Breast adenomyoepitheliomas are composed of a biphasic proliferation of myoepithelial cells around small epithelial-lined spaces. Due to the rarity of adenomyoepitheliomas, the molecular data describing them are limited. Adenomyoepitheliomas are considered to be benign or have low malignant potential, and be prone to local recurrence. Malignant transformation has been associated with homozygous deletion of CDKN2A or somatic mutations in TERT, but remains unexplained in many cases. Here, we describe a case of carcinomatous transformation of both epithelial and myoepithelial cells in an estrogen receptor-negative adenomyoepithelioma caused by amplification of MYC. Break-apart fluorescence in situ hybridization revealed an increase in the MYC gene copy number (3–4 copies/cell in 37%, > 4 copies/cell in 40%). Deregulation of MYC is responsible for uncontrolled proliferation and cellular immortalization in basal-like breast cancers. Our case demonstrates that genomic instability events associated with gene amplification may be involved in the carcinogenesis of malignant adenomyoepitheliomas.

Introduction: Diabetes mellitus is a major global health problem. Needling at CV-12 has reduced blood glucose level in diabetic rats. The aim of this study was to evaluate the effect of needling at CV-12 (Zhongwan) on blood glucose level in patients with type 2 diabetes mellitus (T2DM). Materials and Methods: Forty T2DM patients were recruited and randomized into either the acupuncture group or placebo control group. The participants in the acupuncture group were needled at CV-12 (4 cun above the center of the umbilicus), and those in the placebo control group were needled at a placebo point on the right side of the abdomen (1 cun beside the CV- 12). For both groups, the needle was retained for 30 minutes. Assessments were performed prior to and after the intervention. Statistical analysis was performed using SPSS version 16. Results: There was a significant reduction in random blood glucose level in the acupuncture group compared to baseline. No such significant change was observed in the placebo control group. Conclusion: The result of this study suggests that 30 minutes of needling at CV-12 might be useful in reducing blood glucose level in patients with T2DM.

Introduction: Acupuncture, a key part of traditional Chinese medicine, is used to relieve symptoms of diabetes mellitus. The aim of this study was to evaluate the effect of needling CV-12 (Zhongwan) on blood glucose levels in healthy volunteers. Materials and methods: Thirty-six individuals were recruited and randomized into either the acupuncture group or the placebo control group. The participants in the acupuncture group were needled at CV-12 (4 cun above the center of the umbilicus), and those in the placebo control group were needled at a nonexisting “sham” point on the right side of the abdomen (1 cun beside the CV-12)da nonacupuncture point. For both groups, the needle was retained for 20 minutes without stimulation. Assessments were performed prior to and after the intervention. Statistical analysis was performed using the Statistical Package for the Social Sciences, version 16. Results: The result of this study showed a mild reduction in random blood glucose (RBG) levels in the acupuncture group and a mild increase in RBG levels in the placebo control group. However, these changes were not statistically significant both within and between groups.

Purpose The prognosis of patients with hepatocellular carcinoma (HCC) undergoing transarterial chemoembolization (TACE) is often uncertain. We aimed to utilize analytic morphomics, a high-throughput imaging analysis, to assess if body composition is predictive of post-TACE survival. Materials and Methods We included patients from a single center (Ann Arbor VA) who had TACE as the primary treatment for HCC and had a pre-treatment computed tomography scans. Univariate analysis and multivariate conditional inference tree analysis were utilized to identify the morphomic characteristics predictive of 1-year survival. Results were validated in an external cohort (University of Michigan Health System) of HCC patients who underwent TACE as their primary treatment. Results In the 75 patients in the derivation cohort, median survival was 439 (interquartile range, 377 to 685) days from receipt of TACE, with 1-year survival of 61%. Visceral fat density (VFD) was the only morphomic factor predictive of overall and 1-year survival (p < 0.001). Patients with VFD above the 56th percentile had a 1-year survival of 39% versus 78% for those below the 56th percentile. VFD also correlated with 1-year survival in the external validation cohort (44% vs. 72%, p < 0.001). In a secondary analysis, patients with higher VFD were significantly more likely to experience hepatic decompensation after TACE (p < 0.001). Conclusion VFD served as an objective predictor of mortality in patients undergoing TACE, possibly through its ability to predict hepatic decompensation. VFD may serve as a radiographic biomarker in predicting TACE outcomes.

<P>Ascorbic acid (AsA) is an inevitable antioxidant found abundantly in higher plants. Despite the importance of AsA in plants, how AsA biosynthesis (ABGs; D-mannose/L-galactose pathway) and AsA recycling genes (ARGs) evolved through polyploidization has not been addressed to date. Here, we evaluated the impacts of whole genome triplication (WGT) on ABGs and ARGs in Chinese cabbage (Brassica rapa ssp. pekinensis), which diverged from Arabidopsis thaliana before the WGT event. Twentythree ABGs coded in 13 loci representing nine different enzyme classes and 29 ARGs coded in 19 loci representing five different enzyme classes were identified in the B. rapa genome by whole-genome screening through comparative genomic analyses. Five of these loci maintained three gene copies, 10 loci maintained two gene copies and the majority of the loci (n = 17) maintained single gene copies. Segmental (62 %) and tandem duplication (6 %), and fragment (21 %) and large-scale recombination (10 %) events accelerated the diversification of ABGs and ARGs. Thirteen of the 52 (25 %) identified genes experienced intron losses and two (4 %) experienced intron gains implying that intron losses outnumbered intron gains. The expansion and the retention of ABGs and ARGs were similar to the whole genome gene expansion and retention (P>0.05). These findings provide new insights into the structural characteristics and evolutionary trends of ABGs and ARGs. In addition, our data could become a useful resource to further the functional characterization of these genes.</P>

<P>Cumin seed was investigated for synthesis of gold nanoparticles. Polydispersed particles were obtained at pH 3 and 30 degrees C, and the effect of temperature and pH on synthesis of gold nanoparticles was analyzed. TEM images showed that amount of platelets formed were predominant at lower temperature where as more number of monodispersed spherical particle of size 1-10 nm were perceived at high temperatures. The gold nanoparticles particles formed at higher pH were stable, uniform and spherical in shape. XPS analysis showed the presence of pure gold nanoparticles.</P>

Serine/arginine-rich (SR) gene family memberscan diversify the transcriptome and proteome of eukaryotesby facilitating the alternative splicing (AS) of precursormessenger RNAs. Herein, we investigated the evolutionarydynamics, AS patterns, and expression levels of the Brassicarapa SR (BrSR) gene family in young seedlings treated withabiotic stresses. A comparative genomic analysis identified25 BrSR genes at 18 Arabidopsis loci and three BrSR-likegenes at two Arabidopsis loci. Thirteen of these loci weresingletons, while seven loci carried paralogs. All the duplicatedpairs were determined to be under purifying selection pressure. The expansion of the BrSR gene family was found to be theresult of segmental duplications only. Additionally, theexpression levels of 78.6% (22 of 28) and the AS patterns of60.7% (17 of 28) of the BrSR genes were altered in responseto abiotic stresses. Among the analyzed abiotic stresses,oxidative, cold, and heat treatments induced the largestexpression changes, while cold and heat stresses caused mostAS events. Our findings provide insights into the evolutionarydynamics of BrSR genes following polyploidization events,and provide an important resource for future studies aimed atcharacterizing the specific function(s) of BrSR genes in plantgrowth, development, and defense.

In this paper, we have investigated the impact of the un-doped and recessed gate structure on the performance of the 6 H-SiC Metal Semiconductor Field Effect Transistor. The performance of the 6 H-SiC MESFET analyzed using TCAD simulations. The proposed un-doped gate structure made of high-k dielectric materials and low-k dielectric materials being hafnium oxide (HfO2) and silicon dioxide (SiO2) and it has minimized ionized impurity scattering, leading to increased electron mobility and improved carrier concentration. One of substrate layer of this device grown on Silicon (Si) and β-gallium oxide (β-Ga2O3). Performance metrics such as drain current, transconductance, subthreshold slope, and cutoff frequency are evaluated and compared with conventional SiC MESFET structures. The proposed device exhibits superior current driving capabilities, enhanced transconductance, and reduced leakage currents, leading to improved power efficiency. Moreover, the recessed gate structure contributes to a significant reduction in short channel effects, making the device more suitable for low power applications. The simulation parameters are calculated and compared with conventional MESFET structure with the length of source and drain in submicron technology. Therefore the drain current of this proposed device is improved 68%.
