Scrapie is diagnosed antemortem in sheep by detecting misfolded isoforms of prion protein (PrPSc) in lymphoid follicles of the rectal mucosa and nictitating membranes. Assay sensitivity is limited if (a) the biopsy is collected early during disease development, (b) an insufficient number of follicles is collected, or (c) peripheral accumulation of PrPSc is reduced or delayed. A blood test would be convenient for mass live animal scrapie testing. Currently approved techniques, however, have their own detection limits. Novel detection methods may soon offer a non-animal-based, rapid platform with detection sensitivities that rival the prion bioassay. In anticipation, we sought to determine if diseased animals could be routinely identified with a bioassay using B lymphocytes isolated from blood sample volumes commonly collected for diagnostic purposes in small ruminants. Scrapie transmission was detected in five of six recipient lambs intravenously transfused with B lymphocytes isolated from 5∼10 mL of blood from a naturally scrapie-infected sheep. Additionally, scrapie transmission was observed in 18 ovinized transgenic Tg338 mice intracerebrally inoculated with B lymphocytes isolated from 5∼10 mL of blood from two naturally scrapie-infected sheep. Based on our findings, we anticipate that these blood sample volumes should be of diagnostic value.

Ballasting and deballasting of shipping vessels in foreign ports have been reported worldwide as a vector of introduction of non-native aquatic plants and animals. Recently, attention has turned to ballast water as a factor in the global increase of harmful algal blooms (HABs). Many species of microalgae, including harmful dinoflagellate species, can remain viable for months in dormant benthic stages (cysts) in ballast sediments. Over a period of four years, we surveyed ballast water and sediment of ships docked in two ports of Tampa Bay, Florida, USA. Sampling conditions encountered while sampling ballast water and sediments were vastly different between vessels. Since no single sample collection protocol could be applied, existing methods for sampling ballast were modified and new methods created to reduce time and labor necessary for the collection of high-quality, qualitative samples. Five methods were refined or developed,including one that allowed for a directed intake of water and sediments. From 63 samples, 1,633 dinoflagellate cysts and cyst-like cells were recovered. A native, cyst-forming, harmful dinoflagellate, Alexandrium balechii (Steidinger) F. J. R. Taylor, was collected, isolated, and cultured from the same vessel six months apart, indicating that ships exchanging ballast water in Tampa Bay have the potential to transport HAB species to other ports with similar ecologies, exposing them to non-native, potentially toxic blooms.
Biomechanical Comparison of the Latarjet Procedure with and without Capsular Repair

Background: The purpose of this study was to determine if capsular repair used in conjunction with the Latarjet procedure results in significant alterations in glenohumeral rotational range of motion and translation. Methods: Glenohumeral rotational range of motion and translation were measured in eight cadaveric shoulders in 90° of abduction in both the scapular and coronal planes under the following four conditions: intact glenoid, 20% bony Bankart lesion, modified Latarjet without capsular repair, and modified Latarjet with capsular repair. Results: Creation of a 20% bony Bankart lesion led to significant increases in anterior and inferior glenohumeral translation and rotational range of motion (p < 0.005). The Latarjet procedure restored anterior and inferior stability compared to the bony Bankart condition. It also led to significant increases in glenohumeral internal and external rotational range of motion relative to both the intact and bony Bankart conditions (p < 0.05). The capsular repair from the coracoacromial ligament stump to the native capsule did not significantly affect translations relative to the Latarjet condition; however it did cause a significant decrease in external rotation in both the scapular and coronal planes (p < 0.005). Conclusions: The Latarjet procedure is effective in restoring anteroinferior glenohumeral stability. The addition of a capsular repair does not result in significant added stability; however, it does appear to have the effect of restricting glenohumeral external rotational range of motion relative to the Latarjet procedure performed without capsular repair.

<P>Angiotensin-converting enzyme 2 (ACE2) is a monocarboxypeptidase in the renin-angiotensin system that catalyzes the breakdown of angiotensin II to angiotensin 1-7. We have reported that ACE2 expression in the kidney is reduced in experimental Alport syndrome but the impact of this finding on disease progression has not been studied. Accordingly, we evaluated effects of murine recombinant ACE2 treatment in Col4a3 knockout mice, a model of Alport syndrome characterized by proteinuria and progressive renal injury. Murine recombinant ACE2 (0.5 mg/kg/day) was administered from four to seven weeks of age via osmotic mini-pump. Pathological changes were attenuated by murine recombinant ACE2 treatment which ameliorated kidney fibrosis as shown by decreased expression of COL1 alpha 1 mRNA, less accumulation of extracellular matrix proteins, and inhibition of transforming growth factor-beta signaling. Further, increases in proinflammatory cytokine expression, macrophage infiltration, inflammatory signaling pathway activation, and heme oxygenase-1 levels in Col4a3 knockout mice were also reduced by murine recombinant ACE2 treatment. Lastly, murine recombinant ACE2 influenced the turnover of renal ACE2, as it suppressed the expression of tumor necrosis factor-alpha converting enzyme, a negative regulator of ACE2. Thus, treatment with exogenous ACE2 alters angiotensin peptide metabolism in the kidneys of Co14a3 knockout mice and attenuates the progression of Alport syndrome nephropathy.</P>

<P>Biomass burning (BB) is a large source of reactive compounds in the atmosphere. While the daytime photochemistry of BB emissions has been studied in some detail, there has been little focus on nighttime reactions despite the potential for substantial oxidative and heterogeneous chemistry. Here, we present the first analysis of nighttime aircraft intercepts of agricultural BB plumes using observations from the NOAA WP-3D aircraft during the 2013 Southeast Nexus (SENEX) campaign. We use these observations in conjunction with detailed chemical box modeling to investigate the formation and fate of oxidants (NO<SUB>3</SUB>, N<SUB>2</SUB>O<SUB>5</SUB>, O<SUB>3</SUB>, and OH) and BB volatile organic compounds (BBVOCs), using emissions representative of agricultural burns (rice straw) and western wildfires (ponderosa pine). Field observations suggest NO<SUB>3</SUB> production was approximately 1 ppbv hr<SUP>-1</SUP>, while NO<SUB>3</SUB> and N<SUB>2</SUB>O<SUB>5</SUB> were at or below 3 pptv, indicating rapid NO<SUB>3</SUB>/N<SUB>2</SUB>O<SUB>5</SUB> reactivity. Model analysis shows that >99% of NO<SUB>3</SUB>/N<SUB>2</SUB>O<SUB>5</SUB> loss is due to BBVOC + NO<SUB>3</SUB> reactions rather than aerosol uptake of N<SUB>2</SUB>O<SUB>5</SUB>. Nighttime BBVOC oxidation for rice straw and ponderosa pine fires is dominated by NO<SUB>3</SUB> (72, 53%, respectively) but O<SUB>3</SUB> oxidation is significant (25, 43%), leading to roughly 55% overnight depletion of the most reactive BBVOCs and NO<SUB>2</SUB>.</P> [FIG OMISSION]</BR>
