Numerical analysis of under-designed reinforced concrete beam-column joints under cyclic loading

In the present study, exterior beam-column sub-assemblage from a regular reinforced concrete (RC) building has been considered. Two different types of beam-column sub-assemblages from existing RC building have been considered, i.e., gravity load designed (‘GLD’), and seismically designed but without any ductile detailing (‘NonDuctile’). Hence, both the cases represent the under-designed structure at different time frame span before the introduction of ductile detailing. For designing ‘NonDuctile’ structure, Eurocode and Indian Standard were considered. Non-linear finite element (FE) program has been employed for analysing the sub-assemblages under cyclic loading. FE models were developed using quadratic concrete brick elements with embedded truss elements to represent reinforcements. It has been found that the results obtained from the numerical analysis are well corroborated with that of experimental results. Using the validated numerical models, it was proposed to correlate the energy dissipation from numerical analysis to that from experimental analysis. Numerical models would be helpful in practice to evaluate the seismic performance of the critical sub-assemblages prior to design decisions. Further, using the numerical studies, performance of the sub-assemblages with variation of axial load ratios (ratio is defined by applied axial load divided by axial strength) has been studied since many researchers have brought out inconsistent observations on role of axial load in changing strength and energy dissipation under cyclic load.

<P>This study reports a rapid screening method for the prediction of oral drug bioavailability in humans based on combined immobilized artificial membrane (IAM) chromatographic capacity factor (k<SUB>IAM</SUB>) and in vitro stability in hepatic microsomes. The fraction of drug absorbed (F<SUB>a</SUB>) in humans was predicted for a set of 15 structurally diverse commercial drugs based on k<SUB>IAM</SUB> values using a mobile phase consisting of acetonitrile: Dulbecco's phosphate-buffered saline. The hepatic intrinsic clearance (CL<EM><SUB>int</SUB><SUP>′</SUP></EM>) was calculated from in vitro disappearance half-life, and the oral bioavailability was predicted using in vitro hepatic clearance (CL<SUB>h</SUB>) and F<SUB>a</SUB>. Significant correlations were observed for the relationships between predicted hepatic extraction ratios (ER<SUB>h</SUB>) and actual presystemic metabolism (r = 0.854) and between predicted and observed oral bioavailabilities (r = 0.805; p < 0.01). The IAM capacity factor together with the hepatic microsomal disappearance half-life may be useful in identifying compounds with high oral absorption potential in early drug discovery processes. <P>A rapid and sensitive liquid chromatography/mass spectrometry (LC/MS) method was developed and validated for the determination of roxatidine in human plasma. Roxatidine was extracted by single liquid-liquid extraction with tert-butyl methyl ether, and the chromatographic separation was performed on a C<SUB>8</SUB> column. The total analytical run time was relatively short (5 min), and the limit of assay quantification was 2 ng/mL using 0.1 mL of human plasma. Roxatidine and the internal standard, propranolol, were monitored in selected ion monitoring (SIM) mode at m/z 307.3 and 260.3, respectively. The standard curve was linear over a concentration range from 2–500 ng/mL, and the correlation coefficients were >0.999. The mean intra- and inter-day assay accuracy ranged from 103.4–108.8% and 102.3–110.0%, respectively, and the mean intra- and inter-day precision was between 3.3–8.8% and 5.3–6.2%, respectively. The developed assay method was successfully applied to a pharmacokinetic study in human volunteers after oral administration of roxatidine acetate hydrochloride at a dose of 75 mg.
