      • Saccharomyces cerevisiae에서 인체 Lipocortin-1의 유전자 발현 및 분비

        Phospholipase A2의 저해 단백질로서 항염증 치료제로 알려진 인체 lipocortin을 효모로부터 생산하기 위해 인체 lipocortin-1 유전자의 cloning 및 발현 연구를 수행하였다. 인체 lipocortin-1 cDNA를 CYC1, GALl-GALlO 및 PHO5 유전자의 promoter를 갖는 발현 vector에 cloning시킨 후 유전자의 발현을 조사한 결과 각 promoter의 강도 및 사용한 plasmid의 copy수에 따라 발현효율이 달라져 GALl-GALlO promoter와 $2{\mu}m$ 기원의 multicopy수의 plasmid를 사용했을 경우 인체 lipocortin-1 유전자의 발현효율이 가장 높음이 확인되었다. 효모 내에서 생산된 인체 lipocortin을 배지로 분비시키기 위해 효모의 ${\alpha}$ 교배인자의 pre-pro leader 배열과 인체 lipocortin-1 유전자를 융합시킨 결과 효율적인 post-translational processing을 거쳐 인체 lipocortin만이 일부 분해된 상태로서 체외로 분비됨이 관찰되었다. Human lipocortin is a phospholipase A2 inhibitory protein and known as a potential antiinflammatory agent. Thus, molecular cloning and expression of human lipocortin-1 gene was tried using a number of yeast expression vectors. cDNA of human lipocortin-1 was placed under the control of CYC1, GAL1-GAL10 and PHO5 promoters. The expression levels were varied according to the promoter strength and the plasmid copy number. It was observed that the human lipocortin-1 gene was able to be expressed under the control of all promoters employed but the highest level of gene expression was achieved by use of the divergent GAL1-GAL10 promoter. The $2{\mu}m$ based multicopy number plasmid showed an increased expression level by 50% compared with ARS based single copy number plasmid. To excrete human lipocortin produced in Saccharomyces cerevisiae cells into the culture medium, the pre-pro leader sequence of yeast a mating factor gene was fused with human lipocortin-1 gene. The efficient secretion of the native protein was observed with some extent of degradation in the culture medium after the post-translational processing of the fused protein.

        College students’ attitudes toward business ethics: A comparison between Korea and China

        본 연구는 한국과 중국 학생들의 기업 윤리 태도를 비교 분석하여 양국 기업의 전략적 차원 기반 마련 및 발전을 위한 의견 제시와 고찰을 목표로 삼았다. ATBEQ를 통해 수집된 데이터를 기반하여, 사회 및 개인 차원에서의 한국과 중국 학생들의 윤리관 분석을 위해 사회진화주의, 마키아벨리주의, 윤리객관주의 3가지 이론적 관점을 제시했다. 연구 결과, 한국 학생이 중국 학생보다 윤리객관주의가 더 높았고 중국 학생은 마키아벨리주의와 사회진화주의가 더 높은 것으로 나타났다. 종교와 성별에 따른 경영윤리관 설문 결과에서 한국과 중국은 다른 양상을 보였다. 종교의 유무에 따라 한국 학생은 윤리객관주의와 사회진화주의에 대해 차이를 보였으며 중국 학생은 윤리객관주의에 대해서만 차이를 보였다. 또한, 성별에 따라 한국은 윤리객관주의와 사회진화주의에 대해 차이가 나타났으며, 중국은 윤리객관주의와 마키아벨리주의에 대해서 차이가 나타났다. The purpose of this paper is to get practical conclusions for the construction and development of business ethics between Korea and China by contrasting differences between two countries. Moreover, it is hoped that the paper could contribute to the two countries establishing their strategies in the field of business ethics. This paper explores the factors affecting business ethic from the theoretical perspective. Hypotheses are presented at the social and individual levels to explain the differences in attitudes of Korean and Chinese students towards three business ethical philosophies: Social Darwinism, Machiavellianism, and Moral Objectivism. Through this research, we found that there are differences depending on GDP per person, economic growth rate and corruption degree. And Korean and Chinese students’ management ethics attitudes differed depending on the existence of religion and differ by gender.

      • 필로티 건축물의 지진취약도평가를 통한 변위기반 한계상태 분석

        It is difficult to determine the limit states for the piloti-type building structures consisting of two lateral-resisting systems since the existing drift-based limit states (i.e., interstory drift ratio, IDR) are defined for a lateral-resisting system. This study aims to analyze the existing drift-based limit states through the fragility assessment for piloti-type building structures. The piloti structures consisting of a shear wall system and a RC moment frame was used as a prototype model. The incremental dynamic analysis (IDA) was performed using IDR and concrete strain as the engineering demand parameter (EDP) of the target buildings. The fragility assessment was performed using seismic demand and limit states derived through IDA. As a result, the limit states suitable for the piloti-type building structures should be less than that of the shear wall system in practice.

        산업경계를 확대한 오리온 제과의 혁신전략 : 피터 드러커의 관점에서

        본 논문은 오리온 제과의 혁신사례를 분석하여, 기업의 혁신이 전통적인 산업의 경계가 확대되는 사례를 제시한다. 대형 스낵업체인 오리온 제과는 영양균형과 건강에 주목한 제과제품을 개발하여 제과산업의 경쟁법칙과 산업한계를 동시에 극복하는 성과를 보여주었으며, 향후 새로운 영역으로 제과산업이 확대될 수 있는 계기를 마련하였다. 본 논문에서는 피터 드러커의 혁신관점을 적용하여 오리온 제과의 혁신과정을 설명하고 중요한 교훈을 제시하고 있다. 예상하기 어려운 시장의 변화를 인식하기 위한 주의깊은 마켓센싱, 시장환경의 변화에서 오는 위기를 기회로 재해석할 수 있는 창조적 파괴, 새로운 기회를 실제로 구현하기 위해 필요한 선행적인 내부혁신, 그리고 혁신과정에서 필요한 내부소통과 협업을 가능하게 하는 기업문화 등이 성공적인 혁신의 추진을 위한 중요한 교훈으로 분석되었다. 전통적인 산업경계를 확대를 선도하는 리더십을 발휘하면서 오리온 제과는 빠르게 글로벌 기업으로 발전하고 있다. 본 논문의 사례에서 제시한 오리온 제과의 혁신과 창조적 파괴역량은 21세기의 급변하는 시장환경에서 경쟁하는 다른 기업에게도 중요한 교훈을 제시한다. With a brief historical discussion of Orion's innovation, this paper portrays how organizational innovation may expand traditional boundaries of existing industries. Orion, a major snack manufacturer, has successfully transformed its product positioning. Responding to the changing customer environment, Orion has introduced medically balanced and health conscious snack products, redefining the snack industry. Orion's new market definition includes health and natural foods, successfully enlarging the traditional scope of the snack industry. This paper applies Peter Drucker's innovation framework to explain Orion's innovation process and to identify key lessons. These include (1) market sensing, through which unexpected market disruptions may be recognized, (2) creative destruction, through which the disruptions may be reinterpreted and accepted as innovation opportunities, (3) proactive internal innovations, through which new opportunities may be rapidly realized, and (4) organizational culture and identity, through which internal communication and collaboration may be fully exploited. Taking advantage of its leadership in the redefined snack industry, Orion is rapidly expanding its operations in the global market. Orion's demonstrated competence in innovation and creative destruction provides meaningful lessons to other organizations in the rapidly changing market environment of the 21st Century.

        한국산 거머리로부터 항혈전단백질의 검색과 분리 , 정제

        Two Korean leech species, Hirudo nipponia and Whitmania sp., were examined for the presence of anticoagulant activity. Crude extracts of the leech head parts were fractionated by ethanol and the anticoagulant activity of these extracts was assessed with respect to the inhibition of thrombin, factor Xa and trypsin using the chromogenic substrate assay. While the extract from Hirudo nipponia inhibited all three serine proteases employed in the assay, that of Whitmania sp. inhibited only trypsin, but not thrombin and factor Xa. Thus, the extract from Hirudo nipponia which shows antithrombin activity was subjected to further purification. Three steps of column chromatography including gel filtration on Superose 12 column, anion exchange on Mono-Q and reverse phase HPLC on C-8 column could resolve 3 antithrombin activity peaks from anti-trypsin and anti-factor Xa activities, indicating the presence of multiple forms of thrombin-specific inhibitors in Hirudo nipponia. N-terminal amino acid sequences of the two of these forms showed a limited degree of homology with a portion of heparin cofactor Ⅱ.

        한국산 거머리인 Hirudo nipponia와 Whitmania sp.의 추출액으로 trypsin, factor Xa, thrombin에 대한 저해활성을 조사하였다. Hirudo nipponia로부터 얻은 추출액은 $20{\mu}g/ml$ 농도에서 이 효소들의 활성을 완전히 저해하는 반면 Whitmania sp.의 추출액은 동일조건하에서 trypsin만을 저해하였다. 이 Hirudo nipponia의 추출물로부터 항thrombin 인자를 분리 정제하기 위하여 anion exchange chromatography, size exclusion chromatography 및 HPLC를 이용하였고 최종적으로 세 종류의 단백질(I-A,I-B,I-C)을 얻었다. 이 중 두 종류 단백질(I-A, I-B)의 아미노말단 서열을 부분적으로 분석한 결과 이 단백질이 주로 산성 아미노산잔기로 구성되어 있으며 heparin cofactor II의 일부분과 유사성이 있음이 발견되었다.
