Development of AFLP and SCAR Markers Linked to a Recessive Genic Male Sterile Gene (Bnms3) in Rapeseed for Marker-assisted Selection

        Gui Chun Wang,Jun Ping He,Deng Feng Hong,Yan Zhou Xie,Zheng Hua Xu,Ping Wu Liu,Guang Sheng Yang 한국유전학회 2007 Genes & Genomics Vol.29 No.4

9012AB is a recessive genic male sterility (RGMS) line in rapeseed, of which the male sterility is controlled by two pairs of recessive duplicate male sterile genes (Bnms3 and Bnms4) interacting with a recessive epistatic suppressor gene (esp). The recessive homozygosity at the esp locus (espesp) can suppress the expression of the recessive male sterility trait in homozygous plants (Bnms3Bnms3Bnms4Bnms4) and result in fertility restoration. A F2 population of 188 plants, derived from self-pollinated progenies of a 9012 AB fertile plants (BnMs3Bnms3Bnms4Bnms4EspEsp), was conducted to identify molecular markers linked to the recessive male sterility gene (Bnms3). By amplified fragment length polymorphism (AFLP) assay combining with bulked segregant analysis (BSA), 13 markers linked to Bnms3 were identified. Linkage analysis indicated that 13 AFLP markers were tightly linked to the Bnms3 gene with a genetic distance varying from 1.3 cM to 7.1 cM. Among them, one marker was co-dominant marker, 6 markers were in coupling phase with Bnms3, and the others were in repulsion phase with Bnms3 gene, One AFLP marker with a genetic distance of 1.4cM was further converted into a SCAR marker successfully, which have been applied in marker-assisted selection of RGMS lines and their temporary maintainers effectively.

Adaptive State-of-Charge Estimation Method for an Aeronautical Lithium-ion Battery Pack Based on a Reduced Particle-unscented Kalman Filter

        Wang, Shun-Li,Yu, Chun-Mei,Fernandez, Carlos,Chen, Ming-Jie,Li, Gui-Lin,Liu, Xiao-Han The Korean Institute of Power Electronics 2018 JOURNAL OF POWER ELECTRONICS Vol.18 No.4

A reduced particle-unscented Kalman filter estimation method, along with a splice-equivalent circuit model, is proposed for the state-of-charge estimation of an aeronautical lithium-ion battery pack. The linearization treatment is not required in this method and only a few sigma data points are used, which reduce the computational requirement of state-of-charge estimation. This method also improves the estimation covariance properties by introducing the equilibrium parameter state of balance for the aeronautical lithium-ion battery pack. In addition, the estimation performance is validated by the experimental results. The proposed state-of-charge estimation method exhibits a root-mean-square error value of 1.42% and a mean error value of 4.96%. This method is insensitive to the parameter variation of the splice-equivalent circuit model, and thus, it plays an important role in the popularization and application of the aeronautical lithium-ion battery pack.

Effect of Pomegranate Peel Polyphenols on Human Prostate Cancer PC-3 cells in vivo

        Gui-Zhi Ma,Chun-Mei Wang,Li Li,Xiao-Li Gao 한국식품과학회 2015 Food Science and Biotechnology Vol.24 No.5

Punica granatum possesses strong antitumor properties. In this study, a nude mouse model with a subcutaneous xenograft of human prostate cancer cells (PC-3) was established to observe the in vivo antiproliferative and apoptotic effects of pomegranate peel polyphenols (PP). Levels of the cytokines tumor necrosis factor α (TNF-α) and vascular endothelial growth factor (VEGF) were detected by enzyme-linked immunosorbent assay. Quantitative analysis of ellagic acid, gallic acid, and punicalagin in active fraction was conducted using reverse phase high-performance liquid chromatography with a photo-diode array (HPLC/PDA). PP decreased tumor volume and weight in tumor-bearing nude mice, and significantly increased the rate of apoptosis (p<0.05). In addition, PP increased TNF-α and decreased VEGF in the serum (p<0.05). Ellagic acid (201.3±3.544 mg/g), gallic acid (8.917±0.274 mg/g), and punicalagin (407.0±12.05 mg/g) were the main effectors of the anti-tumor activity.

An investigation of the ferroelectric and magnetic properties of Nd-doped Bi1 − xNdxFeO3 polycrystalline ceramics fabricated by a sol-gel technique

        Chang Chun Chen,Zhonghai Tang,Gui Wang,Chunhua Lu,Zhongzi Xu 한양대학교 세라믹연구소 2012 Journal of Ceramic Processing Research Vol.13 No.2

Nd-modified Bi1 − xNdxFeO3 (x = 0, 0.05, 0.1, and 0.15) ceramics were fabricated by a sol-gel method. The crystal structures of Nd-modified Bi1 − xNdxFeO3 (x = 0, 0.05, 0.1, and 0.15) ceramics were analyzed by X-ray diffraction (XRD). The effects of the Nd content on the ferroelectric, dielectric, and magnetic properties of these Bi1 − xNdxFeO3 ceramics were thoroughly investigated. The XRD patterns indicated that with an increase of the Nd doped content in Bi1 − xNdxFeO3 ceramics, the unwanted impurity phases such as Bi2Fe4O9 and Bi36Fe24O57 are markedly decreased. Clear polarization-electric field (P-E)hysteresis loops were observed for all the Nd-modified Bi1 − xNdxFeO3 ceramics and the shape of the P-E loop of the Bi1 − xNdxFeO3 ceramics was significantly improved due to the Nd substitution. In addition, slim magnetic hysteresis (M-H)loops in the Bi1 − xNdxFeO3 ceramics were also observed, and the saturated magnetization (Ms) of the Bi1 − xNdxFeO3 ceramics also increased with an increase of the Nd content.

The Influence of Construction Scheme of Asymmetric Three-Cabin Utility Tunnelling on the Surface Settlement Behaviour

        Zuo Chun Li,Gui He Wang,Jun Wei Hao,Yao Zhou,Xiao Yang Wang,Heng Xuan,Feng Huang 대한토목학회 2021 KSCE JOURNAL OF CIVIL ENGINEERING Vol.25 No.9

The asymmetric three-cabin structure is one of the favourite choices in urban utility tunnel engineering, as it can provide a higher space utilization and clearer classification for pipelines. However, few work has been done on utility tunnel in such special cross-section during undercutting construction until now. Besides, as the loose soil layers such as sand and silt are often encountered during the utility tunnelling, its influence on surface settlement become one of the main concerns. This study focuses on the influence of the construction scheme on the surface settlement details for an asymmetric three-cabin utility tunnel. A 3D finite element method (FEM) model was established including both the stratum and a utility tunnel based on the Beijing Daxing International Airport (BDIA) Expressway Urban Utility Tunnel project. With this model, the behaviour of the surface settlement with the undercutting construction of the utility tunnel was investigated, and the causes and values of surface settlement in five construction stages were analyzed. By changing the construction spacing and sequence of mid and side cabins, the surface settlement and structural deformation were compared. The numerical results demonstrated a strong correlation between the cross-section shape and surface settlement characteristics. And the symmetry line of the surface settlement trough was offset by 1 m to the side with the larger cross-section. The second (excavation of upper bench in mid-cabin) and fourth (excavation of upper bench in side-cabin) stages caused the largest surface settlement, which were 41.9% and 18.07% of the total settlement, respectively. In addition, after optimizing the field tunnelling scheme, the surface settlement was reduced by 31.9% using the side-tunnel first construction sequence. The proposed numerical model is able to predict the settlement characteristics in each construction stage, which is important and provides a basis for further studies on the surface settlement mechanism and optimal design of the asymmetric three-cabin utility tunnelling scheme.

Generation of Selenoprotein with Glutathione Peroxidase Activity by Chemical Modification of the Single-chain Variable Fragment Expressed in a Single-protein Production System and Its Antioxidant Ability

        Cheng Wang,Gang-Lin Yan,Shao-Wu Lü,Chun-Hong Sui,Yang Zhao,Ya-Wei Xu,Gang Zhao,Jun-jie Xu,Ping-Sheng Gong,Gui-Min Luo,Ying Mu 한국생물공학회 2013 Biotechnology and Bioprocess Engineering Vol.18 No.1

Glutathione peroxidase (GPX) is one of the important members of the antioxidant enzyme family. It can catalyze the reduction of hydroperoxides with glutathione to protect cells against oxidative damage. Single-chain variable fragment (scFv) can be converted into seleniumcontaining single-chain variable fragment (Se-scFv) by chemical modification of the hydroxyl groups in scFv, thus Se-scFv possesses GPX activity and becomes a prodrug. To improve the expression of scFv and simplify its purification steps, Single-protein production (SPP) system was used to express scFv and chemical modification was used to synthesize Se-scFv. Therefore, we must construct a new scFv-WCD1-lessACA gene, which can express its mRNA not containing any ACA sequences and express its amino acid sequence of target protein (scFv) being same to scFv-WCD1. In this way, the scFv-WCD1-lessACA can be only expressed in SPP system and no other background proteins in the cells could be expressed. The expression results showed that high level of scFv-WCD1-lessACA synthesis was at least sustained for 96 h in the virtual absence of background protein synthesis. Then, selenocysteine (Sec) was incorporated into the scFv-WCD1-lessACA by chemical modification and resulted in Se-scFv-WCD1-lessACA. The enzymatic characteristics of Se-scFv-WCD1-lessACA were determined. GPX activity was 2,563 U/μmol,its binding constant for GSH was 0.687 ×105/mol. Moreover,Se-scFv-WCD1-lessACA was confirmed to have a strong antioxidant ability to protect mitochondria against oxidative damage induced by Vc/Fe2+ (mitochondrial damage model),suggesting that Se-scFv-WCD1-lessACA has potential application for protection of mitochondrial damage induced by reactive oxygen species (ROS).

        Rui Tian,Chun-Hua Yang,Xiao-Fei Wei,Er-Na Xun,Ren Wang,Shu-Gui Cao,Zhi Wang,Lei Wang 한국생물공학회 2011 Biotechnology and Bioprocess Engineering Vol.16 No.2

Aeropyrum pernix esterase (APE1547) was successfully used to catalyze the enantioselective transesterification of (R/S)-2-methyl-1-butanol in an ionic liquid (IL). Effects of various reaction conditions on the synthetic activity of the enzyme as well as enantioselectivity, including the type of IL, acyl donor, temperature, water activity, and substrate molar ratio were inverstigated. APE1547showed good catalytic performance (activity > 0.8 μmol/min/mg, E > 25), and the enzyme-IL mixture was recycled five times with only a slight decrease in catalytic performance.

        Hong-Sheng Wang,Ling Sun,Yue-Hu Wang,Ya-Na Shi,Gui-Hua Tang,Fu-Wei Zhao,Hong-Mei Niu,Chun-Lin Long,Ling Li 대한약학회 2011 Archives of Pharmacal Research Vol.34 No.8

A new dihydroflavone, 5-carboxymethyl-7,4'-dihydroxyflavonone (1), and its glucoside 5-carboxymethyl-7,4'-dihydroxyflavonone-7-O-β-D-glucopyranoside (2), and one new monoterpene glucoside, (4Z,6E)-2,7-dimethyl-8-hydroxyocta-4,6-dienoic acid 8-O-β-D-glucopyranoside (3), were isolated from the whole plants of Selaginella moellendorffii. Their structures were determined by spectroscopic methods and chemical transformation. Compound 2 was evaluated for the ability to enhance glucose consumption in normal and insulin-resistant L6 muscle cells induced by high concentrations of insulin and glucose. Glucose consumption in insulin-resistant cells (but not in normal cells) was increased 15.2 ± 3.3% (p < 0.01) by compound 2 at a concentration of 0.1 μM in the presence of insulin (1 nM).

      • Genetic Variations in the HIF1A Gene Modulate Response to Adjuvant Chemotherapy after Surgery in Patients with Colorectal Cancer

        Zhang, Yi,Wang, Peng,Zhou, Xing-Chun,Bao, Guo-Qiang,Lyu, Zhuo-Ming,Liu, Xiao-Nan,Wan, Shao-Gui,He, Xian-Li,Huang, Qi-Chao Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.11

Background: Hypoxia-inducible factor $1{\alpha}$ (HIF-$1{\alpha}$) plays an important role in regulating cell survival and angiogenesis, which are critical for tumor growth and metastasis. Genetic variations of HIF1A have been shown to influence the susceptibility to many kinds of human tumors. Increased expression of HIF-$1{\alpha}$ has also been demonstrated to be involved in tumor progression. However, the prognostic value of single nucleotide polymorphisms (SNPs) inthe HIF1A gene remains to be determined in most cancer types, including colorectal cancer (CRC). In this study, we sought to investigate the predictive role of HIF1A SNPs in prognosis of CRC patients and efficacy of chemotherapy. Materials and Methods: We genotyped two functional SNPs in HIF1A gene using the Sequenom iPLEX genotyping system and then assessed their associations with clinicopathological parameters and clinical outcomes of 697 CRC patients receiving radical surgery using Cox logistic regression model and Kaplan Meier curves. Results: Generally, no significant association was found between these 2 SNPs and clinical outcomes of CRC. In stratified analysis of subgroup without adjuvant chemotherapy, patients carrying CT/TT genotypes of rs2057482 exhibited a borderline significant association with better overall survival when compared with those carrying CC genotype [Hazard ratio (HR), 0.47; 95% confidence interval (95% CI): 0.29-0.76; P < 0.01]. Moreover, significant protective effects on CRC outcomes conferred by adjuvant chemotherapy were exclusively observed in patients carrying CC genotype of rs2057482 and in those carrying AC/CC genotype of rs2301113. Conclusions: Genetic variations in HIF1A gene may modulate the efficacy of adjuvant chemotherapy after surgery in CRC patients.
