate whether the single or fractionated γ-irradiation induced an immune imbalance, we analysed the Th1-related immune response profile until six months after the fractionated whole-body irradiation.</P><P><I>Methods and materials</I>: Mice were exposed to γ-rays at a fractionated 5 Gy cumulative dose for five weeks. At two, four and six months later from the first exposure, experiments were performed. Cell populations in the spleen, the production of IFN-γ, interleukin (IL)- 4 and IL-12p70, natural killer (NK) cell activity and the expression of IL-12 receptors, signal transducer and activator of transcription (STAT) 4 and suppressors of cytokine signaling (SOCS) 3 were detected.</P><P><I>Results</I>: The IFN-γ was lower in the mice exposed by all irradiation conditions than in normal control mice, but the IL-4 had increased in all the irradiated mice. To investigate Th1 profile, NK cell activity, IL-12p70 level and its receptor expression was confirmed. In all fractionated irradiation groups, the NK cell activity as well as the absolute numbers of NK cells was much decreased. Also, all the irradiated mice showed a lower IL-12p70 level. However, the expression of IL-12 receptor β2 was lower in the irradiated mice except the 0.2 Gy ????× ????10 mice group. The phosphoylated STAT4 was lower in all the irradiated mice. This suppression was associated with an overexpression of SOCS3.</P><P><I>Conclusions</I>: The fractionated whole-body irradiations of a dose of 5 Gy appear to be the down-regulation of the Th1-like immune response. These changes, in turn, maintain an immunological imbalance that persists in the long term.</P>
