<P>The petroleum ether and chloroform extracts of the root of Piper chaba showed antimicrobial, antileishmanial and cytotoxic activities. Further bioactivity-guided fractionation led to the isolation of Bornyl piperate (1), piperlonguminine (2) and piperine (3). This is the first report of isolation of compounds (1) and (2) from P. chaba. It was observed that the isolated compounds (1 and 2) showed potent antifungal activity when compared with standard drug Nystatin, and significant cytotoxic activity with the IC?????? values of 0.76 and 0.83???µg???mL???, respectively. These compounds were also found to have weak antibacterial and antileishmanial activities. This is the first report about the antileishmanial activity of Piper isolates.</P>

Total phenolic contents of different fractions of the Psidium cattleianum leaf extract and their antioxidant capacity against several free radicals were examined. Protective effect of the ethyl acetate fraction (EAF) on H2O2-induced DNA damage in HepG2 cells were also evaluated, and the phytochemical profile of EAF was analyzed using tandem mass spectrometry. EAF derived from the 80% methanol extract of the leaf contained a remarkable amount of polyphenol and showed high levels of DPPH and alkyl radical scavenging activity, promoted cell viability, and protected against H2O2-induced DNA damage in HepG2 cells. Phytochemical analysis revealed that the major components in the EAF included quercetin monoglycoside, phloridizin, quercetin 3-diglycoside,quercetin-3-glucuronide, 2,6-dihydroxy-3,5-dimethyl-4-glucopyranosylbenzophenone,phenolic acid, guaijaverin, and naringin. The present study suggests possible synergistic or competitive antioxidant action of the major compounds of cattley guava leaf on H2O2-induced DNA damage in HepG2 cells. These results indicate that the ethyl acetate fraction of the guava leaf could be used as a potential source of natural antioxidants, and these findings will facilitate the utilization of guava leaf as a source of functional food.

Purification, characterization and biochemical properties of α-amylase from post harvest Bangladeshi Potato (Solanum tuberosum L.) were investigated. The α-amylase was purified by successive chromatography on DEAE and CM-cellulose columns with a yield of 24.24%. SDS-PAGE showed a molecular weight of 44 kDa for the enzyme that contain 2.8% sugar. The enzyme lost total activity in the presence of the chelating agent EDTA, confirming it was an α-type amylase. The enzyme displayed optimum activity at pH 7.2 and 37˚C, with an apparent Km value of 0.26% using starch as its substrate. The enzyme was strongly inhibited by Cu2+, Fe2+ and Zn2+; moderately by Li+, Hg+ and Cd2+; and slightly by Ag+, K+, Mn2+ and Mg2+. Conversely, Fe3+ and Na+ appreciably enhanced activity, while adding calcium ion nearly doubled enzyme activity. In addition, the activity of α-amylase gradually decreased with increasing concentrations of urea. Thus, potato α-amylase is an attractive target for study to better understand the structure-function relationships of α-amylases.

Flesh and peel extracts of five mango varieties were compared for their antioxidant and antiproliferative activities. Fozli peel (FP) extract was found to have the highest antioxidant activity and to significantly inhibit the proliferation of HeLa human cervical carcinoma cells in a dose-dependent manner. Hoechst 33342 staining revealed apoptotic nuclei with condensed chromatin, and cell growth arrest was confirmed by flow cytometry. Apoptotic signaling induced by FP extract was characterized by an increased Bax/Bcl-2 ratio and down-regulation of Bcl-2. Moreover, FP extract treatment triggered the proteolytic activation of caspases-3, -8, and -9 and the degradation of poly ADP-ribose polymerase in HeLa cells. These results indicate that FP is an excellent source of phenolic and/or flavonoid compounds, and may have applications in the treatment of human cervical carcinoma and as an antioxidant in the food and pharmaceutical industries. Gas chromatography-mass spectrometry analysis of FP extract revealed that 2,5-dihydroxyphenol was detected with highest amount and other compounds such as 5-hydroxymethyl-2-furancarboxaldehyde, pentadecanoic acid, (Z,Z)-9,12-octadecadienoic acid, and 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one were also found in high amounts.

The crude ethanol extracts (stem and fruits), their fractions and two triterpenes, β-Amyrin and 12-Oleanene 3β, 21β-diol, isolated as a mixture from the chloroform soluble fraction of an ethanolic extract of Duranta repens stem, were evaluated for antibacterial, antifungal activities by the disc diffusion method and cytotoxicity by brine shrimp lethality bioassay. The structures of the two compounds were confirmed by IR, 1H-NMR, 13C-NMR and LC-MS spectral data. The chloroform soluble fraction of stem and ethanol extract of fruits possess potent antishigellosis activity and also exhibited moderate activity against some pathogenic bacteria and fungi but the isolated compound 1 (mixture of β-Amyrin and 12-Oleanene 3β, 21β-diol) showed mild to moderate inhibitory activity to microbial growth. The minimum inhibitory concentrations (MICs) of the extracts (stem and fruits), their fractions and compound 1 were found to be in the range of 32~128 μg/ml. The chloroform soluble fractions of stem and ethanol extract of fruit showed significant cytotoxicity with LC50 value of 0.94 μg/ml and 0.49 μg/ml, respectively against brine shrimp larvae.
