4주간의 고지혈 유도식이를 섭취시킨 후 고지혈증 유도가 이루어진 실험 동물을 이용하여 생식 섭취로 인한 고지혈증개선 효과를 확인하고자 하였다. 고지혈 유도가 확인된 실험 동물에게 고지혈사료에 30% 생식을 혼합한 식이를 투여하였을 경우 혈중 콜레스테롤량을 약 37% 정도 감소시킨 것으로 나타났으며, 간조직중의 콜레스테롤 역시 약 28% 감소한 것으로 나타나 생식의 섭취가 체내의 콜레스테롤 농도를 낮추어 주는 효과가 있는 것으로 확인되었다. 또한, 체내 콜레스테롤 구성에 있어서도 관상동맥 질환을 억제하는 것으로 알려진 HDL-C의 함량은 대조군에 대비하여 생식 섭취군이 약 28%정도 증가되었으며, 동맥벽에 축적되어 관상동맥질환 발병과 연관관계가 높은 것으로 알려진 LDL-C의 함량을 약 39%정도 감소시킨 것으로 나타나 체내 콜레스테롤 수치의 저하와 더불어 콜레스테롤의 구성 양상에도 이로운 점을 제공한 것으로 나타났다. 이러한 결과로부터 순환기계 질환 발병율을 나타내는 지표로 널리 이용되는 동맥경화지수와 심혈관위험지수를 산출한 결과 생식 섭취군은 대조군에 대비하여 동맥경화지수의 약 60%를 저하시켰으며, 심혈관위험지수 역시 약 56% 정도 저하시킨 것으로 나타났다. 이상의 결과를 종합하여 볼 때 생식 섭취는 체내 콜레스테롤의 농도를 저하시킴으로서 순환기계 질환의 위험도를 낮추는데 효과가 있는 것으로 생각된다. The study was conducted to investigate the effect of Saengshik supplementation on lipid metabolism in rats fed high cholesterol diets. Male Spraque-Dawley rats were administrated 1% cholesterol to induce hypercholes-terolemia and were fed on diet containing Saengshik (30%, w/w). Serum and liver lipid profiles and fecal bile acids excretion were examined after 7 weeks of experimental diet. The feeding of diet containing 30% Saengshik significantly decreased total cholesterol (TC) contents in liver and plasma. Since cholesterol was balanced by entero-hepatic circulation through bile acid synthesis and reabsorption, inhibition of bile acid re-uptake in intestine decreases total cholesterol in liver and blood. In this point, significant increament of fecal bile acid excretion in Shaengshik group decreased TC and improved hypercholesterolemia. Also, plasma high density lipoprotein-cholesterol (HDL-C) decreasing risk for coronary heart disease in Saengshik supplemented group was significantly higher than control gorup, whereas low-density lipoprotein-cholesterol (LDL-C) accumulation in arterial vessel wall was significantly lower than control group. In this result, we supposed that supplementation of Saengshik, uncooked natural food, may improve hypercholesterolemia through increment of cholesterol excretion.
산야초 추출물을 함유한 식사 대용식을 이용한 초저열량 식사요법이 성인 비만 여성들의 체중감량과 건강 개선에 미치는 영향

본 연구는 산야초 추출물이 함유된 식사대용식을 이용한 초저열량 식사요법의 체중감량과 건강개선에 미치는 효과를 평가하고자 실시되었다. 그 결과 식사대용식을 이용하여 초저열량 식사요법을 한 성인 비만여성들의 경우 일반 음식으로만 초저열량 식사요법을 한 성인 비만여성들보다 체중과 체지방율이 유의하게 감소하였음을 알 수 있었다. 건강관련 인자는 두 군간에 유의한 차이는 없었으나 산야초 추출물이 함유된 식사대용식을 섭취한 군내에서 식사중재 기간 동안 유의하게 지질 개선 효과가 있었다. 한편, 산야초 추출물이 함유된 식사대용식의 맛에 대한 기호도도 긍정적으로 평가되었다. 그러므로, 본 연구에서 사용된 산야초 추출물을 주성분으로 하여 제조된 식사대용식은 향후 임상에서 비만치료제로 응용함에 있어 크게 도움이 될 것으로 사료된다. 단, 앞으로 요요 현상의 가능성에 대한 보다 장기간에 걸친 임상연구가 이루어져야 할 것으로 사료된다. This study was performed to investigate the effects of very low calorie diet (VLCD) using meal replacements that contain the wild grass extracts that are consisted of mainly Samul-tang ingredients on weight reduction and health promotion in the obese women (BMI≥25㎏/㎡) for four weeks. Fifty six premenopausal women participated in this experiment. Each subject was either classified as part of the control group (C group, n=28) that consumed 3 regular meals a day or the meal replacement group (MR group, n=28) that consumed 1 regular meal and 2 meal replacements within 600 kcal/day. Anthropometric measurements, body composition, health related factors, and psychological factors were assessed before (the initial) and after (the 4^(th) week) the study. Obesity was significantly decreased after the diet intervention. Anthropometric measurements such as weight, waist and hip circumference, and BMI and body composition such as body fat, and fat mass were significantly decreased in all groups after the diet intervention. Anthropometric measurements and body composition of the MR group were significantly decreased more than those of the C group. Serum total cholesterol was significantly decreased in the MR groups but not in the C group and serum triglyceride concentration was significantly decreased in all groups after the diet intervention. All the biochemical measurements were within normal ranges after the diet intervention. Psychological factors such as stress and depression were significantly decreased in two groups after the diet intervention. Therefore, VLCD using meal replacements is very effective for weight reduction in the obese premenopausal women.
식사대용식을 이용한 초저열량 식사요법이 20대 비만 여성들의 심리적 요인과 삶의 질에 미치는 효과

This study was performed to investigate the effects of very low calorie diet (VLCD) using meal replacements that contain the wild grass extracts based on Samul-tang ingredients on psychological factors and quality of life in the obese women (BMI ≥ 25 ㎏/㎡) for four weeks. Seventy five women (20≤ age < 26) participated in this experiment. Subjects were randomly classified three groups: I) General diet group (GD group, n = 27) consumed 3 regular meals within 600 ㎉/day 2) Meal replacements group (MR group, n = 27) consumed 1 regular meal and 2 meal replacements within 600 ㎉/day 3) Herbal Meal replacements group (HMR group, n = 27) consumed 1 regular meal and 2 meal replacements within 600 kcal/day. Physical factors (weight, BMI, fat (%)) of the HMR group significantly decreased more than those of GD and MR groups. Moreover, binge eating habit and environmental factors (surrounding support, emotional reaction, expression of opinion) of the HMR group significantly decreased more than those of GD and MR groups. Psychological factor and quality of life were no significant differences among three groups during the experimental period, because both were significantly decreased in all groups after 4 weeks. Therefore, very low calorie diet using meal replacements that contain the wild grass extracts based on Samul-tang ingredients for 4 weeks was effective on improvement of psychological factor and quality of life as well as weight reduction in the obese preme-nopausal women.
허혈/재관류 환경하에서 청폐사간탕의 염증 관련 반응 억제 효과

Chungpesagan-Tang (CST), a Korean traditional prescription composed of oriental medical herbs, has been used successfully to improve human health and regimen. This study was designed to examine the inhibitory effects of CST against in vitro ischemia/reperfusion-induced inflammatory response. We have characterized the production of prostaglandin E₂and arachidonic acid during ischemia/reperfusion in the human neuroblastoma SK-N-MC and human monocytic macrophage U937 cells and the inhibitory effect of CST on these inflammation-related substance formation has been found out in this study. This result suggested that CST used in this experiment reinforced formation has been found out in this study. This result suggested that CST used in this experiment reinforced antiinflammatory potentials and protected cells against ischemia/reperfusion-induced inflammatory response.
한약내의 리튬이 타이로신 가수분해효소에 미치는 영향에 대한 연구

Lithium, a simple monovalent cation, is the mainstay in treatment of manic-depressive illness, but despite extensive research, its mechanism of action remains to be elucidated. Because lithium requires chronic administration for therapeutic efficacy and because its beneficial effects last well beyond its discontinuation, it has been postulated that lithium may exert major effects at the genomic level. We have previously shown that Yungmijihwanwon(YM) increase TH protein expression through antioxidant activity and unknown effects in vivo. In the present study, we have sought to determine if lithium in herbal medicine also increases the expression of endogenous gene known to be regulated by AP-1 and have therefore investigated the effects of herbal medicine on tyrosine hydroxylase(TH) levels. Male mice were treated with LiCI and herbal medicine for 30 days, and TH levels and dopamine level were measured in striatum using immunoblotting and ECD-HPLC. Herbal medicine treatment resulted in statistically nonsignificant increase in TH levels in mouse striatum. The precise therapeutic relevance of these effects is presently unknown.
4-Vessel Occlusion 허혈동물모델에서의 대규모 유전자 발현 연구

Cerebral ischemia, the most prevalent form of clinical stroke, is a medical problem of the first magnitude. Substantial efforts are being made to develop drugs which will protect the brain from the neurodegeneration followed by an ishemic stroke. A key factor in this process is the increasing an evidence that free radical is involved in the mechanisms of ischemic brain damage. We investigated the macro scale gene expression analysis on the global ischemic induced by 4-vessel occlusion in Wister rats. The recent availability of microarrays provides an attractive strategy for elaborating an unbiased molecular profile of large number of genes during ischemic injury. This experimental approach offers the potential to identify molecules or cellular pathways not previously associated with ischemia. ischemia was induced by 4-vessel occlusion for 10 minutes and reperfused again. RNA from sham control brain and time-dependent ischemed brain were hybridized to microarrays containing 4,000 rat genes. 589 genes were found to be at least 2 fold regulated at one or more time points. These survey data provide the foundation studies that should provide convincing proof for ischemia and oxidative stress on gene expression.
