In this study, we present different physiological responses to cold acclimation between the freezing tolerant barley landrace, Jeonnamjaerae, and the freezing sensitive line, PI283398, chosen by the previous field test. We tried to identify some key factors promoting freezing tolerance in barley by analyzing the physiological responses to cold acclimation. TTC reduction test indicated a significant difference in freezing tolerance between the two genotypes. The apoplastic protein concentration highly increased by cold acclimation at 4oC. Apoplastic protein concentration of PI283398 slowly increased at early stages of cold acclimation, but it decreased with the prolonged cold treatment. In case of Jeonnamjaerae, a low level of the antifreeze activity was detected in the apoplastic extracts. SDS-PAGE showed that most polypeptides were more rapidly induced from leaves of Jeonnamjaerae than PI283398. Our results suggested that difference in concentration and accumulation level of apoplastic proteins between freezing tolerant and sensitive genotypes would be a key factor of freezing tolerance.
