        Antifungal Effect of Triglycerol Monolaurate Synthesized by Lipozyme 435-Mediated Esterification

        This study was designed to synthesize triglycerol monolaurate (TGML) with Lipozyme 435 as the catalyst, and explore its effects on the growth of Aspergillus parasiticus (A. parasiticus) and Aspergillus flavus (A. flavus) and the secretion of aflatoxin b1. The highest content of TGML (49.76%) was obtained at a molar ratio of triglycerol to lauric acid of 1.08, a reaction temperature of 84.93°C, a reaction time of 6 h and an enzyme dosage of 1.32%. After purification by molecular distillation combined with the washes with ethyl acetate and water, the purity of TGML reached 98.3%. Through characterization by electrospray-ionization mass spectrometry, infrared spectrum and nuclear magnetic resonance, the structure of TGML was identified as a linear triglycerol combined with lauroyl at the end. Finally, the inhibitory effects of TGML on the growths of A. parasiticus and A. flavus and the secretion of aflatoxin b1 were evaluated by measuring the colony diameter, the inhibition rate of mycelial growth and the content of mycotoxin in the media. The results indicated that TGML had a stronger inhibitory effects on colony growth and mycelial development of both toxic molds compared to sodium benzoate and potassium sorbate, and the secretions of toxins from A. parasiticus and A. flavus were completely suppressed when adding TGML at 10 and 5 mM, respectively. Based on the above results, TGML may be used as a substitute for traditional antifungal agents in the food industry.

        Study on Mechanical Behaviors of Unsaturated Loess in terms of Moistening Level

        Water sensitivity is the special feature of unsaturated loess, which is the main cause of engineering problems in loessial regions. Previous studies have generally focused on potential changes on mechanical properties of loess when it is moistened to saturation, while the moistening process is often ignored. In order to investigate the mechanical behaviors of unsaturated loess during the moistening process, this paper takes unsaturated loess obtained from Ili, Xinjiang Autonomous Regions, China, as study object. A self-developed oedometer was used which was available for adding water into the soil sample during loading, measuring the coefficient of earth pressure at rest and matrix suction during moistening. Loess samples obtained from 3 buried depths were tested under 4 different vertical loads. The moistening level was defined to refer to the relative level of water content with regard to the initial and saturated water contents of a soil sample. Change tendencies of moistening deformation, the coefficient of earth pressure at rest as well as suction versus the moistening level were investigated.

        Synthesis of a novel triazine flame retardant containing sulfur and its application to cotton fabrics

        A novel triazine flame retardant (FR) has been successfully synthesized by the reaction of cyanuric chloride with sodium sulfanilate followed by diethanol amine. Its structure has been fully characterized by IR and 1H NMR spectroscopy. The effects of reaction solvent, acid-binding agent, reaction temperature, and molar ratio of starting materials on the yield of FR were investigated in detail. Experiments showed that when the molar ratio of intermediate I to diethanol amine is 1 to 1.2 in the presence of sodium carbonate as acid-binding agent in acetone-water at 45 oC, the yield of the obtained FR reaches 81.4%. Thermogravimetry (TG) test indicated that the FR has good thermostability and char-forming ability. In addition, the flame retardancy and thermal behavior of the cotton fabrics treated with FR were studied by limiting oxygen index (LOI), vertical flammability test, and thermogravimetry (TG) to evaluate the flameretardant performance of the target FR. These results demonstrated that the flame retardancy and thermal stability of the cotton fabrics treated with FR were clearly improved.

        PAX8-AS1 knockdown facilitates cell growth and inactivates autophagy in osteoblasts via the miR-1252-5p/GNB1 axis in osteoporosis

        Osteoporosis (OP) is the most common systematic bone disorder among elderly individuals worldwide. Long noncoding RNAs (lncRNAs) are involved in biological processes in various human diseases. It has been previously revealed that PAX8 antisense RNA 1 (PAX8-AS1) is upregulated in OP. However, its molecular mechanism in OP remains unclear. Therefore, we specifically designed this study to determine the specific role of PAX8-AS1 in OP. We first established a rat model of OP and then detected PAX8-AS1 expression in the rats with RT-qPCR. Next, to explore the biological function of PAX8-AS1 in osteoblasts, in vitro experiments, such as Cell Counting Kit-8 (CCK-8) assays, flow cytometry, western blotting and immunofluorescence (IF) staining, were conducted. Subsequently, we performed bioinformatic analysis and luciferase reporter assays to predict and identify the relationships between microRNA 1252-5p (miR-1252-5p) and both PAX8-AS1 and G protein subunit beta 1 (GNB1). Additionally, rescue assays in osteoblasts clarified the regulatory network of the PAX8-AS1/miR-1252-5p/GNB1 axis. Finally, in vivo loss-of-function studies verified the role of PAX8-AS1 in OP progression. The results illustrated that PAX8-AS1 was upregulated in the proximal tibia of OP rats. PAX8-AS1 silencing promoted the viability and inhibited the apoptosis and autophagy of osteoblasts. PAX8-AS1 interacted with miR-1252-5p. GNB1 was negatively regulated by miR-1252-5p. In addition, the impacts of PAX8-AS1 knockdown on osteoblasts were counteracted by GNB1 overexpression. PAX8-AS1 depletion suppressed OP progression by inhibiting apoptosis and autophagy in osteoblasts. In summary, PAX8-AS1 suppressed the viability and activated the autophagy of osteoblasts via the miR-1252-5p/GNB1 axis in OP.
