        Effects of Acupuncture at Zu-San-Li (ST36) on the Activity of the HypothalamicePituitaryeAdrenal Axis during Ethanol Withdrawal in Rats

        The current study investigated the effects of acupuncture at Zu-San-Li (ST36) on the hypothalamicepituitaryeadrenalaxis during ethanol withdrawal in rats. Rats were intraperitoneallytreated with 3 g/kg/day of ethanol or saline for 28 days. Following 24 hours ofethanol withdrawal, acupuncture was applied at bilateral ST36 points or non-acupoints(tail) for 1 minute. Plasma levels of corticosterone (CORT) and adrenocorticotropic hormone(ACTH) were measured by radioimmunoassay (RIA), and the corticotropin-releasingfactor (CRF) protein levels in the paraventricular nucleus of the hypothalamus were alsoexamined by RIA 20 minutes after the acupuncture treatment. RIA showed significantlyincreased plasma levels of CORT and ACTH in the ethanol-withdrawn rats compared withthe saline-treated rats, which were inhibited significantly by the acupuncture at the acupointST36 but not at the non-acupoint. Additionally, ethanol withdrawal promoted CRFprotein expressions in the paraventricular nucleus of the hypothalamus, which were alsoblocked by the acupuncture at ST36. These findings suggest that acupuncture at the specific acupoint ST36 can inhibit ethanol withdrawal-induced hyperactivation of hypothalamicepituitaryeadrenalaxis, and it may be mediated via the modulation of hypothalamicCRF.

        Theoretical Studies on Nitramine Explosives with -NH<sub>2</sub> and -F Groups

        The nitramine explosives with $-NH_2$ and -F groups were optimized to obtain their molecular geometries and electronic structures at DFT-B3LYP/6-31+G(d) level. The theoretical molecular density (${\rho}$), heat of formation (HOF), detonation velocity ($D$) and detonation pressure ($P$), estimated using Kamlet-Jacobs equations, showed that the detonation properties of these compounds were excellent. Based on the frequencies scaled by 0.96 and the principle of statistic thermodynamics, the thermodynamic properties were evaluated, which were respectively related with the temperature. The simulation results reveal that 1,3,5,7-tetranitro-1,3,5,7-tetrazocan-2-amine (molecule B1) performs similarly to the famous explosive HMX, and 2-fluoro-1,3,5-trinitro-1,3,5-triazinane (molecule C1) and 2-fluoro-1,3,5,7-tetranitro-1,3,5,7-tetrazocane (molecule D1) outperform HMX. According to the quantitative standard of energetics and stability as an HEDC (high energy density compound), molecules C1 and D1 essentially satisfy this requirement. These results provide basic information for molecular design of novel high energetic density compounds.

        Purification and Characterization of a β-Glucosidase from Aspergillus niger and Its Application in the Hydrolysis of Geniposide to Genipin

        An extracellular β-glucosidase from Aspergillus niger Au0847 was purified to homogeneity by precipitation with ammonium sulfate, anion exchange, and gel filtration. The purified protein was composed of two subunits with molecular masses of 110 and 120 kDa. Au0847 β-glucosidase exhibited relatively high thermostability and pH stability, and its highest activity was obtained at 65°C and pH 4.6, respectively. As a potential metalloprotein, its enzymatic activity was potently stimulated by manganese ion and DTT. The β-glucosidase displayed avid affinity and high catalytic efficiency for geniposide. Au0847 β-glucosidase has potential value as an industrial enzyme for the hydrolysis of geniposide to genipin.

      • Effects of Parafibromin Expression on the Phenotypes and Relevant Mechanisms in the DLD-1 Colon Carcinoma Cell Line

        Background: Parafibromin is a protein encoded by the HRPT2 (hyperparathyroidism 2) oncosuppressor gene and its down-regulated expression is involved in pathogenesis of parathyroid, breast, gastric and colorectal carcinomas. This study aimed to clarify the effects of parafibromin expression on the phenotypes and relevant mechanisms of DLD-1 colon carcinoma cells. Methods: DLD-1 cells transfected with a parafibromin-expressing plasmid were subjected to examination of phenotype, including proliferation, differentiation, apoptosis, migration and invasion. Phenotype-related proteins were measured by Western blot. Parafibromin and ki-67 expression was detected by immunohistochemistry on tissue microarrays. Results: The transfectants showed higher proliferation by CCK-8, better differentiation by electron microscopy and ALP activity and more apoptotic resistance to cisplatin by DNA fragmentation than controls. There was no difference in early apoptosis by annexin V, capase-3 activity, migration and invasion between DLD-1 cells and their transfectants. Ectopic parafibromin expression resulted in down-regulated expression of smad4, MEKK, GRP94, GRP78, $GSK3{\beta}$-ser9, and Caspase-9. However, no difference was detectable in caspase-12 and -8 expression. A positive relationship was noted between parafibromin and ki-67 expression in colorectal carcinoma. Conclusions: Parafibromin overexpression could promote cell proliferation, apoptotic resistance, and differentiation of DLD-1 cells.

      • Prognostic Value of C-Reactive Protein in Esophageal Cancer: a Meta-analysis

        Background: The classical inflammatory biomarker, C-reactive protein (CRP), has been identified to be related to progression of esophageal cancer. Some research showed that elevated pretreatment serum CRP indicated a poor prognosis, but results have been inconsistent. Materials and Methods: We searched the Medline, Embase and the Cochrane Central Search Library for suitable studies and a meta-analysis of eleven (1,886 patients) was conducted to examine the relationship between elevated serum CRP level and overall survival (OS) in esophageal cancer cases. Moreover, correlation analyses were conducted to assess links between pretreatment serum CRP level and tumor node metastasis (TNM) stage as well as T, N, M grade, respectively. Results: The pooled analysis showed that elevated pretreatment serum CRP level was significantly associated with poorer overall survival (HR 2.09, 95%CI 1.52-2.87, p<0.01). Subgroup analyses were conducted by "country", "cut-off value", "treatment" and "number of patients", and no single factor could alter the result. Elevated pretreatment serum CRP was significantly correlated with more advanced TNM stage and T, N, M grade respectively. Conclusions: Elevated pretreatment serum CRP levels are associated with poorer prognosis in esophageal cancer patients, and could serve as a useful biomarker for outcome prediction.

        Microstructure control of Fe catalyst films for the growth of multiwalled carbon nanotube arrays

        Fe catalyst films were deposited on silicon substrates by using a metal vapor vacuum arc (MEVVA) ion deposition system for the growth of aligned multiwalled carbon nanotubes. The effects of deposition angle and film thickness on the morphologies of catalyst films before and after thermal treatment were investigated. The results show that the uniformities of both 5-nm and 10-nm films are enhanced as the deposition angle decreases. With increase from 5 nm to 10 nm, the surface uniformities of pristine films prepared at higher deposition angles (60˚ and 90˚) are increased, while that of the films produced at 30˚ deposition angle is slightly decreased. The uniformity of Fe catalyst particle sizes after thermal treatment on the whole, clearly decreases as the thickness is increased from 5 nm to 10 nm in the case of the same deposition angle. 5-nm films deposited at 30˚ deposition angle show the most uniform features before and after thermal treatment and can be used for the synthesis of high-quality carbon nanotube arrays.         A Prediction Model Based on the Risk Factors Associated with Pathological Upgrading in Patients with Early-Stage Gastric Neoplasms Diagnosed by Endoscopic Forceps Biopsy

        Background/Aims: The discrepancies between the diagnosis of preoperative endoscopic forceps biopsy (EFB) and endoscopic submucosal dissection (ESD) in patients with early gastric neoplasm (EGN) exist objectively. Among them, pathological upgrading directly influences the accuracy and appropriateness of clinical decisions. The aims of this study were to investigate the risk factors for the discrepancies, with a particular focus on pathological upgrading and to establish a prediction model for estimating the risk of pathological upgrading after EFB. Methods: We retrospectively collected the records of 978 patients who underwent ESD from December 1, 2017 to July 31, 2021 and who had a final histopathology determination of EGN. A nomogram to predict the risk of pathological upgrading was constructed after analyzing subgroup differences among the 901 lesions enrolled. Results: The ratio of pathological upgrading was 510 of 953 (53.5%). Clinical, laboratorial and endoscopic characteristics were analyzed using univariable and binary multivariable logistic regression analyses. A nomogram was constructed by including age, history of chronic atrophic gastritis, symptoms of digestive system, blood high density lipoprotein concentration, macroscopic type, pathological diagnosis of EFB, uneven surface, remarkable redness, and lesion size. The C-statistics were 0.804 (95% confidence interval, 0.774 to 0.834) and 0.748 (95% confidence interval, 0.664 to 0.832) in the training and validation set, respectively. We also built an online webserver based on the proposed nomogram for convenient clinical use. Conclusions: The clinical value of identifying the preoperative diagnosis of EGN lesions is limited when using EFB separately. We have developed a nomogram that can predict the probability of pathological upgrading with good calibration and discrimination value.

        Traveling Wave Type Multi‑Degree‑of‑Freedom Spherical Ultrasonic Motor with Built‑in Stators

        A multi-degree-of-freedom spherical ultrasonic motor with built-in traveling wave stators is proposed, and each traveling wave stator can be controlled independently, the three-degree-of-freedom movement of the spherical rotor is realized by the coordinated control of the three traveling wave stators and the spatial arrangement of the support structures. The motor also has a built-in pre-pressure adjustment structure, which can adjust the pre-pressure of three stators at the same time and make them the same size. The fnite element method is used to analyze the modal of the traveling wave stator, and the transient trajectory of the driving part is simulated. Finally, the working principle was verifed on the prototype, and the mechanical output performance of the prototype was tested: when the excitation voltage of the motor is 400 V and the preload is 100 N, the maximum speed is 45.6 rad/min, the maximum speed output torque is 1.265 Nm; the maximum defection angle is 43°, and the maximum defection speed is 56.3 rad/min, the maximum defection torque is 1.76 Nm, achieves high torque and multiple-degrees-of-freedom. The motor has the advantages of large output force, high integration, and adjustable preload.

      • Associations Between Infiltrating Lymphocyte Subsets and Hepatocellular Carcinoma

        Aims: We aimed to analyze the phenotype of tumor-infiltrating lymphocytes (TILs) and non-tumor infiltrating lymphocytes (NILs) in HCC and non-tumor tissues, and evaluate relationships between changes in these cells and the prognosis of HCC. Methods: Lymphocytes were isolated from HCC and corresponding non-tumor tissues and tested by flow cytometry. For comparison, clinical parameters were analyzed. Results: Compared with the non-tumor tissue, tumor tissue had a lower intensity of NK, NKT andCD8+T cell infiltration. TILs had higher intensity of CD4+CD25+Foxp3+regulatory T cell (Treg cells) infiltration compared with that in NILs. The prevalence of Treg cells was associated with fewer CD8 + T lymphocytes in the HCC immune microenvironment. The frequencies of NK cells and CD8+T cells in TILs of HCC patients with metastasis less than 12 months were lower than those without metastasis. However, the frequency of Treg cells was higher than those without metastasis. Conclusion: These results suggest that the frequencies of CD8+T, NK and NKT cells as well as Treg cells in the tumor tissue of HCC are significantly associated with patient survival, and could be applied as predictive indicators for HCC prognosis.
