On the Disturbance Rejection Control of Flexible-joint Robot: A GPIO-based Approach

A robust disturbance rejection control scheme is addressed for the trajectory tracking problem of a flexible-joint robot (FJR). The system is always severely affected by various types of unknown disturbances including model errors, couplings, changing working environments as well as unmodeled dynamics. These disturbances on the link and actuator side will deteriorate the control performance of FJR. By considering all the disturbances as an unknown lumped time-varying disturbance, a flatness description of FJR is developed. Then, a new output feedback controller is constructed through the estimates of unmeasurable states and unknown lumped disturbance provided by a generalized proportional integral observer (GPIO). The stability of the closed-loop system with the driven of the proposed control scheme is guaranteed under some mild assumptions. Compared with the conventional linear active disturbance rejection control (LADRC) scheme, test results are presented to demonstrate the feasibility and efficacy of the proposed control approach.

Kidney renal clear cell carcinoma (KIRC) remains a significant challenge worldwide because of its poor prognosis and high mortality rate, and accurate prognostic gene signatures are urgently required for individual therapy. This study aimed to construct and validate a seven-gene signature for predicting overall survival (OS) in patients with KIRC. The mRNA expression profile and clinical data of patients with KIRC were obtained from The Cancer Genome Atlas (TCGA) and International Cancer Genome Consortium (ICGC). Prognosis-associated genes were identified, and a prognostic gene signature was constructed. Then, the prognostic efficiency of the gene signature was assessed. The results obtained using data from the TCGA were validated using those from the ICGC and other online databases. Gene set enrichment analyses (GSEA) were performed to explore potential molecular mechanisms. A seven-gene signature (PODXL, SLC16A12, ZIC2, ATP2B3, KRT75, C20orf141, and CHGA) was constructed, and it was found to be effective in classifying KIRC patients into high- and low-risk groups, with significantly different survival based on the TCGA and ICGC validation data set. Cox regression analysis revealed that the seven-gene signature had an independent prognostic value. Then, we established a nomogram, including the seven-gene signature, which had a significant clinical net benefit. Interestingly, the seven-gene signature had a good performance in distinguishing KIRC from normal tissues. GSEA revealed that several oncological signatures and GO terms were enriched. This study developed a novel seven-gene signature and nomogram for predicting the OS of patients with KIRC, which may be helpful for clinicians in establishing individualized treatments.

Synthesis and Enhanced Photocatalytic Activity of Black Porous Zr-doped TiO2 Monoliths

Black Porous Zr-doped TiO2 monoliths were successfully synthesized by a combined route including sol–gel method with polystyrene spheres as template followed by calcination at high temperature in the presence of oxygen. As-synthesized samples were then treated in hydrogen atmosphere at 500℃ for 6 h to obtain black Porous Zr-doped TiO2 monoliths. This as-synthesized black porous Zr-doped TiO2 composite showed relatively narrow bandgap compared to the pure anatase phase of TiO2, which may have its origin from the contributions from the Zr impurity and oxygen vacancies. The photocatalytic activity of the black porous Zr-doped TiO2 monolith was examined by carrying out the dye degradation on Rhodamine B under visible irradiation. Our experimental results indicate that black porous 0.9% Zr-doped TiO2 sample exhibits high photocatalytic activity with the photodecomposition of 95.25% under visible irradiation for 120 min.

Magnetic resonant wireless power transfer (MR-WPT) has become a significant method for powering the measurement units in landslide monitoring boreholes. This paper focuses on the effects of quality factor, coupling distance and load on the power transfer stability, sensitivity and performance of an MR-WPT system. Firstly, through a numerical model of an MR-WPT, the effect of quality factor and coupling distance on the output power, transfer efficiency and frequency splitting phenomenon of MR-WPT systems have been comparatively analyzed. The relationship between the load and the coupling distance region corresponding to optimal transfer performance has been studied. Results show that improving the quality factor of the coils is beneficial for the transfer performance and stability of WPT system. Furthermore, through the selection of the load, a coupling distance region with high transfer stability and an optimal tradeoff between power and efficiency can be obtained. Finally, the above theoretical simulation results have been verified by experimental results.

A novel OxyR (DR0615) with one conserved cysteine that senses hydrogen peroxide in Deinococcus radiodurans had been identified in our previous work. Comparative genomics revealed that D. radiodurans possesses another OxyR homolog, OxyR2 (DRA0336). In this study, we constructed the deletion mutant of oxyR2 and the double mutant of both the OxyR homologs to investigate the role of OxyR in response to oxidative stress in D. radiodurans. Deletion of oxyR2 resulted in an obviously increased sensitivity to hydrogen peroxide, and the double mutant for oxyR and oxyR2 was significantly more sensitive than any of the two single mutants. The total catalase activity of the double mutant was lower than that of any of the single mutants, and reactive oxygen species (ROS) accumulated to a greater extent. DNA microarray analysis further suggested that oxyR2 was involved in antioxidation mechanisms. Site-direct mutagenesis and complementation analysis revealed that C228 in OxyR2 was essential. This is the first report of the presence of two OxyR in one organism. These results suggest that D. radiodurans OxyR and OxyR2 function together to protect the cell against oxidative stress.

RNA interference is an efficient approach for gene function identification and a potential novel strategy for selectively controlling pests. The red flour beetle, Tribolium castaneum (Coleoptera: Tenebrionidae), is a major global storage pest, which causes great economic loss. Juvenile hormone esterase (JHE), a carboxylesterase, is responsible for the degradation of juvenile hormone. However, the knockdown effect of jhe on metamorphosis of pupae or adult in T. castaneum is unknown. In this study, we analyzed the expression profiles of Tc j h e in the larval stage, we found that Tc j h e was expressed throughout the whole larval instars and the expression levels were relatively high right after molting. Furthermore, we knocked down the expression of Tcjhe by injecting dsTc j h e , which significantly increased the mortality of adults and decreased the eclosion rate. In addition, abnormal developmental phenotypes, including wing deformitity, pupal–adult monsters and shrunken adults, were observed. Our finding indicates that JHE plays an important role in the metamorphosis and development in T. castaneum, suggesting that Tc j h e could be used as a potential target for the development of RNAi-based control strategies in T. castaneum.
