Various biometerials have been researched and have been developed for treatment of some disease through transplantation to body. They have been evaluated by in vitro cytotoxicity test using some skin-derived cell lines for prediction of their biocompatibility in vivo. However, the results of experiments using mesenchymal or epithelial cells could not be considered in vivo immune reaction. In this study, we evaluated the biomaterial-elution (elute from high density polyethylene film) using some cell lines (L929, Jurkat, U937) in vitro, and then that results were compared with in vivo results from guinea pig sensitization test. In sensitization test, saline and elution of syringe could not induce erythema, but only DNCB (hypersensitive chemical) induce erythema at guinea pig sensitization test. In cell experiment, the cytotoxicity results of inflammatory cells (Jurkat; T lymphocyte, U937; monocyte) was no difference with L929 (fibroblast) in the overall trend. However, inflammatory cell lines were only secreted inflammatory cytokine (TNF-α, INF-γ) in some materials (biomateriallution, FAC, DNCB). And the biomaterial-elution did not have toxicity to the cells, but it induced the inflammatory cytokines in inflammatory cell lines only. So, we were predicted inflammatory reaction through the cytokine resultes of inflammatory cell lines, and it was more correlated with in vivo results than cytotoxicity test. Therefore, we suggested that the inflammatory cytokine assay using inflammatory cell lines are more effective method in vitro for evaluation of biocompatibility of biomaterials or chemicals.

Alopecia is the medical description of the loss of hair from the head or body. Alopecia tends to be involuntary and unwelcome. Alopecia does not vitally influence one`s life but patients who undergo alopecia appeal severe mental stress. The number of alopecia patients increases steadily. The current therapies used for treating alopecia have a number of limitations and to overcome such problems, many researchers have attempted to revive hair follicles by in vitro culturing hair follicle cells and implanting them in the treatment area. In this study, umbilical cord-derived mesenchymal stem cells(UC-MSCs) could be isolated and expanded successfully from the Wharton`s Jelly. Culture-expanded UC-MSCs formed aggregates similar to native dermal papilla(DP) in size and morphology under the special medium. Also the aggregates showed similar protein expression pattern of primary dermal papilla cells(DPCs). To confirm whether the aggregates could induce hair follicle instead of DP, they were transplanted with outer root sheath cells(ORSCs) in athymic nude mice`s inner-dermis where cells underwent proliferation and differentiation. After 6 weeks of transplantation, we observed new hairs on the mice`s scalp apparently. In conclusion, our results suggest that UC-MSC would be one of the cell sources for the treatment of alopecia in the future.
삼백초 추출물의 3T3-L1 세포에 대한 지방축적 억제효과

Obesity increases the risk of many adult diseases, such as atherosclerosis, diabetes, hypertension, ischemic heart disease and breast cancer. Inhibition of adipogenesis is an effective way in the anti-obesity management. Because of main components of Saururus chinensis is flavonoid, it has been showed some improvement by its antioxidant effects on the atherosclerosis, heart disease and diabetic hyperlipidemia. But mechanism of anti-obesity effect of S. chinensis is not clear. We investigated the effects of ethanol extracts of S. chinensis on adipogenesis in the 3T3-L1 pre-adipocyte. The 3T3-L1 cell line is commonly used to study adipogenesis in vitro. In this study, ethanol extracts of S. chinensis significantly decrease the lipid accumulation in the 3T3-L1 cells proved by measuring triglyceride contents and Oil red O staining. The proposed mechanism of inhibition of adipogenesis in the 3T3-L1 cells with ethanol extracts of S. chinensis is down-regulation of transcriptional factors and adipocytespecific genes such CCAAT/enhancer binding protein α (C/EBPα) and Peroxisome proliferator activated receptor γ (PPARγ) in concentration dependent pattern. These results suggest that ethanol extracts of S. chinensis inhibits adipognesis in the 3T3-L1 cells and can be used as a safe and efficient natural substance to manage anti-obesity.
