Knockdown of Chloride Channel-3 Inhibits Breast Cancer Growth In Vitro and In Vivo

Purpose: Chloride channel-3 (ClC-3) is a member of the chloride channel family and plays a critical role in a variety of cellular activities. The aim of the present study is to explore the molecular mechanisms underlying the antitumor effect of silencing ClC-3 in breast cancer. Methods: Human breast cancer cell lines MDAMB- 231 and MCF-7 were used in the experiments. Messenger RNA and protein expression were examined by quantitative realtime polymerase chain reaction and western blot analysis. Cell proliferation was measured by the bromodeoxyuridine method, and the cell cycle was evaluated using fluorescence-activated cell sorting. Protein interaction in cells was analyzed by co-immunoprecipitation. Tumor tissues were stained with hematoxylin- eosin and tumor burden was measured using the Metamorph software. Results: Breast cancer tissues collected from patients showed an increase in ClC-3 expression. Knockdown of ClC-3 inhibited the secretion of insulin-like growth factor (IGF)-1, cell proliferation, and G1/S transition in breast cancer cells. In the mouse xenograft model of human breast carcinoma, tumor growth was significantly slower in animals injected with ClC- 3-deficient cells compared with the growth of normal human breast cancer cells. In addition, silencing of ClC-3 attenuated the expression of proliferating cell nuclear antigen, Ki-67, cyclin D1, and cyclin E, as well as the activation of extracellular signalregulated protein kinases (ERK) 1/2, both in vitro and in vivo. Conclusion: Together, our data suggest that upregulation of ClC- 3 by IGF-1 contributes to cell proliferation and tumor growth in breast cancer, and ClC-3 deficiency suppresses cell proliferation and tumor growth via the IGF/IGF receptor/ERK pathway.

Kaiso is a Pox Virus and Zinc Finger (POZ-ZF) transcription factor with bi-modal DNA-binding specificity. Here, we demonstrated that Kaiso expression is inversely correlated with glucocorticoid receptor (GR) expression in breast carcinomas. Knockdown of Kaiso increased GR expression, while overexpression of Kaiso inhibited GR expression in breast cancer cells. Furthermore, Kaiso repressed GR proximal promoter-reporter activity in a dose-dependent manner. Remarkably, ChIP experiments demonstrated that endogenous Kaiso was associated with the GR promoter sequence in a methylation-dependent manner. Since glucocorticoids inhibit chemotherapyinduced apoptosis and have been widely used as a co-treatment of patients with breast cancer, we assessed the role of Kasio in GR-mediated anti-apoptotic effects. We found that overexpression of Kaiso attenuated the anti-apoptotic effects of glucocorticoids in breast cancer cells. Our findings suggest that GR is a putative target gene of Kaiso and suggest Kaiso to be a potential therapeutic target in GC-combination chemotherapy in breast cancer. Study on Corner Reflectors Identification in Highway Deformation Monitoring

CRInSAR is a newly developed technique to monitor ground deformation. In CRInSAR algorithm, the identification of Corner Reflectors in SAR images is necessary. Due to the uncertainty of traditional identification method, a new method based on the intensity and correlation coefficient of each CR candidates in SAR images is presented. The method has been successfully used to determine the locations of 11 CRs installed along a highway in six SAR images over the study area. The results show that the identification accuracy of the new method is about 1 pixel. It is effective and reliable especially in the area with lots of lightspots around the CR points. The method proposed can play important role in the highway deformation monitoring within CRInSAR algorithm.

In order to investigate the characteristics of the gradient fluorinated polyacrylate and polyurethane latex blendfilms, the fluorinated polyacrylate emulsion and the polyurethane emulsion were synthesized, and then the both emulsionswere blended at a series of ratios. The effects of content of the fluorinated polyacrylate on the gradient structure and surfaceproperty of the blended films were assessed by AFM, XPS, SEM-EDX and surface free energy measurements. Itappeared that, while the content of the fluorinated polyacrylate latex was up to 30%, the fluorinated polyacrylate particleswere selectively gathered on the film-air (F-A) and film-glass (F-G) interfaces at room temperature. When the contentof the fluorinated polyacrylate was under 30%, the gradient structure of fluorinated component was not evident. The furtherincreasing of fluorinated polyacrylate in the mixed system facilitated the formation and enlargement of gradient structure,but the adhesion of film decreased a little.

Aim: To analyze the significance of different clinical factors for prognostic prediction in diffuse large B-cell lymphoma (DLBCL) patients. Methods: Two hundred and twenty-seven DLBCL patients were retrospectively reviewed. Patients were managed with cyclophosphamide, doxorubicin, vincristine, and prednisone (CHOP) regimen or rituximab plus the CHOP (RCHOP) regimen. Results: Lactate dehydrogenase (LDH), ${\beta}2$-microglobulin (${\beta}2$-M), B symptoms, Ann Arbor stage and genetic subtypes were statistically relevant in predicting the prognosis of the overall survival (OS). In the CHOP group, the OS in patients with germinal center B-cell-like (GCB)(76.2%) was significantly higher than that of the non-GCB group (51.9%, P=0.032). With RCHOP management, there was no statistical difference in OS between the GCB (88.4%) and non-GCB groups (81.9%, P=0.288). Conclusion: Elevated LDH and ${\beta}2$-M levels, positive B symptoms, Ann Arbor stage III/IV, and primary nodal lymphoma indicate an unfavorable prognosis of DLBCL patients. Patients with GCB-like DLBCL have a better prognosis than those with non-GCB when treated with the CHOP regimen. The RCHOP treatment with the addition of rituximab can improve the prognosis of patients with DLBCL.

Understanding soybean (Glycine max) domestication and improvement at a genetic level is important to inform future efforts to further improve a crop that provides the world's main source of oilseed. We detect 230 selective sweeps and 162 selected copy number variants by analysis of 302 resequenced wild, landrace and improved soybean accessions at >11× depth. A genome-wide association study using these new sequences reveals associations between 10 selected regions and 9 domestication or improvement traits, and identifies 13 previously uncharacterized loci for agronomic traits including oil content, plant height and pubescence form. Combined with previous quantitative trait loci (QTL) information, we find that, of the 230 selected regions, 96 correlate with reported oil QTLs and 21 contain fatty acid biosynthesis genes. Moreover, we observe that some traits and loci are associated with geographical regions, which shows that soybean populations are structured geographically. This study provides resources for genomics-enabled improvements in soybean breeding.

L-glutamate oxidase (GLOD) and L-amino acid oxidase (AAO) were reported to be capable of convert L-glutamic acid to α-aketoglutaric acid (α-KG). These two enzymes gene have been successfully expressed by using pHBM905BDM in Pichia pastoris to produce α-aketoglutaric acid from L-glutamic acid in our previous studies. Here these two enzymes were displayed on P. pastoris to achieve the conversion. We constructed multi-copy expression plasmids using plasmid pHBM905BDM. By using this plasmid, multi-copy strains were constructed and named as PGLOD(1-3)-AGα1 and PAAO(1-3)-AGα1, respectively. The following results showed that expression of GLOD(1-3)- AGα1 and AAO(1-3)-AGα1 in multi-copy strains increased as designed and strain PGLOD3-AGα1 and PAAO3-AGα1 was chosen for high-density fermentation and enzyme activity experiments. By using a multi-copy expression approach and high-density fermentation, we achieved a GLOD expression yield of 688.5 U/g dry cell weight and AAO expression yield of 626.7 U/g dry cell weight. By using displayed GLOD, the average production rate of L-glutamic acid to α-KG was 6.22 g/L/h and the highest α-KG titer (124.5 g/L) was converted from 135 g/L L-glutamic acid. By using displayed AAO, the average production rate of L-glutamic acid to α-KG was 5.78 g/L/h and the highest α-KG titer (115.6 g/L) was converted from 135 g/L L-glutamic acid. It showed that displaying enzymes on P. pastoris are suitable for use in industrial applications.

Purpose: To compare the relative merits of imatinib and allogeneic hematopoietic stem cell transplantation (allo-HSCT) for chronic myelogenous leukemia (CML). Materials and Methods: This cohort study was designed to compare the outcomes of imatinib (n=292) versus allo-HSCT (n=141) for CML, the clinical data of these patients being retrospectively analyzed so as to compare the event free survival (EFS) and overall survival (OS) between these two groups with patients in the chronic phase (CP) and advanced phases, including accelerate (AP) and blast phases (BP). Results: (1) Patients treated with imatinib (278 in the CP) demonstrated superior EFS, OS, 5-year EFS and 5-year OS rates of 88.5% versus 70.0% (P<0.05), 93.2% versus 80.0% (P<0.05), 84% versus 75.0% (P<0.05) and 92% versus 79.0% (P<0.05), respectively, to those treated with allo-HSCT (120 patients in the CP). (2) Both treatments resulted in similar survival, with EFS and OS rates of 42.9% versus 47.6% (P>0.05), 42.9% versus 57.1% (P> 0.05), respectively, for imatinib (14 patients in the AP and BP) and allo-HSCT (21 patients in the AP and BP). Conclusions: Imatinib confers significant survival advantage (EFS and OS) for CML patients with CP compared with allo-HSCT treatment. However, the outcomes are equally good with both treatments in AP and BP patients.
Uniform DNA Biosensors Based on Threshold Voltage of Carbon Nanotube Thin-Film Transistors

Uniformity is a key parameter to assure the accuracy of biosensor devices. In this work, highly uniform carbon nanotube thin-film transistors (CNT-TFTs) with a standard deviation of threshold voltage (Vth) as small as 0.04 were achieved by accurately controlling the fabrication process, which is so far the most stable distribution to our knowledge. On-state current (Ion), off-state current and on/off current ratio also exhibit high uniformity with low standard deviation of 0.50, 0.72 and 0.54, respectively. Given the high uniformity, high stability and high sensitivity, the CNT-TFTs are used as ultra-sensitive 5-hydroxymethyl cytosine (5-hmC) detecting devices for the first time, which is one of the important modified bases in DNA and plays an important role in epigenetics. After attachment of 5-hmC DNA, a reproducible and stable shift of 18.7–59% in Vth as well as a 31–54% change in Ion were observed in the transfer characteristics curves of CNT-TFTs. Thus, a detecting device of 5-hmC in DNA segments could be designed based on the highly uniform CNT-TFTs.
