        실험연구 : 열 (熱) Stress 및 유영 (游泳) Stress에 대한 귀비탕과 (歸脾湯) Ascorbic Acid의 항 (抗) Stress효능 비교연구

        In order to compare the anti-stress effect of Guibitang with that of ascorbic acid, after these medicines were administered to guinea pigs induced by heating and swimming stress, the changes of the weight of body and organ, and content of plasma catecholamines, serum total cholesterol, free cholesterol, triglyceride, protein, glucose and cortisol were measured. The results were as follows : 1. The weight of the body was increased with statistical significance in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on both heating and swimming stress. 2. The weight of spleen decreased with statistical significance in the groups administered ascorbic acid, Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress, but in case of swimming stress, the weight of spleen decreased with statistical significance in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. The weight of adrenal decreased with statistical significance in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress alone. 3. Plasma norepinephrine content decreased with statistical significance not only in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid but in the group administered ascorbic acid with Guibitang as compared with in the group administered ascorbic acid on heating stress. In case of swimming stress, norepinephrine decreased with statistical significance in the groups administered ascorbic acid, Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. 4. Plasma dopamine content decreased with statistical significance only in the group administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on both heating and swimming stress. 5. Serum total cholesterol content decreased with statistical significance in the groups administered ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress, but in case of swimming stress, it decreased with statistiscal significance only in the group administered ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. 6. Serum triglyceride content decreased with statistical significance not only in the groups administered ascorbic acid, Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid but in the group administered ascorbic acid with Guibitang as compared with in the group adminstered ascorbic acid on heating stress. In case of swimming stress, triglyceride decrease with statistical significance only in the group administered ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. 7. Serum glucose content increased with statistical significance in the groups administered Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on both heating and swimming stress, particulaly in case of swimming stress, in the group administered ascorbic acid with Guibitang, it increased with statistical significance as compared with in the group administered ascorbic acid. 8. Serum cortisol content decreased with statistical significance only in the group administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress.

        사향소합원이 (麝香蘇合元) 흰쥐의 방사형 미로 학습과 기억에 미치는 영향

        Purpose: This study was conducted to find out the effects od Sahyangsohapwon on learning and memory of rats. Method: In the experiment, rats were divided two groups. One was control group which was adminstered Sahyangsohapwon and the other was sample group administered placebo. Numbers of each group were 13 rats. 8-arm radial maze task was used in it, and working memory test and retention(reference memory) test were done. Before the beginning of the test, the rats were deprived of water for 24hrs. In the frist test, each of eight arm was baited with water and a rat was permitted to remain on the maze until all eight arms were entered. A working memory error was defined as revisit of any previously entered arm. When a rat made an error not exceeding one time in consecutive 3 days-performance, it was regarded as learning criteria and the test was ended. The reference memory was evaluated with total days which it took rats to pass the learning crtirtia. The second test was performed after 24 hours when the first test was over. When a rat entered 4 arms, the entrance of arm was cut off during 30 seconds. Here the number if errors which was produced during a rat find remaining 4 tracks was regarded as the index of memory. This experiment compared the number of error at the control group with that of the sample group. Result: 1. In the first test, it was shown that the sample group took 7.69±1.11 days and the control group 9.31±1.97 days to pass the learning criteria. There was statistically significant reference mernory development at the sample group. 2. In the second test, the frequency of errors made by the two groups were 0.92±1.32 times for the control group and 1.23±1.59 times for the Sahyangsohapwon group. There was no difference between the groups in frequency of errors. Conclusion: It is suggested that Sahyangsohapwon has effects on the improvement of learning and memory.

        실험연구 : 귀비온담탕이 (歸脾溫膽湯) 흰쥐의 항스트레스와 면역반응에 (免疫反應) 미치는 영향

        In order to investigate the Anti-stress effect of Guibiondamtang in the immobilization stressed rats, the level of serum catecholamine, the change of body weight, the humoral and cellular immune response were studied. The results were as follows; 1. The decrese of the body weight was significantly inhibited in test group for Guibiondamtang, comparting to the control group. 2. The increase of the level of serum norepinephrine was significantly inhibited in test group, comparing to the control group. 3. The increase of the level of serum epinephrine was significantly inhibited in test group, comparing to the control group. 4. In the hemagglutinaton titer, the control group was decreased on the serum antibody titer but test group was inhibitory effect on the decrease of sereum antibody titer. 5. In the plaque formation test, the control and test group were not shown significant differences.

        치매에 대한 한의학적 임상연구

        1. Out of 21 examples on a clinical base Alzheimer type dementia were 8 examples(38%), 11 vascular types(52%), 1 Alzheimer and vascular mixed type and rest 1 was secondary dementia type(NPH). 2. Between the gendes, there were 5 males and 3 females for Alzheimer types, 6 males and 5 females for the vascular types, 1 male for Alzhrimer and vascular mixed type, and 1 frmale for secondary dementia type(NPH). 3. For the degrees of Alzheimer type dementia there were 2 mild dementia, 4 moderate dementia, and 2 severe dementia. 4. Among the Alzheimer types 2 mild dementia were almost recovered back to normal in approximately 2 months, 2of 4 moderate dementia became significantly better in approximately 2 months and recovered to almost normal state in 3 to 5 months. The rest 2 have been under treatment for 3 months, but showed a little improvement. Out of 2 severe dementia examples, one showed a little improvement even if it has been under treatment over 2 years. The other exampe did not show any improvement, but dementia did not proceed any more. 5. Among the 11 vascular examples, 7 recovered in 2 to 3 months, 1 in 5 months, 2 recovered in a year, and the treatment was stopped arbitrary for the last one. 6 In the case of Alzheimer and vascular mixed type dementia, even the moderate dementia did not show any apparent result in 6 months. 7. For NPHI (Normal Pressure Hydrocephalus), there was an improvement on dementia in 2 months after the treatment, but gait disturbance and urinary incontinence did not show any noticeable difference. As a conclusion, almost all the Alzheimer and vascular type patients recovered, but the treatment periods varied depending on the types of the dementia and the degree of seriousness, especially in the case of the severe Alzheimer patients, the treatment oniy suppression the progression of the dementia. The most important aspect from clinical point of view was even if almost all the patients were almost completely cired, they need to keep being cured for a long period of time. The methol with respect to Four Constitutions is thought of the most desirable.

        실험연구 : 귀비탕과 (歸脾湯) Ascorbic Acid가 열 (熱) 및 유영 (遊泳) Stress Guinea Pig의 뇌 (腦) Catecholamine 함량에 미치는 영향

        This study aimed to compare the anti-stress effect of Guibitang with Ascorbic Acid on the guinea pigs in heat or forced swimming stress. the experimental animals were heated in hot room(34±2℃) for 2 hours in a day during 7 days. Forced swimming stress was loaded by forcing guinea pigs to swim in a oval tub for 2 minutes in a day during 7days. And administered 1.0㎎/100g of Ascorbic Acid or 86.0㎎/100g of Guibitang extract formonce befor stress. 1. In brain, the contents of norepinephrine in group of the heat of forced swimming stress, in case administered Ascorbic Acid and in case administered Guibitang extract with Ascorbic Acid significantly decreases as compared with these in case administered non Ascoric Acid. 2. In brain, the contents of epinephrine in group of the forced swimming stress, in case administered Guibitang extract with Ascorbic Acid significantly decreases as compared with these in case administered non Ascorbic Acid. 3. In brain, the contents of dopamine in group of the heating stress, in case administered Guibitang extract with Ascorbic Acid significantly decreases as compared with these in case administered non Ascorbic Acid, and these in group of the forced swimming stress in case administered Ascorbic Acid and in case administered Guibitang extract without Ascorbic Acid and in case administered Guibitang extract with Ascorbic Acid significantly decreases as compared with these in case administered non Ascorbic Acid.

      • 청화보심탕(淸火補心湯)이 우울증(憂鬱症) 막형동물(模型動物)의 절망행동(絶望行動), 불안(不安) 및 뇌(腦)의 TH 와 c-Fos 발현(發展)에 미치는 효과(效果)

        Objective : This study was designed to assess the protective effects of Chengwhabosimtang on the animal model of depression, chronic mild stress(CMS). Method : Male Sprague-Dawley rats Were used for this experiment. The subjects Were divided into 3 groups (1. CMS-drug: Chengwhabosimtang administered during CMS treatment, 2. CMS-vehicle: water administered, 3. normal ). After 4 weeks of CMS treatment they were executed Forced swimming test(FST) and Elevated plus maze. Tyrosine hydroxylase(TH) in ventral tegmental area(VTA) and c-Fos in paraventricular nucleus(PVN) were measured. Result : 1. In FST, CMS-drug group showed significantly decreased immobility behavior. 2. CMS-drug group showed no significantly lower TH level in VTA than CMS-vehicle group. 3. CMS-drug group showed significantly less c-Fos expressed cell bodies in PVN than CMS-vehicle group. 4. In Elevated plus maze, CMS-drug group showed no significantly anxiety. Conclusion : These results suggest that Chengwhabosimtang may have protective antidepressant effects in CMS model rats. And these effects could be explained by the elevated stress-copying behaviors which are related with PVN of hypothalamus and dopaminergic neurons in VTA.
