초임계 이산화탄소를 이용한 겨우살이(Mistletoe, Viscum album) 추출물의 폴리페놀 함량 및 항산화 활성

소수성 저분자 물질의 추출에 효과적이며, 인체에 무해하고 친환경적 추출법인 초임계 이산화탄소를 이용하여 한국산 겨우살이를 추출한 후 추출물의 추출수율, 전자공여능 등의 항산화활성 평가 및 총 폴리페놀 함량을 측정하였다. 실험 결과 초임계 이산환탄소를 이용한 추출수율은 300 bar, 40℃에서 12.16%, 정제수와 주정을 이용한 추출물은 18.32, 23.84%로 초임계 추출물보다 높은 추출수율을 보였다. 전자 공여능은 초임계 이산화탄소 추출물이 15.73%(400 bar, 30℃), 정제수와 주정을 이용한 추출물이 36.73, 70.53%로 초임계 이산화탄소를 이용한 추출물의 활성은 일반 용매추출법으로부터 얻어진 추출물들에 비해 EDA 활성이 낮은 편이었다. 그러나 SOD 유사활성은 초임계 이산화탄소를 이용한 추출물이 22.54%(200 bar, 40℃), 정제수와 주정을 이용한 추출물이 7.85, 6.12%로 초임계 이산화탄소를 이용한 추출물이 일반용매를 이용한 추출물보다 우수한 활성을 나타내었다. 총 폴리페놀 함량은 초임계 이산화탄소 추출물이 17.57 wt%(200 bar, 50℃), 정제수와 주정을 이용한 추출물이 4.79와 5.46 wt%로 초임계 이산화탄소를 이용한 추출물이 일반용매 추출물보다 높은 함량을 보였다. 결론적으로 초임계 이산화탄소를 이용한 겨우살이 추출물의 전자공여능은 높지 않지만 우수한 SOD 유사활성과 총 폴리페놀 함량이 풍부하여 항산화 건강기능성 식품의 가능성을 갖는 것으로 사료되었다. This study was performed to investigate antioxidant activities of Mistletoe (MS) extracts by measuring electron donating ability (EDA), super oxide dismutase (SOD)-like activity and total polyphenol content (TPC). The extracts were obtained using supercritical carbon dioxide, hot-water and ethanol extraction. EDAs was the higher in ethanol extract (70.53%) than water (36.73%) or supercritical carbon dioxide extract (15.73% at 400 bar, 30℃) at concentration of 10 ㎎/mL. However, SOD-like activities and TPCs were the highest in the supercritical carbon dioxide extract. At concentration of 1 ㎎/mL, supercritical carbon dioxide extract (22.54% at 200 bar, 40℃) was higher SOD activity than water (7.85%) and ethanol extract (6.12%). Regarding TPCs of MS extracts, the content was the highest in supercritical carbon dioxide extract (17.57 wt% catechin equivalent at 200 bar, 50℃) followed by ethanol and water extract. These results suggest that it can be useful to develop functional foods using antioxidative active compounds of MS with high polyphenol contents.

To investigate the mechanism for the main chain-elongation process and the possibility of putative precursors as stater units in the biosynthesis of the Monascus pigments, feeding experiments with possible poly-β-ketide intermediates were carried out. Both crotonic acid and sorbic acid. especially In low concentrations, enhanced the pigment production while not increasing the dried mycelium weight appreciably. Also, it was observed that the feeding of sorbic acid and its ethyl ester was about two folds efficient in the pigment production than the feeding of crotonic acid and its ethyl ester. In addition to these acids, cinnamic acid and vinylacrylic acid were examined for their possibility as starter units. It was observed that the color of the culture fed with cinnamic acid was dominantly dark-red, but with overall decrease in the pigment production. Whey its ethyl ester was administered to the culture. however, the pigment production increased significantly. Also noted in 2D TLC study o1 the pigments was the increased production of red pigment and the formation of new red pigments.

Fe₃O₄ particles, prepared by a sol-gel method, were examined for their structural characteristic, particle shapes and sizes, and their magnetic properties. Two different chemical compositions (using a mol rate Fe<SUP>2+</SUP>/Fe<SUP>3+</SUP> = 1/2 and only Fe<SUP>2+</SUP>) and 2-methoxyethanol were used for making proper solutions. And the solutions were refluxed and dry in a dry oven and the samples were fired at 200~600 ℃ in the N₂ atmosphere. The formation of single-phased spinel ferrite powders was identified with the X-ray diffraction measurement as they were fired at above 250 ℃. The result of scanning electron microscopy measurement showed the increase of annealing temperature yielded the particle size increased. The magnetic transition was observed using the Mossbaur spectroscopy measurement. As the ferrite, prepared with the chemical composition (Fe<SUP>2+</SUP>/Fe<SUP>3+</SUP> = 1/2), was fired at 250 ℃, 78% of the ferrite had a ferrimagnetic property and 22 % of the ferrite was non-magnetic. In case of preparing the sample with only Fe<SUP>2+</SUP> and annealed at 200 ℃, it had a single phased spinel structure but its particle size was too small to be ferrimagnetic. The annealing temperature above 250 ℃ made powders a spinel structure regardless of the preparation method. They had a typical soft magnetic property and their saturation magnetization and coercivity became larger as the annealing temperature increased.

The intent of this study is to propose a campus masterplan for Dongshin University, which is a physical plan based on the prepared academic plan, space program and other useful resources. The procedural steps taken for the proposal is as follows: The close examination on the comprehensive academic plan of the university and the general space program of required buildings; The second step is to analyze the existing condition of the site including buildings with a view to grasping problems or difficulties and potentials; Two specific planning goals are sought and interpreted into several design concepts for a systematic design approach; Finally, a masterplan derived from these design concepts is proposed.
