        New Evidence of Alleles (V199I and G52S) at the PRKAG3 (RN) Locus Affecting Pork Meat Quality

        The porcine PRKAG3 (RN) gene encodes the regulatory gamma subunit of adenosine monophosphate-activated protein kinase (AMPK), which is a good candidate gene affecting meat quality. In this study, the effects of two missense mutations A595G (Ile199Val) and G154A (Gly52Ser) in porcine PRKAG3 gene on meat quality traits were studied in M. Longissimus dorsi (LD), M. Semispinalis capitis (SC) and M. Biceps femoris (BF) from different populations of 326 pigs. The PRKAG3 alleles 199I, 199IV, 52S and 52G were identified with PCR-RFLPs and all genotypes - 199I/199I, 199I/199V, 199V/199V, 52S/52S, 52S/52G and 52G/52G - were found. The frequency of V allele was larger than that of I allele in all populations. I allele frequency was zero in Chinese Meishan pigs (population D) especially. G allele frequency was larger than that of S allele in all populations except Large White (population A). Both variations at the PRKAG3 locus significantly affected these meat quality traits. The pork meat quality has not previously been established in Meishan or crosses thereof. The results suggested that generally pH of LD, SC and BF was higher in Meishan pigs than that in other populations. Moreover, Meishan pigs showed higher water-holding capacity and intramuscular fat (IMF), lower water content and water loss percentage compared to other populations in terms of the two variations. The results present here supply new evidence that alleles V199I and G52S at the PRKAG3 locus affect pork meat quality and provide useful information on pork production.

        Feasibility study of dual-targeting paclitaxel-loaded magnetic liposomes using electromagnetic actuation and macrophages

        This work aims to develop a novel electromagnetic and macrophage-mediated drug delivery system to transport anticancer drug (Paclitaxel, PTX)-encapsulated magnetic liposomes (PTX-MLPs) to tumors. The PTX-MLPs are engulfed into macrophages by phagocytosis of murine macrophages (J774A1). Therefore, the macrophages with PTX-MLPs can be dually targeted both by an external electromagnetic field and by chemotaxis of the macrophages to the tumors. Under the electromagnetic field, a single macrophage with PTX-MLPs can be controlled following predefined paths and can reach an average velocity of 10.48+/-4.43μm/s. Meanwhile, using chemotaxis, the macrophages with PTX-MLPs can infiltrate through a 5μm pore membrane to chemo-attractants in transwell migration assays, with infiltration rate of up to 73.70+/-4.63%. In addition, their therapeutic efficacy is investigated using an in-vitro cytotoxicity assay on breast cancer (4T1) and colorectal cancer (CT26) models. The results confirm that the macrophages with PTX-MLPs can carry a sufficient amount of drug to kill the cancer cells with the IC<SUB>50</SUB> values for the 4T1 and CT26 cells of 52.97+/-1.06μg/mL and 117.04+/-1.82μg/mL, respectively. The outcomes of this study suggest that a potential active cancer therapy can be developed through the dual targeting system using macrophages with PTX-MLPs and magnetic actuation.

        Evaluation of Ni Film Interfacial Energy Release Rate on Titanium and Stainless Steel Substrates

        An expression including the effect of residual stress on the interfacial energy release rate is proposed for peeling experiments according to the energy-balance argument. The influence of residual stress on the external work is also contained in the expression. Two numerical methods are employed to evaluate the values of the work expenditure G_db, which is the actual energy dissipated during bending of the peel arm near the peel front. The peeling method is employed to test the interfacial energy release rates, G, for Ni films on Titanium and stainless steel substrates. The results indicate that the value of G for Ni films on stainless steel substrate is about 5.47-6.03 N/m, while 5.23-6.71 N/m for Ni films on titanium substrate; the interfacial energy release rates, G, do not depend on the residual stress in film, film thickness nor peel angle. The effect of residual stress in film on peel strength P/h is also discussed.

      • A combined stochastic diffusion and mean-field model for grain growth

        A combined stochastic diffusion and mean-field model is developed for a systematic study of the grain growth in a pure single-phase polycrystalline material. A corresponding Fokker-Planck continuity equation is formulated, and the interplay/competition of stochastic and curvature-driven mechanisms is investigated. Finite difference results show that the stochastic diffusion coefficient has a strong effect on the growth of small grains in the early stage in both two-dimensional columnar and three-dimensional grain systems, and the corresponding growth exponents are ~0.33 and ~0.25, respectively. With the increase in grain size, the deterministic curvature-driven mechanism becomes dominant and the growth exponent is close to 0.5. The transition ranges between these two mechanisms are about 2-26 and 2-15 nm with boundary energy of 0.01-1 J $m^{-2}$ in two- and three-dimensional systems, respectively. The grain size distribution of a three-dimensional system changes dramatically with increasing time, while it changes a little in a two-dimensional system. The grain size distribution from the combined model is consistent with experimental data available.

        A numerical study on mixed convection in a lid-driven cavity with a circular cylinder

        A two-dimensional numerical simulation is carried out in this study to investigate mixed convection in a lid-driven cavity with an isothermal circular cylinder. The simulation is conducted at three Reynolds numbers of Re = 100, 500, and 1000 under a fixed Grashof number of Gr = 105. The top wall of the cavity moves to the right at a constant velocity and is kept at a low temperature of Tc, whereas the stationary bottom wall is kept at a constant high temperature of Th. The immersed-boundary method, which is based on the finite volume method, is adopted for the boundary of the circular cylinder that is present in the square cavity. The present study aims to investigate the effects of circular cylinder on fluid flow and heat transfer in a cavity at different locations. The fluid flow and heat transfer characteristics in the cavity strongly depend on the position of the circular cylinder as well as on the relative magnitude of the forced convection and the natural convection caused by the movement in the top wall of the cavity and the heating at the hot bottom wall, respectively.

        MADS-box genes (Schwarz-Sommer et al. 1990) contribute significantly to the development of inflorescences and flowers, e.g. by acting as floral meristem or organ identity genes (for recent reviews, see Thei?en and Saedler 1995; Thei?en et al. 1996;Riechmann and Meyerowitz 1997). Up to now, MADS-box genes have mainly been studied in the scientific context of developmental biology, and in eudicotyledonous model plants of little commercial value, such as Antirrhinum and Arabidopsis. The focus of our group is to apply our current knowledge about flower development to breeding research and evolutionary biology. Therefore, we are currently characterizing the MADS-box gene family in the important monocotyledonous crop plant maize (Zea mays ssp. mays), and in other phylogenetic informative taxa, such as the monocots lily (Lilium regale) and tulip (Tulipa gesneriana), the basal angiosperm Cabomba, and non-flowering plnats including the gymnosperm Gnetum gnemon and the ferns Ceratopteris and Ophioglossum.

      • Comparison of the osteogenesis and fusion rates between activin A/BMP-2 chimera (AB204) and rhBMP-2 in a beagle's posterolateral lumbar spine model

        Background Context: Activin A/BMP-2 chimera (AB204) could promote bone healing more effectively than recombinant bone morphogenetic protein 2 (rhBMP-2) with much lower dose in a rodent model, but there is no report about the effectiveness of AB204 in a large animal model. Purpose: The purpose of this study was to compare the osteogenesis and fusion rate between AB204 and rhBMP-2 using biphasic calcium phosphate (BCP) as a carrier in a beagle's posterolateral lumbar fusion model. Study Design: This is a randomized control animal study. Methods: Seventeen male beagle dogs were included. Bilateral posterolateral fusion was performed at the L1-L2 and L4-L5 levels. Biphasic calcium phosphate (2@?cc), rhBMP-2 (50@?@?g)+BCP (2@?cc), or AB204 (50@?@?g)+BCP (2@?cc) were implanted into the intertransverse space randomly. X-ray was performed at 4 and 8 weeks. After 8 weeks, the animals were sacrificed, and new bone formation and fusion rate were evaluated by manual palpation, computed tomography (CT), and undecalcified histology. Results: The AB204 group showed significantly higher fusion rate (90%) than the rhBMP-2 group (15%) or the Osteon group (6.3%) by manual palpation. On x-ray and CT assessment, fusion rate and the volume of newly formed bone were also significantly higher in AB204 group than other groups. In contrast, more osteolysis was found in rhBMP-2 group (40%) than in AB204 group (10%) on CT study. In histologic results, new bone formation was sufficient between transverse processes in AB204 group, and obvious trabeculation and bone remodeling were observed. But in rhBMP-2 group, new bone formation was less than AB204 group and osteolysis was observed between the intertransverse spaces. Conclusions: A low dose of AB204 with BCP as a carrier significantly promotes the fusion rate in a large animal model when compared with the rhBMP-2. These findings demonstrate that AB204 could be an alternative to rhBMP-2 to improve fusion rate.

        OPTIMIZATION OF HOT FORMING PROCESS USING DATA MINING TECHNIQUES AND FINITE ELEMENT METHOD

        In the process of hot forming, many design variables have effects on the final results in different and complex ways, such as geometry feature and forming process parameters. It is difficult to understand the relationship between design variables and results, which is very important to guide the design. In this paper, Data Mining (DM) was introduced to explore the influence of the parts geometric feature and the hot forming parameters on hot forming results of an automobile B-pillar model, and the optimum parameter ranges were determined. Firstly, a series of variable parameters were chosen and 100 groups of experimental data were generated with super Latin method, then the FEM ananlysis results were calculated respectively. Secondly, analysis and evaluation of simulation results were carried out by making full use of the Decision Tree (DT) algorithm. Finally, a series of B-pillar hot forming rules were refined, such as the initial temperature of the sheet metal should be controlled between 720 ℃ to 800 ℃. The fillet radius is recommended to be bigger than 10㎜ and the height gradient should be controlled under 67㎜, etc. A real B-pillar model was designed to testify the rules and the result shows that the rules are correct and effective.

        Law and Politics of Legislative Interpretation in China

        This article analyzes the constitutional basis and operational principles of legislative interpretation in China. Based on this analysis, it also proposes some reforms on this unique institutional arrangement in order to improve its performance in terms of its own stated reasons and justifications. I choose institutional competence, impartiality, procedural fairness, reasoning and transparency as the major parameters to evaluate the current system and to calibrate my suggestions, because these values have been accepted by a variety of political and legal systems with different agendas and commitments as essential to an institution with some concern for justice. 이 논문은 중국에서 법률해석의 헌법적 기초와 운영 원리를 분석한 것이다. 아울러 이러한 분석에 기초하여 개선을 위한 제도적 개혁을 제안한다. 필자는 현행 제도를 평가하는 지표와 제안의 근거로 기관의 역량, 공평성, 절차적 공정성, 합리성 그리고 투명성을 선택했다. 왜냐하면 이러한 가치들은 다양한 의제와 목표를 갖는 정치적 법적 시스템에서 정의를 이루기 위해 제도에 필수적으로 있어야 하는 것으로 받아들여져 왔기 때문이다.
