The approximate degree reduction of ball non-uniform rational B-splines (NURBS) curves is a pivotal and knotty technique in computer-aided design/computer-aided manufacture. As we all know, the multi-degree reduction of NURBS ones is a mathematical optimization problem that a swarm intelligence algorithm can deal with. This paper uses an improved orca predation algorithm (IOPA) to accomplish the optimal multi-degree reduction of NURBS curves. Firstly, by incorporating a dimension learning strategy and opposition-based learning strategy into the orca predation algorithm (OPA), an IOPA is developed to increase the population diversity and enhance its capability of jumping out of the local minima. Secondly, the superiority of the proposed IOPA is comprehensively verified by comparing it with the original OPA and numerous celebrated and newly developed algorithms on the IEEE Congress on Evolutionary Computation (CEC) 2014 test suite and IEEE CEC2017 benchmark functions, respectively. Meanwhile, the practicability of IOPA is also highlighted by solving three real-world engineering design problems. Furthermore, statistical testing of IOPA has been conducted to validate its significance. Finally, the optimization model of multi-degree reduction for NURBS curves is established by minimizing the distance between the original curve and the approximate curve. The IOPA is utilized to solve the optimization model, and the optimal approximate NURBS curves are obtained. Some representative numerical examples illustrate the ability of the proposed IOPA to effectively solve the multi-degree reduction problem of NURBS curves in terms of precision, robustness, and convergence characteristics.
Pressure measurements on inclined square prisms

This study investigated aerodynamic characteristics of an inclined square prism experimentally. Pressure measurements were performed on a static square prism with a series of inclinations including forward inclinations (inclined to the upwind direction) and backward inclinations (inclined to the downwind direction). The prism with a vertical attitude was also tested for comparisons. Based on the pressure data, influences of the inclinations on aerodynamic characteristics (e.g., force coefficients, pressure distributions on the surfaces, and vortex shedding features) of the square prism were evaluated in detail. The results show that the inclinations have significant effects on these aerodynamic characteristics. Furthermore, the influences of the forward and backward inclinations are quite different.

Excessive activation of the NLRP3 inflammasome is a key component contributing to the pathogenesis of various inflammatory diseases. However, the molecular mechanisms underlying its activation and regulation remain poorly defined. The objective of this study was to explore the possible function of the K+ channel pore-forming subunit Kir6.1 in regulating NLRP3 inflammasome activation and insulin resistance. Here, we demonstrate that Kir6.1 depletion markedly activates the NLRP3 inflammasome, whereas enhanced Kir6.1 expression produces opposing effects both in mice in vivo and in primary cells in vitro. We also demonstrate that Kir6.1 controls insulin resistance by inhibiting NLRP3 inflammasome activation in mice. We further show that Kir6.1 physically associates with NLRP3 and thus inhibits the interactions between the NLRP3 inflammasome subunits. Our results reveal a previously unrecognized function of Kir6.1 as a negative regulator of the NLRP3 inflammasome and insulin resistance, which is mediated by virtue of its ability to inhibit NLRP3 inflammasome assembly. These data provide novel insights into the regulatory mechanism of NLRP3 inflammasome activation and suggest that Kir6.1 is a promising therapeutic target for inflammasome-mediated inflammatory diseases.

The association between the NAD(P)H:quinone oxidoreductase 1 (NQO1) gene C609T polymorphism (rs1800566) and gastric cancer has been widely evaluated, but a definitive answer is so far lacking. We first conducted a case-control study to assess this association in a large Han Chinese population, and then performed a meta-analysis to further address this issue. Although our case-control association study indicated no significant difference in the genotype and allele distributions of C609T polymorphism between gastric cancer patients and controls, in the meta analysis involving 4,000 subjects, comparison of alleles 609T and 609C indicated a significantly increased risk (46%) for gastric cancer (95% confidence interval (95%CI) for odds ratio (OR)=1.20-1.79) in individuals with the T allele. The tendency was similar to the homozygote (OR=1.81, 95%CI: 1.16-2.84), dominant models (OR=1.41, 95%CI: 1.12-1.79), as well as recessive model (OR=1.58, 95%CI: 1.06-2.35). Stratified analysis by study design demonstrated stronger associations in population-based than in hospital-based studies. And ethnicity-based analysis demonstrated a significant association in Asians. We conclude that the NQO1 gene C609T polymorphism increases the risk for gastric cancer, especially in Asian populations.

Molecular Cloning and Characterization of a Lipocalin in the Bumblebee Bombus Ignitus

We have cloned and characterized a lipocalin from the bumblebee Bombus ignitus (Bi-lipocalin). The Bi-lipocalin gene spans 2284 bp and consists of four exons coding for 270 amino acid residues. Sequence analysis revealed that Bi-lipocalin possesses three structurally conserved regions (SCTs) that characterize lipocalins. Recombinant Bi-lipocalin, expressed as a 37 kDa protein in baculovirus-infected insect cells, was N-glycosylated, indicating that the carbohydrate moieties are necessary for secretion. Tissue distribution analysis revealed ubiquitous expression of Bi-lipocalin in all tissues examined. Bi-lipocalin transcripts were upregulated by stress, such as wounding, H2O2 exposure, and external temperature shock. These results indicate that Bi-lipocalin is a stress-inducible protein that acts on wounding, H2O2 overexposure and temperature stimulation.

A muscle-specific lipase gene of the bumblebee Bombus ignitus was cloned and characterized. This gene, which we named Bi-Lipase, consists of seven exons encoding 317 amino acid residues. Bi-Lipase possesses all the features of lipases, including GXSXG consensus motif and Ser-Asp-His catalytic triad. Expressed as a 37-kDa polypeptide in baculovirus-infected insect Sf9 cells, recombinant Bi-Lipase showed an optimal pH of 9.0 and exhibited its highest catalytic activity at 40℃. Furthermore, through the addition of tunicamycin to the recombinant virus-infected Sf9 cells, recombinant Bi-Lipase was found to be N-glycosylated. Northern and western blot analyses indicated that Bi-Lipase was expressed in the wing, thorax, and leg muscles. These results show that Bi-Lipase is a muscle-specific lipase, suggesting a possible role of Bi-Lipase in the utilization of lipids for muscular activity in B. ignitus.
The effects of LNG-tank sloshing on the global motions of FLNG system

This paper addresses a study of inner-tank sloshing effect on motion responses of a Floating Liquefied Natural Gas (FLNG) system, through experimental analysis and numerical modeling. To investigate hydrodynamic characteristics of FLNG under the conditions of with and without LNG-tank sloshing, a series of numerical simulations were carried out using potential flow solver SESAM. To validate the numerical simulations, model tests on the FLNG system was conducted in both liquid and solid ballast conditions with 75% tank filling level in height. Good correlations were observed between the measured and predicted results, proving the feasibility of the numerical modeling technique. On the verified numerical model, Response Amplitude Operators (RAOs) of the FLNG with 25% and 50% tank filling levels were calculated in six degrees of freedom. The influence of tank sloshing with varying tank filling levels on the RAOs has been presented and analyzed. The results showed that LNG-tank sloshing has a noticeable impact on the roll motion response of the FLNG and a moderate tank filling level is less helpful in reducing the roll motion response.
