Crocetin Induces Cytotoxicity in Colon Cancer Cells Via p53-independent Mechanisms

Objective: Crocin has been proposed as a promising candidate for cancer chemoprevention. The purpose of this investigation was to investigate the chemopreventive action and the possible mechanisms of crocin against human colon cancer cells in vitro. Methods: Cell proliferation was examined using MTT assay and the cell cycle distribution fractions were analyzed using fow cytometric analysis after propidium iodide staining. Apoptosis was detected using theTUNEL Apoptosis Detection Kit with laser scanning confocal microscope. DNA damage was assessed using the alkaline single-cell gel electrophoresis assay, while expression levels of p53, cdk2, cyclinA and P21 were examined by Western blot analysis. Results: Treatment of SW480 cells with crocetin (0.2, 0.4, 0.8 mmol/L) for 48 h signifcantly inhibited their proliferation in a concentration-dependent manner. Crocetin (0.8 mmol/L) signifcantly induced cell cycle arrest through p53-independent mechanisms accompanied by P21 induction. Crocetin (0.8 mmol/L) caused cytotoxicity in the SW480 cells by enhancing apoptosis and decreasing DNA repair capacity in a time-dependent manner. Conclusions: This report provides evidence that crocetin is a potential anticancer agent, which may be used as a chemotherapeutic drug.

4-Methylsulfinyl-3-butenyl isothiocyanate (MTBITC) found in the radish (Raphanus sativus L.), is a wellknown anticancer agent. In this study, the mechanisms of the MTBITC induction of cell apoptosis in human A549 lung cancer cells were investigated. Our PI staining results showed that MTBITC treatment significantly increased the apoptotic sub-G1 fraction in a dose-dependent manner. The mechanism of apoptosis induced by MTBITC was investigated by testing the change of mitochondrial membrane potential (${\Delta}{\Psi}m$), the expression of mRNAs of apoptosis-related genes by RT-PCR, and the activities of caspase-3 and -9 by caspase colorimetric assay. MTBITC treatment decreased mitochondrial membrane potential by down-regulating the rate of Bcl-2/Bax and Bcl-xL/Bax, and activation of caspase-3 and -9. Therefore, mitochondrial pathway and Bcl-2 gene family could be involved in the mechanisms of A549 cell apoptosis induced by MTBITC.

This paper proposed a discrete wavelet transform based method for time-varying physical parameter identification of shear type structures. The time-varying physical parameters are dispersed and expanded at multi-scale as profile and detail signal using discrete wavelet basis. To reduce the number of unknown quantity, the wavelet coefficients that reflect the detail signal are ignored by setting as zero value. Consequently, the time-varying parameter can be approximately estimated only using the scale coefficients that reflect the profile signal, and the identification task is transformed to an equivalent time-invariant scale coefficient estimation. The time-invariant scale coefficients can be simply estimated using regular least-squares methods, and then the original time-varying physical parameters can be reconstructed by using the identified time-invariant scale coefficients. To reduce the influence of the ill-posed problem of equation resolving caused by noise, the Tikhonov regularization method instead of regular least-squares method is used in the paper to estimate the scale coefficients. A two-story shear type frame structure with time-varying stiffness and damping are simulated to validate the effectiveness and accuracy of the proposed method. It is demonstrated that the identified time-varying stiffness is with a good accuracy, while the identified damping is sensitive to noise.
Damage detection of nonlinear structures with analytical mode decomposition and Hilbert transform

This paper proposes an analytical mode decomposition (AMD) and Hilbert transform method for structural nonlinearity quantification and damage detection under earthquake loads. The measured structural response is first decomposed into several intrinsic mode functions (IMF) using the proposed AMD method. Each IMF is an amplitude modulated-frequency modulated signal with narrow frequency bandwidth. Then, the instantaneous frequencies of the decomposed IMF can be defined with Hilbert transform. However, for a nonlinear structure, the defined instantaneous frequencies from the decomposed IMF are not equal to the instantaneous frequencies of the structure itself. The theoretical derivation in this paper indicates that the instantaneous frequency of the decomposed measured response includes a slowly-varying part which represents the instantaneous frequency of the structure and rapidly-varying part for a nonlinear structure subjected to earthquake excitations. To eliminate the rapidly-varying part effects, the instantaneous frequency is integrated over time duration. Then the degree of nonlinearity index, which represents the damage severity of structure, is defined based on the integrated instantaneous frequency in this paper. A one-story hysteretic nonlinear structure with various earthquake excitations are simulated as numerical examples and the degree of nonlinearity index is obtained. Finally, the degree of nonlinearity index is estimated from the experimental data of a seven-story building under four earthquake excitations. The index values for the building subjected to a low intensity earthquake excitation, two medium intensity earthquake excitations, and a large intensity earthquake excitation are calculated as 12.8%, 23.0%, 23.2%, and 39.5%, respectively.

<P>Understanding site-selective fluorescence is one of valuable importance for spectrum modulation. In this Letter, we observed the existence of two non-equivalent Gd-activated crystallographic sites in an Er3+-doped hexagonal Na1.5Gd1.5F6 phosphor. It is proved that two green emissions from the S-4(3/2) level separately originate from the Gd1 (540 nm) and Na2/Gd2 (550-555 nm) crystallographic sites, and the 657 nm red emission from the F-4(9/2) level only originates from Na2/Gd2 site through using the time-resolved luminescence spectra. The 142.2% absolute enhancement of the red emission is realized through the synergistic effect of ultraviolet downconversion and infrared upconversion induced by the 370 nm and 1.54 mu m dual-mode excitation. A non-thermal plasma (NTP) reactor was used to generate ozone for diesel particulate filter (DPF) regeneration. The kinetic mechanism of ozone thermal decomposition reaction is explored, and the effect of temperature on the change of ozone concentration is analyzed. The changes of the internal temperature and the concentration of regeneration products such as CO2 and CO during the regeneration under constant temperature and non-constant temperature conditions are then studied and the influence of different regeneration environments on the regeneration is analyzed in combination with the decomposition law of ozone. In the non-constant temperature condition, DPF surface temperature changes significantly with time. The results show that when using an NTP reactor to generate ozone, the activation energy of the decomposition reaction is 2.80755 × 104 J/mol and the law of thermal decomposition reaction can be described as 190.76 . /. During DPF regeneration, the overall regeneration rate and ozone utilization rate in the nonconstant temperature environment are higher than in the constant temperature environment and the temperature change (delta-T) peak rises with the increase of PM deposition. The regeneration with NTP under non-constant thermal condition is an effective way to improve the efficiency of DPF regeneration.

<P>We report the discovery of an ultra-luminous quasar J030642.51+185315.8 (hereafter J0306+1853) at redshift 5.363, which hosts a supermassive black hole with M-BH = (1.07 +/- 0.27) x 10(10) M-circle dot. With an absolute magnitude M-1450 = -28.92 and a bolometric luminosity L-bol similar to 3.4 x 10(14) L-circle dot, J0306+1853 is one of the most luminous objects in the early universe. It is not likely to be a beamed source based on its small flux variability, low radio loudness, and normal broad emission lines. In addition, a z = 4.986 damped Ly alpha system (DLA) with [M/H] = -1.3 +/- 0.1, among the most metal-rich DLAs at z greater than or similar to 5, is detected in the absorption spectrum of this quasar. This ultra-luminous quasar puts strong constraints on the bright end of the quasar luminosity function and massive end of the black hole mass function. It will provide a unique laboratory for the study of BH growth and the co-evolution between a BH and the host galaxy with multi-wavelength follow-up observations. The future high-resolution spectra will give more insight into the DLA and other absorption systems along the line of sight of J0306+1853.</P>

The COPS3 gene has stimulating effect on cell proliferation and progression of osteosarcomas and related cells. However, the features of COPS3 and its potential application as a therapeutic target in other cancers has not yet been studied. In this study, therefore, the effect of COPS3 silencing via COPS3 siRNA on lung cancer cell proliferation was examined. Expression levels of COPS3 gene in COPS3 siRNA infected cells and control siRNA infected cells were compared with real time PCR and Western blot analysis. Cell proliferation levels were comprehensively analyzed by MTT, BrdU incorporationy, and colony formation assays. For mechanistic assessment the effects of COPS3 silencing on cell cycle and apoptosis were analyzed using flow cytometry. Results showed that successful silencing of the COPS3 gene at both translational and transcriptional levels significantly reduced the proliferation and colony formation by lung cancer cells (p<0.01). Flow cytometry showed cell cycle arrest in the G0/G1 phase after COPS3 silencing, and more importantly, apoptosis was induced as a result of COPS3 knockdown, which negatively affected cell survival. Therefore, these results provide another piece of important evidence that the COPS3 gene expressed in lung cancer cells may play a critical role in stimulating proliferation. Down-regulation of COPS3 could significantly inhibit lung cancer cell growth, which was most likely mediated via induction of cell cycle arrest in G0/G1 phase and apoptosis.
