고수(Coriandrum sativum L.) 지상부 추출물의 항산화 및 항염증 활성 효과

Coriandrum sativum L., an annual herbaceous plant of Apiaceae family. The present study evaluated the anti-oxidant activities and anti-inflammatory effects of ethanol extracts of C. sativum. The anti-oxidant activities of C. sativum were measured by total contents of polyphenol, flavonoid, DPPH and ABTS radical scavenging and reducing power activity. And anti-inflammatory effects of C. sativum were measured by LPS-induced RAW 264.7 cells. The results showed that the contents of total polyphenol and flavonoid were 76.03±1.36 mg of gallic acid equivalents/g and 182.23±4.32 mg of rutin equivalents/g at concentration 1 mg/ mL of C. sativum. The DPPH radical scavenging activity was found to be 52.8% at 500 µg/mL. The ABTS radical scavenging activity was shown in 58.3% after exposure to 1,000 µg/mL. Reducing power activity was found to be 66.8% at 2,000 µg/mL. The inhibitory effect of NO production was found to be 65% concentration 500 µg/mL. In the generation quantity of inflammatory cytokines such as TNF-α and IL-1β in cell culture medium, the expression levels of inflammatory proteins in cells were showed decrease with the increase of concentration. Therefore, we suggest that the C. sativum should be a potential source of alternative anti-inflammatory drug with good anti-inflammatory effects.
좀민들레(Taraxacum hallaisanense Nakai) 추출물의 항산화 및 항염증 활성 효과

본 논문은 가축의 면역 증진을 위한 천연 첨가제로서 좀민들레의 활용 가능성을 검토하고자 항산화 및 항염증 활성 평가를 실시하였다. 총 폴리페놀 및 플라보노이드 함량은 94.95, 86.33 mg/g으로 나타났고 DPPH, ABTS radical 소거능은 각각 100, 200 μg/mL의 농도에서 50%의 억제율을 보였으며, 1000 μg/mL에서 50%의 환원력을 나타냈다. LPS와 함께 처리한 Raw 264.7 cell에서는 좀민들레에 의한 세포 독성이 나타나지 않았으며 염증 매개 인자 NO와 염증성 사이도카인 IL-1β의 생성량을 유의하게 감소시켰다. 또한 염증성 단백질 발현량을 측정하기 위해 western blotting을 통해 확인한 결과, 400 μg/mL으로 처리하였을 때 LPS 처리구에 비해 염증성 단백질 발현 수준을 유의하게 감소시킨 것으로 확인되었다. 본 연구 결과, 좀민들레 추출물은 세포에 대한 독성이 없이 유의한 항산화 활성과 항염증 활성을 나타냄으로써 가축의 질병예방을 위한 면역 증진 및 생산성 향상에 기여할 수 있는 안전한 대체 천연 첨가제로 이용될 수 있다고 생각된다. Taraxacum hallaisanense (pr), a species of the family Compositae is a perennial herb plants that inhabit to Jeju Island. In the study, we performed to determine the anti-inflammatory effects in LPS-induced RAW 264.7 cells and antioxidant activity of ethanol extracts from T. hallaisanense whole plants. The antioxidant activity of extracts was measured by contents of polyphenol and flavonoid, DPPH radical scavenging, and reducing power activity. The anti-inflammatory effect of T. hallaisanense extracts was measured by NO and IL-1β production inhibitory activity and the expression of pro-inflammatory in lipopolysaccharide (LPS)- induced Raw 264.7 cells. Also, the expression of pro-inflammatory genes such as iNOS, COX-2 and NF-kB protein were reduced. In the cytotoxicity measurement by cytotoxicity kit, the extract was exhibited Raw 264.7 cell viabilities as nontoxic result in concentration of 25~400 μg/ml. These results indicated that ethanol extracts of T. hallaisanense whole plants expected development possibility as nutrial additives through high anti-inflammatory effects and antioxidant activity.

Veronica persica (V. persica) is a perennial plant that is broadly distributed in Europe, Asia and so on. V. persica is used for pain about the lower abdomen and low back. The aim of this study was to investigate the anti-oxidant and antiinflammatory effects of V. persica ethanol extract in LPS-induced RAW 264.7 cells. To evaluate the anti-oxidant activity, the DPPH and ABTS radical scavenging, total polyphenol and flavonoid contents, and reducing power activity were carried out. The DPPH and ABTS radical scavenging activity were evaluated as 72.0% and 73.0% at the concentrations of 200 and 500 μg/mL, respectively. Total polyphenol and flavonoid contents of V. persica extracts were measured as 65.22 mg/g and 43.82 mg/g at the concentration of 1 mg/mL. The reducing power activity measurement showed 53.0% activity at 1 mg/mL. The anti-inflammatory effects of the V. persica extract were evaluated in LPS induced RAW 264.7 cells. In the evaluation of cell viability by proliferation & cytotoxicity assay kit, the cytotoxicity of the extract was not confirmed at 0~800 μg/mL concentration. And the V. persica significantly inhibited NO production in a concentration dependent manner. The inhibition effects of NO in cell medium of V. persica was over 80% at 800 μg/mL. The V. persica also suppressed the expression of iNOS, COX-2, and phosphorylation of NF-κB and IkB-α proteins. These results indicate that the V. persica has anti-oxidant and anti-inflammatory effects by modulating NF-κB signaling pathways and can be used as natural functional materials.
