To address low H<sub>2</sub> production yield by dark fermentation, co-production of H<sub>2</sub> and ethanol by E. coli from glucose, was suggested in this study. Moreover, ethanol is one of the attractive biofuel producible through the fermentation. In order to increase the cofactor availability for ethanol production, operation of Pentose-Phosphate (PP) pathway was tried by deleting pgi in the base strain (E. coli BW25113 ΔhycA ΔhyaAB ΔhybBC ΔldhA ΔfrdAB). Additional over-expression of G6PHD and 6PGDH recovered the cell growth of Δpgi mutant as well as increased co-production yield by decreasing acetate formation. Resulting strain increased co-production yield (1.65 for H<sub>2</sub> and 1.38 mol/mol for ethanol) which corresponds to ~85% energy recovery from glucose showing the possibility of operating PP pathway under anaerobic condition.

The purpose of this study is to those with ingrown toenails had no difficulties inwalking and their pains were alleviated after they underwent non-surgical operationtreatment using B/S SPANGE's nail tips rather than surgical operation treatments intreating ingrown toenails. The first, The respondents said that the toenail treatment usingB/S SPANGE was simpler than the existing many other treatments, and that they had noinconveniences in walking after the treatment, and that their pain was effectivelyalleviated. The secord, The study subjects replied that the B/S SPANGE's glassreinforced fiber tips were hardly shown with the naked eye and were not distinguishedeven when nail enamel was put on the tips, and that they were highly satisfied with theaesthetic effects. The third, Regarding the observation of the tip size change through theprior- and post-toenail form before and after the test, it was found that B/S SPANGE'stip size before the test was 16mm∼18mm, but that the tip size after the test was 20mm∼22mm. which means that toenail form changed after the treatment. Accordingly, thisresearcher tried to provide this study as a fundamental material in alleviating thepsychological pressure of those with ingrown toenails caused by operational pains andeasing the concerns about the pains after the operation, and in suggesting an effectiveconservative treatment in the short operation time and aesthetic aspects.

This study attempted to determine public knowledge regarding what constitutes abnormal nails (both fingernails and toenails) in a nail salon and investigate changes in awareness of aesthetic nail treatment. For this, an empirical survey was conducted, and the study results found the following: First, in terms of changes in perception of abnormal nails depending on nail care habits, as the length of the nail increased, the ability to judge nail damage improved. In addition, the view of nail care was favorable. As nails got longer, even though the ability of judgment on nail damage declined, the view of nail care was still favorable. Second, according to an analysis of correlations between nail shape and the experience of abnormal nails, the shape of fingernails and toenails was correlated with the experience of abnormal nails. Third, no difference was found in terms of perception of abnormal nails depending on whether or not the respondents visited or did not visit the nail salon in all questions. In other words, perception of abnormal nails was high regardless of whether or not they visited or did not visit the nail salon. Fourth, in terms of differences in perception of cosmetic nail procedures at a nail salon, those who had visited a nail salon before displayed a higher level of awareness. In terms of the level of perception on abnormal nails between nail experts and non-experts, in addition, the former was higher than the latter.
Co-production of 3-hydroxypropionic acid and 1,3-propandiol by recombinant Klebsiella pneumoniae

3-hydroxypropionic acid (3-HP) production from glycerol has gained much attention due to the importance of 3-HP as platform chemical. 3-HP production is composed of two reactions i.e. conversion of glycerol to 3-HPA oxidation to 3-HP. Klebsiella pneumoniae, one of the candidates for 3-HP production, has advantage of natural synthesis of B<sub>12</sub> under anaerobic condition. However, further reaction of 3-HPA to 3-HP involves the reduction of NAD<sup>+</sup> and consequently NAD<sup>+</sup> regeneration is very important for efficient production of 3-HP. On the other hand, production of 1,3-PDO by NADH-dependent oxidoredcutase of K. pneumoniae has been studied extensively. Hence, in this study, we tried co-production of 3-HP and 1,3-PDO using K. pneumoniae mutants and studied the effect of various environmental factors and genetic backgrounds on co-production yields change in K. pneumoniae.

Microbial 3-HP production from glycerol requires glycerol dehydratase (GDHt) and aldehyde dehydrogenase (ALDH). GDHt requires coenzyme B12 and ALDH requires NAD+ as cofactors for the production of 3-HP from glycerol. The imbalance between GDHt and ALDH activities cause the accumulation of toxic intermediate, 3-hydroxypropionaldehyde (3-HPA). As a solution to these problems, co-production of 1,3-PDO along with 3-HP has been suggested. 1,3-PDO production could regenerate NAD+ and utilize 3-HPA, thus avoiding 3-HPA accumulation and also facilitate the process to be performed under microaerobic conditions. But acetate was produced as major metabolite, which is very toxic to the cells. Therefore, in this study, we attempted the down-regulation of glycolytic pathway and disruption of byproducts formation pathway. We also attempted to operate TCA cycle/glyoxylate shunt to reduce the carbon traffic at pyruvate node.
Production of 1,3-Propanediol from glucose using Klebsiella pneumoniae

With the increase in demand for biodegradable polyester composed of 1,3 propanediol (1,3-PD), the microbial production of 1,3-PD has received much attention. However, till now, native microorganisms directly producing 1,3-PD from glucose was not identified and/or studied. Most microorganisms prefer glucose and glucose takes most portion in lignocellulosic biomass composed of various carbohydrates. Although using glucose is beneficial, there are only few reports on the production of 1,3-PD from glucose. In this study, we focus on development of strains that can convert glucose to 1,3-PD. For these purpose, two genes encoding glycerol-3-phosphate dehydrogenase and glycerol-3-phosphatase from Saccharomyces cerevisiae were overexpressed. Further, the produced glycerol was converted to 1,3-PD using native enzymes in Klebsiella pneumoniae J2B. Efficient recombinant strains developed in our study could lead to economical production of 1,3-PD.
Characterization of 1, 3-PD oxidoreductase (DhaT) from Klebsiella pneumoniae J2B

With the development of a new biodegradable and biocompatible polyester composed of 1, 3 propanediol (1, 3-PD), the bioconversion of glycerol and/or glucose to 1, 3-PD has gain much interest. In this biological system, glycerol is dehydrated to 3-HPA by glycerol dehydratase and 3-HPA is then reduced to 1, 3-PD by NADH dependent 1, 3-PD oxidoreductase encoded by dhaT gene. Despite practical importance of DhaT for 1, 3-PD production, there are few studies characterizing DhaT in terms of toxicity of 3-HPA intermediate and reversibility of DhaT. This work aims at kinetic characterization of DhaT from K. pneumoniae. The dhaT gene encoding 1, 3-PD oxidoreductase was cloned from the recently isolated K. pneumoniae J2B, and expressed and purified. Then, the reaction kinetics of DhaT for the forward and backward reactions were investigated using several aldehydes and alcohol as substrates.
