In order to study the level of the selected elements(Ca, Zn and Cd, etc.) in milk and their interrelationships with milk protein, raw milk samples in the vicinity of Seoul and market milk samples were collected in the summer, winter and early spring. The average Ca and Zn content of milk from different areas and different products varied between 0.06∼0.13% and 2∼5ppm, respectively, and the Cu content is below 0.lppm. Cd(regarded as a toxic element) content in milk is not detected and the Hg content is below the detection limit of AAS instrument in the milk produced in Korea. The interrelationship of these elements with milk components(specially with casein) is also studied. Generally, inorganic elements(Ca, Mg and Zn) are contained in the portion of casein and whey. The fraction of α_s -casein contains higher Ca amounts than that of β- or k-casein does, but we have not found out that these elements are combined with milk component or not.

In order to study the toxic level of dietary chromium and its interaction with other trace elements such as manganese and molybdenum on the growth, mortality and chromium accumulation in the internal organs, two experiments were conducted with day old chicks for a total period of 4 weeks. The body weight gain and mortality of the growing chicks were measured by two experiments. In the first experiment, the liver, kidney and pancreas were analyzed to determine chromium retention.. Growing chicks (0-2 wks), fed 1200 ppm and 1500 ppm of hexavalent chromium, exhibited considerable growth depression and mortality. Chicks (0-2 wks) fed 4000 ppm of trivalent chromium showed adverse effects on growth rate, but suffered no mortality. There was antagonistic interaction between hexavalent chromium and manganese and hexavalent chromium and molybdenum; however the mode and degree did not correspond to dietary chromium levels. Antagonistic interaction between trivalent chromium and managanese did not appear. In order to study the toxic level of dietary hexavalent chromium and its interaction with copper and zinc on the growth, mortality and accumulation of chromium in organs, an experiment was conducted with 720 chicks (Hisex White) for a period of 8 weeks. Four levels of hexavatent chrominm (O0ppm, 200ppm, 400ppm, and 600ppm) were added in the absence or presence of copper 1000ppm or zinc 400ppm. The results obtained from the experiment are summarized as follows: 1. Growing chicks(0-8wk) fed 200ppm of hexavalent chromium showed no adverse effects on the growth rate and mortality, but those fed 400ppm to 600ppm of hexavalent chromium exhibited remarkable growth depression and increased mortality(p$lt;0.05). 2. Dietary copper and zinc partially reduced the toxicity of hexavalent chromium, but the modes and degrees were not consistent with the various dietary chromium levels. 3. Chromium accumulation in the internal organs of the growing chicks was slightly increased as the dietary hexavalent chromium level was increased from 200 to 600ppm. And it was found that hexavalent chromium accumulation in the internal organs was partially reduced by dietary copper and zinc supplementation.

Lour levels of trivalent chromium(0ppm, 500ppm, 1,500ppm and 2,500ppm) were added in the absence or presence of cobalt 150ppm or zinc 600ppm. The results obtained from the experiment are summarized as follows: 1. Growing chicks(0-5wk) fed 500ppm of trivalent chromium showed no adverse effects on the growth rate and mortality, but those fed 1,500ppm to 2,500ppm of trivalent exhibited considerable growth depression(p$lt;0.05) without mortality. 2. Dietary cobalt and zinc did not reduced the toxicity of trivalent chromium. 3. Chromium accumulation into internal organs was slightly increased as the dietary trivalent chromium level was increased from 500 to 2,500ppm, but the degree and mode of the accumulation was different depending upon the internal organ. And adding cobalt and zinc failed to reduce the chromium accumulation.

In order to study the toxic level of dietary trivalent or hexavalent chromium and its interaction with silver, copper and zinc on the growth, mortality and feed requirement, an experiment was conducted with 780 chicks(Hanhyub 603) for a period of 3 weeks. The results obtained from the experiment are summarized as follows: 1. Chicks(0-3wk) fed 4,000ppm of trivalent chromium exhibited considerable growth depression(p $lt; 0.05), but showed no adverse effects on the mortality. 2. Growth depression induced by dietary trivalent chromium was not relived by silver or zinc, but relived slightly by copper. 3. Chicks(0-3wk) fed 1500ppm of hexavalent chromium exhibited considerable growth depression and mortality(p $lt; 0.05). Growth depression induced by dietary hexavalent chromium was not relieved by copper, zinc and copper + zinc, but mortality was significantly(p $lt; 0.05) relieved only by zinc. 4. The feed requirement was increased as the level of dietary chromium was increased, and adding silver, copper, zinc and copper+zinc failed to improve the feed requirement.

In order to study the high level toxicity of dietary chromium (III, VI) and its interaction with copper and zinc, an experiment was conducted with 480 chicks(Hanyub 603) for a period of 4 weeks. The results obtained from the experiment are summarized as follows: 1. Chicks(0-4wk) fed 5,000ppm of trivalent chromium exhibited considerable growth depression and mortality(p $lt; 0.05) 2. Growth depression and mortality induced by trivalent chromium were relieved by copper. 3. Chicks(0-4wk) fed 1200ppm of hexavalent chromium exhibited considerable growth depression and moutality(p $lt; 0.05). Growth depression induced by dietary hexavalent chromium was not relieved by copper and zinc, but mortality was significantly relieved by only zinc. 4. The feed requirement was increased as the level of dietary chromium (III, VI) was increased, and adding copper and zinc failed to improve the feed requirement.

한국인 인유 ( 人乳 ) 유청단백질의 Sephadex G-150 Gel 여과에 의한 분별 및 아미노산 조성

This experiment was carried out to study on the fractionation of Korean human whey protein by gel filteration and its amino acid composition. Human whey protein was fractionated by Sephadex G-150 column chromatography using THU buffer and observed by means of polyacrylamide gel electrophoresis. And then percentage yields of each fraction were datermined, and amino acid composition of human whey protein was also analyzed. The results are summarized as follows: 1. Korean human whey protein was separated into four fractions by Sephadex G-150 column chromatography. 2. In polyacrylamide gel electrophoresis, fraction-I was postulated as the mixture of lactoferrin, immunoglobulin and lysozyme, and fraction-II and III appeared as serum albumin and α-lactalbumin, respectively. 3. Percentage yields of fractions were 33.9% for the mixture of LF, IG and lysozyme, 5.6% for serum albumin and 58.2% for α-lactalbumin. 4. Analitical results of amino acid composition of human whey protein indicated that Asp, Ala, Leu and Lys contents in human whey protein were lower than α-lactalbumin in cow`s milk, but Phe contents were higher than that in cow`s milk.
