Aflatoxin B₁was converted to aflatoxin B₁derivative to get a coupling site in the presence of bromine: carbontertrachloride mixture, yielding 82.9% and then the derivative was conjugate to BSA. Hybridoma cell antibody against aflatoxin B₁conjugate was produced by fusing myeloma cells with spleen cells of Balb/c female mice immunized with aflatoxin B₁-BSA conjugate. The antibody obtained was specially reacted with aflatoxin B₁. Competition assats as performed by incubation of different aflatoxin analogues with the peroxidase conjugate shpwed that aflatoxin B₁caused the most inhibition of conjugate binding to antibody. Aflstoxin G₁, G₂and M₁inhibited the conjugate binding to a lesser degree, whereas aflatoxin B₂, M₂and sterigmatocystin had no effect on the assay. It took only 45minutes to do whole ELISA procedure for the analysis of aflatoxin B₁by the direct competitive ELISA system.

To investigate the effect of cultural condition on the aflatoxin production of Aspergillus flavus ATCC 15517, mixed culture with AspergiI/us niger, better kind of media and size of cu1tural vessels were examined. YES medium was better than SLS medium for this study. Small scale test tube culture was showed the possibility to simply examine the growth, total acidity, pH and aflatoxin production during cultivation, and also could reduce the second contamina. tion of aflatoxin B 1 from large scale broth cultured. Especially ELISA method is simple, sensitive and specific and therefore well suited to small scale of test tube culture. Mixed culture significantly reduced the aflatoxin production of AspergiI/us flavus ATCC ) 5517 and showed almost 95% inhibition of that level during the incubtation.

This study was designed to observe the effect of temperature cycling on the production of aflatoxin by Aspergillus parasiticus R-716 in rice, bar1ey , peanut and soybean. In those media, temperature cyc1ing resulted in more total aflatoxin production by the strain of R-716 than constant incubatïon at 280C and natural condition did. Especially, high level of total aflatoxin (1 826μg/30g) in rice medium at temperature cyc1ing was produced. The intensity of yellow color of chlbroform extracts correlated with the concentration of aflatoxin, and the ratio of aflatoxin 8 1 to at1atoxin B2 ,G1 ,G2 is lower at temperature cyc1ing condition than at 280 C.
