볏짚의 사료가치 증진을 위한 적정 알카린 H2O2 처리 수준에 관한 연구

This study was carried out to determine the adequate levels of alkaline hydrogen peroxide treatment for the improvement of nutritive value of rice straw. In vitro digestibility and chemical analysis after several treatments were measured. Treatment variables were soaking time(12, 24, 48 and 96 hrs), temperature(5, 25, 50 and 75℃) of alkaline H₂0₂ solution, H₂0₂ concentration (0.5, 1, 2, 3 and 4%) and substrate /solution ratio (1, 2, 3, 4 and 5g/ml). The results obtained were summarized as follows. 1. The DM digestibilities or rice straw treated with H₂O₂ for 24 and 48 hours were higher than those of the others(p$lt;0.05). There was no differ ences when the rice straw was treated for 12, 72 or 96 hours. Cell wall contents were not affected by the time of treatment. 2. The DM digestibilities were higher at pH 11.5 or above(p$lt;0.05) and was decreased as pH declined. When rice straw was treated with H₂O₂ at pH 11.5 or higher, NDF, ADF and cellulose contents were increased. However, lignin content and DM recovery percentages were decreased. 3. The DM digestibilities were not influenced by the ratio of straw versus liquid. The contents of NDF and cellulose were not affected up to the ratio of 4g rice straw/50m1, but were decreased at 5g rice straw/50m1. Lignin content and DM recovery percentages, however, were increased at 5g rice straw /50m1. 4. The DM digestibilities were not different among treatments at 5, 25 and 50℃. However, the rice straw treated at 75℃ showed lowest digestibility of all treatment (p$lt;0.05). The content of NDF, ADF and cellulose was increased by increasing temperature. However, lignin content and DM recovery percentages were decreased by increasing temperature. 5. The AHP treated rice straw had higher content of ADF and cellulose, and lower content of hemicellulose and lignin as the concentration of H₂O₂, increased. But hemicellulose, lignin and DM recovery percentages were decreased. The NDF content was not different among five different concentrations of H₂0₂ solution. In conculsion, based upon the results of present experiments the most desirable method is to soak rice straw in 1% alkaline H₂0₂ solution at pH 11.5, at room temperatrve (25℃), for 24∼48 hours and at the ratio of 4g rice straw /50m1 solmtion.
천연 항산화제로서 포도씨 분말 첨가가 육용계의 성장 및 항산화 작용에 미치는 영향

본 연구는 포도씨 분말 첨가 사료가 육계의 성장, 사료 이용성 및 체 조직내 항산화 효소 및 관련 생화학 지표 인자에 미치는 영향을 조사하기 위하여 수행되었다. 전체 50수 3일령 육계를 2주간 예비 실험 기간을 거친 다음 각 군당 10수씩 선발하여 대조구 및 포도씨 분말 1% 첨가구 등 2개 구를 설정하여 3주간 사양 시험을 실시하고 혈액 생화학 성분 및 체조직의 항산화 지표를 분석하였다. 그 결과는 다음과 같다. 1. 육계 사료내 1% 수준의 포도씨 분말 첨가는 체중, 일당 증체량, 사료 섭취량, 사료 요구율 및 장기 무게 등에 유의적인 영향을 미치지 않았다. 2. 혈액내 total antioxidant status(TAS)는 포도씨 첨가구에서 유의적(P<0.05)으로 증가하였으며, 혈중 생화학적 성분과 조직 손상을 나타내는 AST, ALT, LDH 등의 수준은 모든 구에서 비슷한 수준을 보였다. 3. 포도씨 분말 첨가는 소장 점막 세포내 SOD 활성도를 유의적으로(P<0.05) 증가시켰다. 그러나 간 조직내 GSHPx, SOD, GST 등과 같은 항산화 효소, 환원형 glutathione 및 지질 과산화(MDA) 수준은 대조구 및 포도씨 분말 급여구간에 차이는 없었다. 이상의 결과를 종합해 볼 때 천연 항산화제로서 포도씨 분말 1% 첨가 사료는 성장 및 사료 이용성에 특이적 영향을 미치지 않았으나, 혈액내 TAS 및 소장내 SOD활성도를 현저히 증가시키므로 육계에서 천연 항산화원으로서의 사용이 가능할 것으로 사료된다. A total of twenty, 2-wk-old male broiler chickens were allotted into control diet(CON) or a diet supplemented with 1% ground grape seed(GGS). They had free access to feed and water for 3 wk. Growth performance and antioxidant markers in plasma, intestine and liver were determined. Dietary addition of 1% GGS did not affect weight gain, feed intake, feed conversion and organ weight in 35 day-old broiler chickens significantly. There was no difference in plasma levels of glucose, triglyceride, cholesterol, AST, ALT and LDH activity. However, total antioxidant status(TAS) in blood increased(P<0.05) in chickens fed the diet supplemented with 1% GGS compared to those fed the control diet. In addition, the specific activity of intestinal superoxide dismutase(SOD) increased(P<0.05) in birds fed the diet supplemented with GGS. However, the activities of intestinal gluthathione peroxidase(GSHPx) and gluthathione -S- transferase(GST) and hepatic SOD, GSHPx and GST were not affected by the dietary GGS. The levels of reduced glutathione and lipid peroxidation in the small intestine and liver were not different between the two groups. In conclusion, dietary supplementation of 1% GGS did not result in a negative effect on growth performance. In addition, some antioxidant indicators including blood TAS and intestinal SOD were markedly elevated in response to dietary GGS. Therefore, dietary addition of 1% GGS may be beneficial to improve antioxidant capacity in broiler chicken.
