Background: TP73-AS1 is a novel antisense long noncoding RNA and plays an important role in cell proliferation and cancer development. However, the link between TP73-AS1 and colorectal cancer (CRC) has not yet been reported. Objective: To explore the association of genetic variants in TP73-AS1 and its expression with CRC susceptibility and prognosis. Methods: A case-control study (including 507 CRC cases and 503 controls) and bioinformatics analysis were conducted. Results: rs9800 polymorphism was significantly related to higher risk in CRC [adjusted odds ratio (AOR) = 1.33, 95% confidence interval (CI) = 1.02-1.75, P = 0.034 in heterozygote codominant model]. There was no difference between TP73-AS1 polymorphisms and different tumor node metastasis (TNM) stages in the adjusted model. Moreover, TP73-AS1 expression level was positively related to different TNM stages. After adjusted for age, gender and TNM, higher TP73-AS1 expression levels were related to shorter recurrence-free survival time [hazard ratio (HR) = 1.66, 95% CI = 1.02-2.71, P = 0.043]. Conclusion: TP73-AS1 polymorphisms and expression may be associated with susceptibility and prognosis of CRC.

A low-cycle loading experiment of 16 transfer column specimens was conducted to study the influence of parameters, likes the extension length of shape steel, the ratio of shape steel, the axial compression ratio and the volumetric ratio of stirrups, on the shear distribution between steel and concrete, the concrete damage state and the degradation of lateral stiffness. Shear force of shape steel reacted at the core area of concrete section and led to tension effect which accelerated the damage of concrete. At the same time, the damage of concrete diminished its shear capacity and resulted in the shear enlargement of shape steel. The interplay between concrete damage and shear force of shape steel ultimately made for the failures of transfer columns. With the increase of extension length, the lateral stiffness first increases and then decreases, but the stiffness degradation gets faster; With the increase of steel ratio, the lateral stiffness remains the same, but the degradation gets faster; With the increase of the axial compression ratio, the lateral stiffness increases, and the degradation is more significant. Using more stirrups can effectively restrain the development of cracks and increase the lateral stiffness at the yielding point. Also, a formula for calculating the yielding lateral stiffness is obtained by a regression analysis of the test data.

Our previous study has shown that sodium selenite can cause apoptosis in acute promyelocytic leukemia-derived NB4 cells in a caspase-dependent manner, but the detailed mechanism is unknown. Here we demonstrate a requirement for extracellular signal-regulated protein kinase (ERK) in mediating sodium selenite -induced apoptosis in NB4 cell. Though no apparent elevation of ERK activity was observed during the apoptosis in NB4 cells caused by 20 μM sodium selenite treatment, PD98059 and U0126, specific chemical inhibitors of the MEK/ERK signaling pathway, were shown to strongly prevent the apoptosis process, while ERK activator TPA enhanced the process. It is also known that p38 MAPK inhibitor SB203580 and JNK inhibitor SP600125 had slight effects on apoptosis. Further study indicated that ERK exerted its proapoptotic effect only at the early stage of apoptosis and played an antiapoptotic role at the later stages. Taken together, our findings suggest that ERK plays an active role in mediating sodium seleniteinduced apoptosis in NB4 cells .

Artemisinin is an endoperoxide sesquiterpenelactone isolated from the aerial parts of Artemisia annua L.,and is presently the most potent anti-malarial drug. Owingto the low yield of artemisinin from A. annua as well as thewidespread application of artemisinin-based combinationtherapy recommended by the World Health Organization,the global demand for artemisinin is substantially increasingand is therefore rendering artemisinin in short supply. An economical way to increase artemisinin production is toincrease the content of artemisinin in A. annua. In thisstudy, three key genes in the artemisinin biosynthesispathway, encoding farnesyl diphosphate synthase, amorpha-4, 11-diene C-12 oxidase and its redox partner cytochromeP450 reductase, were over-expressed in A. annuathrough Agrobacterium-mediated transformation. Thetransgenic lines were confirmed by Southern blotting andthe over-expressions of the genes were demonstrated byreal-time PCR assays. The HPLC analysis showed that theartemisinin contents in transgenic lines were increasedsignificantly, with the highest one found to be 3.6-foldhigher (2.9 mg/g FW) than that of the control. Theseresults demonstrate that multigene engineering is aneffective way to enhance artemisinin content in A. annua.
