A Hydrogel-based First-Aid Tissue Adhesive with Effective Hemostasis and Anti-bacteria for Trauma Emergency Management

        Dongjie Zhang,Li Mei,Yuanping Hao,Bingcheng Yi,Jilin Hu,Danyang Wang,Yaodong Zhao,Zhe Wang,Hailin Huang,Yongzhi Xu,Xuyang Deng,Cong Li,Xuewei Li,Qihui Zhou,Yun Lu 한국생체재료학회 2023 생체재료학회지 Vol.27 No.00

Background Clinical tissue adhesives remain some critical drawbacks for managing emergency injuries, such as inadequate adhesive strength and insufficient anti-infection ability. Herein, a novel, self-healing, and antibacterial carboxymethyl chitosan/polyaldehyde dextran (CMCS/PD) hydrogel is designed as the first-aid tissue adhesive for effective trauma emergency management. Methods We examined the gel-forming time, porosity, self-healing, antibacterial properties, cytotoxicity, adhesive strength, and hemocompatibility. Liver hemorrhage, tail severance, and skin wound infection models of rats are constructed in vivo, respectively. Results Results demonstrate that the CMCS/PD hydrogel has the rapid gel-forming (~ 5 s), good self-healing, and effective antibacterial abilities, and could adhere to tissue firmly (adhesive strength of ~ 10 kPa and burst pressure of 327.5 mmHg) with excellent hemocompatibility and cytocompatibility. This suggests the great prospect of CMCS/ PD hydrogel in acting as a first-aid tissue adhesive for trauma emergency management. The CMCS/PD hydrogel is observed to not only achieve rapid hemostasis for curing liver hemorrhage and tail severance in comparison to commercial hemostatic gel (Surgiflo ®) but also exhibit superior anti-infection for treating acute skin trauma compared with clinical disinfectant gel (Prontosan ®). Conclusions Overall, the CMCS/PD hydrogel offers a promising candidate for first-aid tissue adhesives to manage the trauma emergency. Because of the rapid gel-forming time, it could also be applied as a liquid first-aid bandage for mini-invasive surgical treatment.

Effects of alpha-linolenic acid and essential amino acids on the proliferation and differentiation of C2C12 myoblasts

        Dongjie Zhou,Xiao-Han Li,Song‑Hee Lee,Geun Heo,Xiang-Shun Cui 한국동물생명공학회(구 한국수정란이식학회) 2022 한국동물생명공학회지 Vol.37 No.1

Alpha-linolenic acid is an important polyunsaturated fatty acid that exhibits anticancer, anti-inflammatory, and antioxidative effects. In this study, we investigated the protective effects of alpha-linolenic acid on the cell proliferation and differentiation of C2C12 cells under essential amino acid-deficient conditions. Different concentrations of alpha-linolenic acid and essential amino acids were added to the growth and differentiation media. The concentrations of 10 μM of alphalinolenic acid and 2% essential amino acid were chosen for subsequent experiments. Supplementation with alpha-linolenic acid and essential amino acids improved the proliferation and differentiation of C2C12 cells and significantly increased the mRNA levels of catalase, superoxide dismutase, B-cell lymphoma-2, and beclin-1 as well as the protein levels of PPARγ coactivator-1α compared to those in the controls. Moreover, supplementation with alpha-linolenic acid and essential amino acids reduced the levels of phosphorylated H2A.X variant histone, Bcl-2-associated X, p53, and light chain 3 during C2C12 cell proliferation, and increased the expression levels of myogenic factors 4 (myogenin) and 5 during C2C12 cell differentiation. Overall, we determined that alpha-linolenic acid and essential amino acids maintained the cell proliferation and differentiation of C2C12 cells via their anti-oxidative, anti-apoptotic, and anti-autophagic effects.

Optimal operating points of PEM fuel cell model with RSM

Dongji Xuan,Zhenzhe Li,김진환,김영배 대한기계학회 2009 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.23 No.3

The output power efficiency of the fuel cell system mainly depends on the required current, stack temperature, air excess ratio, hydrogen excess ratio, and inlet air humidity. Therefore, the operating conditions should be optimized to get maximum output power efficiency. In this paper, a dynamic model for the fuel cell stack was developed, which is comprised of a mass flow model, a gas diffusion layer model, a membrane hydration, and a stack voltage model. Experiments have been performed to calibrate the dynamic Polymer Electrolyte Membrane Fuel Cell (PEMFC) stack model. To achieve the maximum output power and the minimum use of hydrogen in a certain power condition, optimization was carried out using Response Surface Methodology (RSM) based on the proposed PEMFC stack model. Using the developed method, optimal operating conditions can be effectively selected in order to obtain minimum hydrogen consumption.

Experimental study on the cable rigidness and static behaviors of AERORail structure

Fangyuan Li,Peifeng Wu,Dongjie Liu 국제구조공학회 2012 Steel and Composite Structures, An International J Vol.12 No.5

This paper presented a new aerial platform-AERORail for rail transport and its structure evolution based on the elastic stiffness of cable; through the analysis on the cable properties when the cable supported a small service load with high-tensile force, summarized the theoretical basis of the AERORail structure and the corresponding simplified analysis model. There were 60 groups of experiments for a single naked cable model under different tensile forces and different services loads, and 48 groups of experiments for the cable with rail combined structure model. The experimental results of deflection characteristics were compared with the theoretical values for these two types of structures under the same conditions. It proved that the results almost met the classical cable theory. The reason is that a small deflection was required when this structure was applied. After the tension increments tests with moving load, it is verified that the relationships between the structure stiffness and tension force and service load are simple. Before further research and applications are made, these results are necessary for the determination of the reasonable and economic tensile force, allowable service load for the special span length for this new platform.

Simplified Analytical Model for Investigating the Output Power of Solar Array on Stratospheric Airship

Zhang, Yuanyuan,Li, Jun,Lv, Mingyun,Tan, Dongjie,Zhu, Weiyu,Sun, Kangwen The Korean Society for Aeronautical and Space Scie 2016 International Journal of Aeronautical and Space Sc Vol.17 No.3

Solar energy is the ideal power choice for long-endurance stratospheric airships. The output performance of solar array on stratospheric airship is affected by several major factors: flying latitude, flight date, airship's attitude and the temperature of solar cell, but the research on the effect of these factors on output performance is rare. This paper establishes a new simplified analytical model with thermal effects to analyze the output performance of the solar array. This model consisting of the geometric model of stratospheric airship, solar radiation model and incident solar radiation model is developed using MATLAB computer program. Based on this model, the effects of the major factors on the output performance of the solar array are investigated expediently and easily. In the course of the research, the output power of solar array is calculated for five airship's latitudes of $0^{\circ}$, $15^{\circ}$, $30^{\circ}$, $45^{\circ}$ and $60^{\circ}$, four special dates and different attitudes of five pitch angles and four yaw angles. The effect of these factors on output performance is discussed in detail. The results are helpful for solving the energy problem of the long endurance airship and planning the airline.

METTL3 m6A-dependently promotes miR-21-5p maturation to accelerate choriocarcinoma progression via the HIF1AN-induced inactivation of the HIF1A/VEGF pathway

        Ye Kefan,Li Lingchuan,Wu Bao,Wang Dongjie 한국유전학회 2022 Genes & Genomics Vol.44 No.11

Background: Gestational choriocarcinoma is a highly malignant neoplastic disease derived from pathological changes in trophoblastic cells. Recent evidences have shown that N6-methyladenosine (m6A) modifications play important role in modulating the development of multiple cancers, but the detailed mechanisms by which m6A-mediated choriocarcinoma progression have not been fully delineated. Objectives: This study aimed to investigate the role of m6A in choriocarcinoma and reveal its underlying molecular mechanisms. Methods: The expression of METTL3, miR-21-5p and HIF1AN was detected using RT-qPCR in tissues and cells. The protein expression of METTL3, HIF1AN, HIF1A and VEGF were measured by western blot. The luciferase reporter assays and RNA immunoprecipitation (RIP) were used to verify the relationship between miR-21-5p and HIF1AN. The CCK-8, colony formation and transwell assays were used to detected cell proliferation and cell migration, respectively. Results: Here, we demonstrated that the m6A methyltransferase-like 3 (METTL3) was aberrantly high-expressed in the clinical choriocarcinoma tissues and choriocarcinoma cell lines compared to the corresponding normal counterparts. The following functional experiments verified that silencing of METTL3 suppressed cell proliferation, migration, epithelial-mesenchymal transition (EMT) and tumorigenesis in vitro and in vivo to hamper the aggressiveness of choriocarcinoma. Next, the mechanical experiments confirmed that METTL3 promoted the maturation of miR-21-5p in an m6A-dependent manner, and elevated miR-21-5p subsequently degraded its downstream hypoxia-inducible factor asparagine hydroxylase (HIF1AN) by targeting its 3' untranslated regions (3'-UTR), resulting in the activation of the tumor-promoting HIF1A/VEGF pathway. Finally, the rescuing experiments verified that METTL3 ablation-induced inhibitory effects on the malignant phenotypes in choriocarcinoma were all abrogated by both miR-21-5p overexpression and HIF1AN downregulation. Conclusions: Collectively, this study firstly reported the involvement of the METTL3/m6A/miR-21-5p/HIF1AN signaling cascade in regulating the progression of choriocarcinoma, which provided novel biomarkers for the diagnosis and treatment of this disease.
