        Oligodeoxyribonucleotide의 화학적 합성 ; Deoxyribonucleoside의 인산화와 이량체 합성 방법의 개선

        Base의 아미노기와 5'-산소가 아실 및 dimethoxytrityl기로 각각 보호된 deoxyribonucleosidedml 3'-산소를 인산화시약인 o-chlorophenylphosphoroditriazole과 반응시켜 인산화중간체를 합성함에 있어서 수득률을 증가시킬 수 있는 반응조건을 조사하였다. 이 과정에서 주로 이용하고 있는 디옥산단일용매 보다 디옥산에 소량(5%)의 피리딘을 포함시키면 반응속도가 크게 증가하였으며, 단일용매 사용에서의 단점인 부반응도 일어나지 않았다. 인산화방법이 개선되므로 인산화중간체의 합성을 포함한 몇 단계의 반응을 쉽게 수행하여 모두 보호된 이량체를 합성하는 방법을 개발했다. 이렇게 개선된 방법으로 합성한 5종의 이량체들로부터 Hind Ⅲ인식자리를 지닌 십이량체를 합성하였다. The study was done with a focus on making the optimum condition on phosphorylation of deoxyribonucleoside with o-chlorophenylphosphoroditriazole as a phosphorylating agent. The result showed that the addition of 5 volume % pyridine to the dioxane solution accelerated the rate of reaction to a great extent and turned out to nearly quantitative yields on phosphorylation. On the basis of this improvement of optimum reaction conditions, a more efficient method to synthesize all-protected dideoxyribonucleotide from N, 5-O-blocked deoxyribonucleoside was developed. The dodecamer with a Hind Ⅲ recognition site was readily synthesized from five different dimers which were prepared through the newly improved method.

      • 1-Hydroxy-7-octylphenazine의 합성과 그의 항균성

        1-Hydroxy-7-octylphenazine was synthesized using borontribromide by the demethoxylation of methoxy group on the phenazine ring which was prepared by the condensation of 3,4-diaminooctylbenzene and pyrogallolmonomethylether. 3,4-Diaminooctylbenzene was prepared by the reducdtion of 3-nitro-4-aminooctylbenzene and pyrogallolmonometylether by the oxidation of o-vanilline (2-hydroxy-3-methoxybenxaldehyde). Surface tension of aqueous solution of this phenazine derivative was determined by Wilhermy model surfacetensionmeter. It was found that the surface tension of 1-hydroxy-7-octylphenazine solution was lowered to about 40 dyne/cm at 10-³mol/ℓ concentration, and its CMC (critical micelle concentration) was about 1.5×10-⁴M. Antimicrobial activity of 1-hydroxy-7-octylphenazine was also observed in this study. The identification of all the reaction products and structure determination of them were performed by melting point measurement and IR/NMR Spectroscopy.

        복무외상의 임상적 제특성에 대한 고찰

        This is a retrospective clinical study of 195 cases of abdoinal trauma treated at the Department of Surgery, Hanyang University Hospital during the period from july 1981 to june 1991. The results are as follow ; 1) The sex distribution was 141 males (72.3%) and 54 females (27.7%) with the ratio of 2.6 : 1, and by age, high incidence was seen in the 3rd and the 4th decade(54.4%). 2) The seasonal distribution was not characteristic, but the hourly distribution showed the high incidence from day to evening in blunt trauma, evening to night in penetrating trauma. 3) Among the 195 cases of trauma, blunt trauma was 128 cases (65.6%) and penetrating trauma 67 cases (34.4%), with the ratio of 1.9:1. Traffic accident was most common in the former, stab would in the later. 4) The time lapse between trauma and arrival at hospital was predominantly within 2 hours (40.5%). The operation was predominantly performed from 8 hours to 12 hours (34.7%) after trauma. 5) The clinical manifestations were consisted of abdominal pain (88.7%), tenderness (82.6%), rebound tenderness (50.3%), rigidity (42.6%), and decreased peristalsis (34.3%) in order of frequency. 6) The preoperative value of Hb and WBC were below 12gm/dl in 53.4%, and above 10.000/ml in 77.0% respectively. The result of paracentesis was positive in 79.2% (sensitivity ; 82.1%, specificity ; 41.7%). 7) Among the diagnostic measures used to determine the surgical indications, the clinical manifestations were most commonly used in 32.3% (22.6% in blunt trauma, 50.7% in penetrating trauma). CT in 25.1%, paracentesis in 24.1%, and ultrasonography in 18.5%. 8) The abdominal organs injured were spleen (22.7%), liver(16.7%), mesentery (13.1%), small bowel injury. Multiple organ injury was 48.6% in blunt trauma, 22.7% in penetrating trauma. 9) The associated extra-abdominal injuries were seen in 52.3% of cases suffered abdominal organ injury ; in 61.9% of blunt trauma, and in 29.5% of penetrating trauma. They were head injury (42.8%), chest injury (23.6%) low extremity fracture (10.9%), pelvic bone fracture (6.9%) in order of frequency. 10) Of the total 195 patients, operation was performed in 167 cases (85.6%), and 28 cases (14.4%) were treated conservatively. 11) Transfusion of blood was done in 144 cases (73.8%), and not required in 51 cases (26.2%) 12) Post-operative complications occurred in 54 cases (32.3%), and pulmonary complication was most common. 13) The mortality rate was 5.1% (10 cases), and the causes of death were hypovolemia, sepsis, and renal failure in order of frequency.

      • Internucleotide Phosphotriester Bond 형성에 따른 Arylsulfonyl azolide계 축합시약의 반응성

        Arylsulfonylimidazoles, arylsulfony-1,2,4-triazoles and arylsulfonyl-3-nitro-1,2,4-triazoles were synthesized and employed in the formation of a phosphotriester bond between appropriately protected monodeoxyribonucleotides. New condensing agents, benzenesulfonyl-3-nitro-1,2,4-triazole, and p-toluenesulfonyl-3-nitro-1,2,4-triazole were found to be excellent in the dideoxyribonucleotide synthesis via the phosphotriester method.

      • Dideoxyribonucleotide의 비효소적인 방법에 의한 합성과 분리방법

        A protected dinucleotide, 5'-O-dimethoxytritylthymidylyl-(3'→5')-3'-O-acetyl-N??-benzoyldeoxyadenosine, was synthesized by 'Phosphodiester Method' and separated by solvent extraction/recrystalization. The amino group of the base, 3'-hydroxyl group of deoxyadenosine-5'-monophosphate, and 5'-hydroxyl group of thymidine were previously protected with benzoyl, acetyl, and di-p-methoxytrityl group respectively before the condensation between the nucleotide and the nucleoside was conducted. The condensation reaction was more efficiently conducted with triisopropyl benzenesulfonyl chloride(TPS) than with dicyclohexylcarbodiimide(DCC). The dinucleotide synthesized was purified utilizing solvent systems isopropyl ether/0.5M triethylamminium bicarbonate (TEAB), ethyl ether-ethyl acetate (5:5, v/v)/0.5M TEAB, and 0.5M TEAB/CH₂CL₂-CCl₄(4:6, v/v). This method was more efficient than the ordinary diethylaminoethyl (DEAE) cellulose column chromatography.

      • Deoxyribonucleoside의 보호와 di-p-Methoxytrityl기의 제거에 대한 Alcohol류의 효과

        5'-O-Dimethoxytritylthymidine and N-benzoyl-5'-O-dimethoxytrityldeoxyade-nosine were synthesized from the corresponding deoxynucleosides and some factors influencing their yields were also investigated. To remove the dimethoxytrityl group from 5'-O-Dimethoxytritylthymidine and N-benzoyl-5'-O-dimethoxytrityldeoxyadenosine, ZnBr₂/CH₃NO₂, ZnBr₂/CH₃NO₂in the presence of alcohols, and CH₃COOH/CH₃NO₂systems were employed. The rates of the detritylation as well as the extent of side reaction in these systems were estimated and the effect of alcohols on the reactions was discussed.
