      • 저온성 Pseudomonas sp.에 의한 지방분해효소의 최적 생산조건에 관한 연구

        In order to isolate the bacteria which have lipolytic activities and to evaluate the effect of lipolytic activity on the milk and milk product quality, the raw milk samples were collected at several farms over Chon-Buk area. Each sample was spread on the tributyrin agar plates for the screening of lipase producing bacteria at 5℃ for 10days. Of the colonies grown on the tributyrin agar plate, an isolate B15 strain was selected for its strongest lipolytic activity and identified as a Genus of Pseudomonas sp. which was used for further studies. The cultural conditions for the lipase production were examined. As a result, the optimum cultural pH and temperature for the lipase production of Pseudomonas sp. were appeared at pH 8.0 and 25℃, respectively. Addition of 2.0%(w/v) starch as a carbon source and 0.5%(w/v) NaNO_(3) as a nitrogen source to the cultural medium was effective to the production of lipase.

      • Pseudomonas sp.K101이 생산하는 단백분해효소의 특성에 관한 연구

        Protease of Pseudomonas sp. K101 isolated from raw milk was characterized. The enzyme was most active at 40℃ and pH 7.0, and showed rapid decrease in its activity at higher temperature above 50℃ Activities of the protease were affected by the presence of metal ions tested, of which ?? and ?? stimulated the enzyme activity but ?? didn't. Notably, ?? ion could not replace the stimulatory effect of ?? ion. The thermostabilities of the enzyme treated at 35℃ and 40℃ were nearly constant at pH range of 5.0 to 8.0 but decreased sharply at higher alkaline pH. Addition of Ca2+ ion into the reaction mixture did not increase heat-stability of the protease whereas skim milk exerted preventive effect on denaturation by heat. Both PMSF, inhibitor of serine protease, and thiol reagents including iodoacetate and SDS inhibited considerably the enzyme activity. As to higher substrate specificity, β-casein was degraded best by the protease followed by whole casein, K-casein, α-casein, bovine serum albumin in that order. When commercial market milks inoculated with 50 microorganisms/ml and 500 microorganisms/ml of Pseudomonas sp. K101 were kept in refrigerator, tyrosine was detected by 9 days afterward in the former and 6 days in the latter.

        고준위 방사성폐기물 처분 기술개발 현황

        The amount of the high level radioactive wastes in Korea will be increased up to 14,297 MTU about 2010 year. Most of countries adopt the concept of deep burial repository in high level radioactive waste disposal. Because the high level radioactive wastes are very toxic in biosphere and to human, the data verifing its never return to the biosphere are requisite for the disposal. Presently, the evaluating techniques for the high level radioactive waste disposal are not fully developed. Therefore, in order to dispose the high level radioactive wastes in proper time the R & D of it is urged in our country. The R & D and/or the international joint research programme for the disposal of high level wastes have already been proceeded. In our country no plan for its disposal has been prepared. It is the time that the direction of the R & D is to be discused seriously. The R & D for the disposal of high level radioactive wastes in Korea is believed to be focused on developing the pecular techniques such as in situ characteristics of groundwater flowage, and change of properties of in situ rock mass at thermal effects.

      • 韓國産 人蔘成分의 變化에 關한 硏究 : 第2報: Saponin과 Sapogenin 成分의 比較 2. Comparison of Saponins and Sapogenins

        The changes of ginseng component was derived from the application of ginseng tissue culture, and ginseng callus produced here was proved to be similar to growing ginseng roots in a scent, tastes and content of components, and others. To study on the changes in saponins from callus mass by tissue culture, the callus was derived from the petiole of Korean Ginseng(Panax Ginseng C. A. Meyer) and cultivated on Murashige and Skoog's agar medium supplemented with 2.4-dichlorophenoxy-acetic acid and kinetin for 8 months. Then, well grown callus was analyzed for its components estimation. The results obtained are as follows: (1) When saponins isolated from callus mass were chromatographed on a silica gel plate, and determined by the thinchrograph TFG-10, the ratio of Rb, c to Rg(f) in saponins was 2.16 to 1 and Rb, c, d to Re, g(f) was 1 to 1.63, while in the case of saponins from the root of Panax Ginseng grown by soil culture, the ratio of Rb, c to Rg(f) was 1.03 to 1 and the ratio of Rb, c, d to Re, g (f) was 1 to 1.17. (2) Sapogenins were obtained from the hydrolysates of saponins, and determined by thinchrograph TFG-10. The ratio of panaxadiol to panaxatriol in sapogenins from callus saponins was 2.66 to 1, while the ratio of panasadiol to panaxatriol in sapogenins from ginseng root saponins was 1.86 to 1. From the results above mentioned. we concluded that the relative con- tents of sapogenins in saponins from callus mass by tissue culture were different from those in saponins from ginseng root b5- soil culture.

      • 백악기 진안분지 마이산 역암층의 고배수지 환경고찰

        According to an analysis of paleotopography in the circumference of the Cretaceous Maisan Conglomerate, Jinana Basin, the elevation was high in the east and low in the west, and the slope was dipping westward. The paleodrainage area for the Maisan Conglomerate is calculated to be about 734㎢. Analyzed by GUMBEL, IWAI and LOG-PEARSON TYPE Ⅲ, the PMP(probable maximum precipitation) and the PMF(probable maximum flowage) per 24 hours from 1 to 500 return-year-period are calculated to be 82 - 316㎜ and 697 - 2685㎥/sec, respectively. Comparison with the Roaring alluvial fan(Blair, 1987), which serves as an modern analogue for the Maisan Conglomerate in terms of depositional conditions, reveals the at the flow velocity at variant PMF with 1 - 500 return-year-period is about 24 - 94 m/sec and the clast size transported by the flow might have reached at 25 - 380m. These values are not realistic in modern environments. Conversly using the maximum clast intermediate diameter about 1m in the Maisan Conglomerate, the return-year-period at the inferred velocity of 5m/sec and PMF of 148㎥/sec is calculated to be less than 1 year, which seems common even in terms of the usual time scale. The calculated values of cross sectional flowage area using variant paleoslopes show much wider and deeper than those of the recent Roaring alluvial fan with the gentlest slope of 0.005.

        A bove all we must meet the following essential prerequisites in order to preclude various environmental pollutions: 1) We must have economical ability to bear the considerate expenses required to clean up the pollutions. 2) We must strive morally and willingly to work together for the social betterment of out posterity in future, to say nothing of giving up short-sighted selfishness determinedly. 3) Scientists or engineering specialists must disclose how harmful environmental pollutions are and study thoroughly how to eradicate their causes at once. The best and optimum means against pollution is scientific and technological one; control of motor exhaustion, appropriate disposal of waste matters issued from a variety of factories or stock farms or draining aqueducts, possible removal of the exhausted harmful gas from sylinders in power plants-these are to be taken into consideration so carefully that the amount of damagte from environmental pollutions can be minimized.

        We dealt with author of God's creationism, historical background, order of creation, character in science about the universe creation which is written in the Genesis and the Proverbs, and hypothesis in relation to evolutionism. Also, we considered recently prospect in differentiation of species by Genetic engineeing (Gene manipulation method) which is the latest science in modern Genetics. In the differentiation of species in biololgical evolution, as molecular biololgical research, though the gene of sex chromosome is demonstrated expressing by mutation or, that change of base sequence in gene, chromosome partial aberration (or, Deletion, Duplication, Translocation, Inversion) as well as numeral aberration in chromosome (like that : aneuploidy and polyploidy) etc, is able to induce of variety about species of some living things that has no prescribe proof of variety in original species yet. So we are able to explain complementary between Creationism and Evolution Theory. However, it is eternal truth to homeostasis for system sustenance of all living things that are the first creatures. Creationism which was recorded by ancients should be comprehensioned basis on science and knowledge of ancient. Christian promulgate that creationism the truth research in foundation of modern science, forever and should be keep Creation theology with redemption theology.
