노랑초파리 발생과정에서 sta 유전자의 발현 양상에 관한 연구

Drosophila stuvarista(sta) gene encodes the ribosomal protein D-p40, consisting of 270 amino acids. The D-p40 is highly conserved between such evolutionarily distinct organisms as human(63% identity) and hydra(58% identity) and has the effect of Minute phenotype such as other ribosomal proteins. We identified the sta gene in the process of Drosophila cDNA library screening and analyzed the developmental expression pattern of sta gene in the transcription level using in situ hybridization. Each animal stage and tissues of 3rd instar larva and adults were showed ubiquitously expression pattern. These results were similar to other ribosomal proteins that represented Minute phenotype. Also, we expressed the 31kDa recombinant proteins of D-p40 using his-tag expression system in E. coli and purified by affinity chromatography. Immunoblot result of the developmental stages was showedthat strongly detection only the stage of embryo and pupa. This result suggested that the expression of sta gene were regulated in the level of post-translation modification ailed considered that sta gene had to other function during development as well as to function as a core ribosomal component.
노랑초파리 ecdysone 결핍 돌연변이(ecd1ts)에서 기능적 ecdysone수용체 구성분의 발현

In Drosophila, the steroid hormone ecdysone triggers the key regulatory cascades controlling the coordinated changes in the developmental pathway of molting and metamorphosis. Ecdysone action is mediated by heterodimer consisting of the ecdysone receptor(EcR) and ultraspiracle proteins(Usp). Heterodimers of these proteins bind to ecdysone response element and ecdsone to modulate gene transcription. In this study, the transcripts of functional ecdsone receptors and ecdysone response genes(Fbp1 and Uro) in ecdysoneless(ecd-1ts) mutation were analyzed by using a RT-PCR and competitive RT-PCR method. The expression of functional ecdysone receptor components were not correlated with tissue-specific ecdysone response genes(Fbp1 and Uro), Suggesting that several other factors involved in the complexity of various ecdysone responses in Drosphila melanogaster. But we showed that the transcripts of EcR-B1 isoforms were expressed with higher level than those of EcR-A in the late third instar larvae of ecdysoneless(ecd-1ts) mutation.
흰쥐에서 사염화탄소에 의해 유도된 간 상해에 미치는 키토산 올리고당의 영향

Chitosan is the deacetylated product of chitin. Chitosan and its derivatives have many properties that make them attractive for a wide variety of health applications. We reported recently that water soluble chitooligosaccharides have hepatoprotective effect on ethanol induced liver injury in the mouse. The current study examined the effect of water soluble chitooligosaccharides on CC1₄-induced liver injury in rats. Male Sprague-Dawley rats were orally treated with CC1₄without or with administration of 5% chitooligosaccharides. After 5 weeks after CC1₄treatment, the histological sections and the expression of mRNA of interleukin-12(IL-12), hepatocyte growth factor(HGF), and tumor necrosis factor(TNF- ) were examined. The liver section of rats supplemented with chitooligosaccharides showed attenuated effects of the necrosis, steatosis, cirrhosis compared with those of non-supplemented rats. The level of TNF- mRNA in CC1₄-treated rats were higher than those in the rats supplemented with chitooligosaccharides, while the levels of IL-12 and HGF mRNA were higher in the rats supplemented with chitooligosaccharides than those of in CC1₄-treated rats. These findings suggested that water soluble chitooligosaccharides may have a hepatoprotective activity against CCl₄-induced liver injury in rats.
Developmental Profiles of the Functional Ecdysone Recepytor Transcripts Using RT-PCR

노랑초파리에서 스테로이드 호르몬인 ecdysone은 탈피와 변태의 발생과정에서 coordinated changes의 key regulatory cascades을 시발한다. Ecdysone의 작용은 ultraspiracle 단백질(USP)과 ecdysone receptor(EcR)의 이합체에 의해 매개된다. 이들 단백질의 이합체는 ecdysone response element(EcRE)에 결합하여 유전자의 전사를 조절한다. 최근에 기능적인 ecdysone receptor는 USP와 ECR isoforms간에 형성된 이합체 복합체라는 사실이 알려지게 되었다. 본 연구에서는 RT-PCR(Reverse TRanscription Polymerase Chain REaction)방법을 이용하여 기능적인 ecdysone receptor전사체의 발생단계별 양상을 조사하였다. usp과 EcR전사체들은 비록 양적인 차이는 있지만 분석된 전 발생단계에서 검출되었다. 본 연구결과는 기능적인 ecdysone receptor의 유전자의 발현은 초파리 생애에서 관찰되는 ecdysone titer와 밀접하게 관련되어 있음을 암시해 주었다. It is known recently that the functional ecdysone receptors are complexes produced by the heterodimerization between USP and three EcR isoforms. In this study, the transcripts of functional ecdysone receptors during development were analyzed by using a RT-PCR(Reverse Transcription Polymerase Chain Reaction) assay. The transcripts of usp and EcR isoforms were detected in all developmental stages. This study revealed that the expression patterns of functional ecdysone receptors were correlated with ecdysone titers showing during Drosophila life cycle.
