Alnus japonica cortex has been used as antidiarrhea, antihemorrhage and the remedy of indigestion. This study was performed to investigate the effectiveness of the methanol extracts of the Alnus japonica cortex on the gastric lesion and ulcer. The methanol extract was fractionated with hexane, chloroform and butanol, followed by bioassay on antigastritic and antiulcer activity. The methanol extract showed low acute toxicity with minimum lethal dose of more than 5000 ㎎/㎏,p.o. in mice. The chloroform and the butanol fraction reduced gastric lesion in HCl. ethanol induced gastritic model. On gastric secretion in pylorus ligated rat, the hexane and chloroform fraction decreased the volume and acidity. The butanol fraction had significant inhibitory effects on aspirin and Shay`s ulcer. The butanol fraction showed a tendency to inhibit the decrease of mucin secretion due to ingestion of absolute ethanol.

In a preliminary screening of plant extracts for the antigastritic and antiulcer actions in rats, the extracts of head of Panax ginseng Radix showed positive activity in HCl ethanol-induced gastric lesion. Among the systematic fractions of hexane, chloroform, butanol and water, the most potent butanol fraction reduced significantly HCl ethanol-induced gastric lesion at the oral dose of 500 mg/kg. In pylorus ligated rats, hexane and butanol fraction showed decreases in the volume of gastric secretion and acid output, of which effects were stronger in butanol fraction. Further assays with butanol fraction disclosed that it significantly suppressed the aspirin-induced and Shay ulcer. The butanol fraction at the intraduodenal dose of 500 mg/kg showed significant stimulation of mucus secretion.

The effect of Cimicifuga·heracleifolia on CCl₄-induced hepatotoxicity was investigated. Cimicifuga heracleifolia has been used to diaphoresis, antipyretics and detoxification in oriental remedy. We examined the effect of Cimicifuga heracleifolia methanol extract by blood chemical analysis and histopathologic examination. ALT and AST were decreased by 38% and 67% in pretreatment group of Cimicifuga heracleifolia compared to CCl₄ control group respectively. There were significant changes neither in total protein, albumin, triglyceride and cholesterol nor in BUN and creatinine. In histopathologic examination, there were severe necrosis and hemorrhage with infiltration of inflammatory cells around the central vein, zone 3 in the liver of CCl₄ treated rat. Ballooning degeneration of hepatocytes were frequently noted in the periphery of the hemorrhagic necrosis. In Cimicifuga heracleifolia pretreatment group, we observed mild degree of ballooning degeneration and inflammatory cell infiltration. No gross necrosis was observed. We measured malondialdehyde (MDA) formation by TBA method. It showed that the formation of MDA in Cimicifuga heracleifolia pretreatment group was decreased compared to the CCl₄, control group. We got the result of the effect of Cimicifuga heracleifolia on CCl₄-induced hepatotoxicity by decreasing serum ALT and AST. It seems that the decrease of lipid peroxidation is related to the recovery effect.

The rhizoma of Zingiber officinalehas been used as antiemetic, expectorants, stomachache relieving drugs and digestive accelerators. From the observation of antigastritic action of the methanol extract of the rhizoma, it was fractionated with hexane, chloroform, ethyl acetate and butanol, followed by bioassay on antigastritic and antiulcerative activity. The hexane and the chloroform fraction reduced significantly HCl.ethanol induced gastric lesion at the dose of 370 and 210mg/kg, p.o., respectively. On the gastric ulceration and gastric secretion in pylorus-ligated rats, the hexane fraction decreased the volume of gastric secretion and acid output, and also increased pH at the dose of 370mg/kg, i.d.. It showed considerable curative ratio of acetic acid induced ulcer without inhibition of indomethacin induced gastric lesion. The methanol extract showed low acute toxicity with minimum lethal dose of more than 3000mg/kg, p.o. in mice. In conclusion, Zingiberis rhizoma exhibited antigastric and antiulcerative activity which might be attributable to inhibition of gastric secretion. It is revealed that the active component may be present in the hexane fraction.
Calcium Channel Blocker 가 사염화탄소에 의한 간손상에 미치는 영향 및 조직학적 검경

Carbon tetrachloride (CCl₄) induces the hepatotoxicity due to the reactive free radicals generated by cytochrome P-450 (CYP-450) enzyme, which result in destabilization of cellular membrane. Diltiazem, a calcium channel blocking agent, has been known to suppress the CYP-450 enzyme activities. To study the effect of diltiazem in CCl₄-treated rats, we measured the activities of alanine aminotransferase (ALTS, aspartate aminotransferase (AST) and glutathione S-transferase (GST), contents of bilirubin, albumin, total protein, cholesterol, triglyceride, blood urea nitrogen, creatinine malondialdehyde and calcium. Also we conducted liver histopathologic examinations. Diltiazem, when administered 1 hour prior to CCl₄ treatment, significantly reduced the activities of ALT and AST, the contents of microsomal malondialdehyde and calcium in liver and microsome as compared with those of CCl₄-treated rats. In addition, histopathologic examination showed that diltiazem prevented the development of centrilobular necrosis induced by CCl₄ in liver tissue. Our results suggested that diltiazem could inhibit the formation of free radicals and the influx of calcium. Therefore diltiazem may be applied to suppress the liver damage caused by CCl₄.

Antihyperlipidemic effect of taurine was investigated in the hyperlipidemic rats induced by feeding a diet supplemented with cholesterol (1.5% in diet), vitamin D2(1.25 million IU/kg of diet) and cholic acid (0.5% in diet). The rats were fed the diet containing 1% and 3% of taurine for 8 weeks. The contents of the cholesterol and triglycerides in the serum and liver of the hyperlipidemic rats were increased as compared with those of the control group. Feeding taurine resulted in decreases in total cholesterol and triglyceride levels. The HDL-cholesterol level in serum was decreased in the hyperlipidemic rats, but by administration of taurine its level was increased. In the aorta of the animals, total cholesterol and triglycerides contents were reduced significantly by treatment with taurine. The contents of calcium in the heart of hyperlipidemic rats were greatly increased as compared with those of the control group. Treatment of taurine produced significant decreases in calcium contents in the heart muscle of the animals. These results showed that the hyperlipidemic states in this model of rats were reversed by treatment of taurine.

The bark of Ulmus davidiana var. japonica(Ulmaceae) has been described in traditional books to have diuretic and laxative actions, to ameliorate edematous disease, and to be used in insomnia and gastrointestinal disorders. Thus this study was carried out to elucidate some of the pharmacological activities of the plant extracts. The results obtained in this experiment indicated that its methanol extract elicited remarkable inhibition of HCl-ethanol induced gastric lesion, Shay ulceration and gastric secretion. However, it showed no anti-inflammatory action. The acute toxicity of the extract was low, that is, the minimum lethal dose was more than 2000mg/kg by oral administration in mice. The systematic fractionation of the methanol extract by hexane, ether, ethylacetate and butanol resulted in potent prevention of gastric erosion in butanol and water fractions.

Although Sulfonamides have broad Antibacterial ability, they have frequently administrated for maintance therapeutic level because protein binding ability is large, and crystalluria, their acetylmetabolite, has been reported frequently. In order to diminished those problem, Probenecid was administrated to rabbit concomitant, before 1 hour, after 1 hour of Sulfonamides. The results was follows: 1. Before 1 hour Sulfonamides, plasma level was increase and after 1 hour Sulfonamides, half-life was increase. 2. Plasma level of Acetylsulfadiazine, their acetylmetabolite, was diminished in concomitant, before 1 hour, after 1 hour of Sulfonamides.
